Measurements were performed in thin film silicon on insulator (SOI) nMOSFETs and it was observed a transition region in the subthreshold slope, larger than the theoretically expected, when the back interface (silicon film/buried oxide) changes from accumulation to depletion. Also, it was observed a non-constant plateau in the subthreshold slope when the back interface is accumulated.

MEDICI numerical bidimensional simulations were performed in order to analyze this transition region. It was verified that there is a back gate voltage range where a part of the back interface is not depleted over the whole subthreshold region, depending on the front gate voltage, which influences strongly the determination of the subthreshold slope, resulting in a non-abrupt transient region.

It is proposed a method for extracting the interface trap density in gate oxide/silicon film and silicon film/buried oxide interfaces minimizing the influence of the back accumulation layer in the subthreshold slope with the back interface accumulated. This method was also applied experimentally.  相似文献

In high-pressure-assisted freezing, samples are cooled under pressure (200 MPa) to - 20 °C without ice formation then pressure is released (0.1 MPa) and the high super-cooling reached (approx. 20 °C), promotes uniform and rapid ice nucleation. The size and location of ice crystals in large meat pieces (Longissimus dorsi pork muscle) as a result of high-pressure-assisted freezing were compared to those obtained by air-blast and liquid N(2). Samples from the surface and centre of the frozen muscle were histologically analysed using an indirect technique (isothermal-freeze fixation). Air-blast and cryogenic fluid freezing, having thermal gradients, showed non-uniform ice crystal distributions. High-pressure-assisted frozen samples, both at the surface and at the central zones, showed similar, small-sized ice crystals. This technique is particularly useful for freezing large pieces of food when uniform ice crystal sizes are required.  相似文献

Several variants of the enzyme pyridox(am)ine 5′-phosphate oxidase (PNPO), responsible for a rare form of vitamin B6-dependent neonatal epileptic encephalopathy known as PNPO deficiency (PNPOD), have been reported. However, only a few of them have been characterised with respect to their structural and functional properties, despite the fact that the knowledge of how variants affect the enzyme may clarify the disease mechanism and improve treatment. Here, we report the characterisation of the catalytic, allosteric and structural properties of recombinantly expressed D33V, R161C, P213S, and E50K variants, among which D33V (present in approximately 10% of affected patients) is one of the more common variants responsible for PNPOD. The D33V and E50K variants have only mildly altered catalytic properties. In particular, the E50K variant, given that it has been found on the same chromosome with other known pathogenic variants, may be considered non-pathogenic. The P213S variant has lower thermal stability and reduced capability to bind the FMN cofactor. The variant involving Arg161 (R161C) largely decreases the affinity for the pyridoxine 5′-phosphate substrate and completely abolishes the allosteric feedback inhibition exerted by the pyridoxal 5′-phosphate product.  相似文献

Osteosarcoma (OS) is a skeletal tumor affecting mainly children and adolescents. The presence of distance metastasis is frequent and it is localized preferentially to the lung, representing the main reason for death among patients. The therapeutic approaches are based on surgery and chemotherapeutics. However, the drug resistance and the side effects associated with the chemotherapy require the identification of new therapeutic approaches. The understanding of the complex biological scenario of the osteosarcoma will open the way for the identification of new targets for its treatment. Recently, a great interest of scientific community is for extracellular vesicles (EVs), that are released in the tumor microenvironment and are important regulators of tumor proliferation and the metastatic process. At the same time, circulating extracellular vesicles can be exploited as diagnostic and prognostic biomarkers, and they can be loaded with drugs as a new therapeutic approach for osteosarcoma patients. Thus, the characterization of OS-related EVs could represent a way to convert these vesicles from antagonists for human health into therapeutic and/or diagnostic agents.  相似文献

This paper presents the results obtained in computer simulations and laboratory measurements of interference caused by FM-TV signals to band limited quasi synchronous code division multiple access (BLQS-CDMA) signals. Certain parameters, such as frequency spacing between wanted and interfering carriers and type of modulating TV signal (75 per cent saturated colour bar or saturated red field), were varied during the measurements and simulations, in order to study their influence on the amount of interference suffered by the BLQS-CDMA signal. Computer simulations were validated by hardware measurements, in order to have available an in-house software tool for future analyses. Protection ratio (PR) masks have been obtained and show that BLQS-CDMA is well adapted to a VSAT network operating in an FM-TV interference environment. © 1997 John Wiley & Sons, Ltd.  相似文献

