Untreated paired helical filaments (PHFs) and pronase-digested PHF-core filaments were stereoscopically imaged with a freeze-drying vertical platinum-carbon replication preparation method for TEM. The untreated PHF have an average wide region (W) = 22.8 +/- 2.4 nm, a narrow region (T) = 10.6 +/- 1.7 nm, and a helical turn period (L) = 78.6 +/- 13.4. The surfaces of the untreated PHF's fuzzy coat appears disorganized. The widths of the pronase-treated PHF-core filaments were significantly reduced (W(d) = 14.8 +/- 1.2 nm, T(d) = 5.7 +/- 1.0 nm, and L(d) = 75.4 +/- 17 nm). The surfaces of the untreated PHF contained approximately 1.1 nm strands, the same size as tau monomer ( approximately 1.0 nm). The pronase-digested PHF cores mostly contained approximately 1.6 +/