The exact mechanism underlying selective dopaminergic neurodegeneration is not completely understood. The complex interplay among toxic alpha-synuclein aggregates, oxidative stress, altered intracellular Ca2+-homeostasis, mitochondrial dysfunction and disruption of mitochondrial integrity is considered among the pathogenic mechanisms leading to dopaminergic neuronal loss. We herein investigated the molecular mechanisms leading to mitochondrial dysfunction and its relationship with activation of the neuroinflammatory process occurring in Parkinson’s disease. To address these issues, experiments were performed in vitro and in vivo in mice carrying the human mutation of α-synuclein A53T under the prion murine promoter. In these models, the expression and activity of NCX isoforms, a family of important transporters regulating ionic homeostasis in mammalian cells working in a bidirectional way, were evaluated in neurons and glial cells. Mitochondrial function was monitored with confocal microscopy and fluorescent dyes to measure mitochondrial calcium content and mitochondrial membrane potential. Parallel experiments were performed in 4 and 16-month-old A53T-α-synuclein Tg mice to correlate the functional data obtained in vitro with mitochondrial dysfunction and neuroinflammation through biochemical analysis. The results obtained demonstrated: 1. in A53T mice mitochondrial dysfunction occurs early in midbrain and later in striatum; 2. mitochondrial dysfunction occurring in the midbrain is mediated by the impairment of NCX3 protein expression in neurons and astrocytes; 3. mitochondrial dysfunction occurring early in midbrain triggers neuroinflammation later into the striatum, thus contributing to PD progression during mice aging.  相似文献

Understanding the mechanisms of colorectal cancer progression is crucial in the setting of strategies for its prevention. δ-Valerobetaine (δVB) is an emerging dietary metabolite showing cytotoxic activity in colon cancer cells via autophagy and apoptosis. Here, we aimed to deepen current knowledge on the mechanism of δVB-induced colon cancer cell death by investigating the apoptotic cascade in colorectal adenocarcinoma SW480 and SW620 cells and evaluating the molecular players of mitochondrial dysfunction. Results indicated that δVB reduced cell viability in a time-dependent manner, reaching IC50 after 72 h of incubation with δVB 1.5 mM, and caused a G2/M cell cycle arrest with upregulation of cyclin A and cyclin B protein levels. The increased apoptotic cell rate occurred via caspase-3 activation with a concomitant loss in mitochondrial membrane potential and SIRT3 downregulation. Functional studies indicated that δVB activated mitochondrial apoptosis through PINK1/Parkin pathways, as upregulation of PINK1, Parkin, and LC3B protein levels was observed (p < 0.0001). Together, these findings support a critical role of PINK1/Parkin-mediated mitophagy in mitochondrial dysfunction and apoptosis induced by δVB in SW480 and SW620 colon cancer cells.  相似文献

A construct based on the electrostatic layer-by-layer self assembly technique has been fabricated, to be used as a tailored device to encourage nerve regeneration. A multilayered nanocoating composed of three precursor bilayers of cationic poly(dimethyldiallylammonium) chloride (PDDA) and anionic poly(styrenesulfonate) (PSS), followed by bilayers of poly-D-lysine (PDL) and antibody specific to transforming growth factor 1 (anti-TGF-1), has been deposited on HYAFF 11. The assembly process has been monitored by quartz crystal microbalance (QCM) for its characterisation and then it has been used on HYAFF 11. Structural studies of the resulting multilayers confirmed stepwise deposition of anti-TGF-1, with an average layer thickness of 2.2+/-0.2 nm and an average surface density of 0.36+/-0.03 mug cm(-2). Scanning electron microscopy has been used to characterise multilayer uniformity. Finally, the immunological activity of the multilayered structure has been assessed. The results show that anti-TGF-1 can be included in its active form in a predetermined multilayered structure onto HYAFF 11 with quantitative control of layer thickness and weight, providing a high tool with great potential in tissue engineering.  相似文献

Research into novel internal combustion engines requires consideration of the diversity in future fuels in an attempt to reduce drastically CO2 emissions from vehicles and promote energy sustainability. Hydrogen has been proposed as a possible fuel for future internal combustion engines and can be produced from renewable sources. Hydrogen’s wide flammability range allows higher engine efficiency than conventional fuels with both reduced toxic emissions and no CO2 gases. Most previous work on hydrogen engines has focused on spark-ignition operation. The current paper presents results from an optical study of controlled autoignition (or homogeneous charge compression ignition) of hydrogen in an engine of latest spark-ignition pentroof combustion chamber geometry with direct injection of hydrogen (100 bar). This was achieved by a combination of inlet air preheating in the range 200–400 °C and residual gas recirculated internally by negative valve overlap. Hydrogen fuelling was set to various values of equivalence ratio, typically in the range ? = 0.40–0.63. Crank-angle resolved flame chemiluminescence images were acquired for a series of consecutive cycles at 1000 RPM in order to calculate in-cylinder rates of flame expansion and motion. Planar Laser Induced Fluorescence (LIF) of OH was also applied to record more detailed features of the autoignition pattern. Single and double (i.e. ‘split’ per cycle) hydrogen injection strategies were employed in order to identify the effect of mixture preparation on autoignition’s timing and spatial development. An attempt was also made to review relevant in-cylinder phenomena from the limited literature on hydrogen-fuelled spark-ignition optical engines and make comparisons were appropriate.  相似文献
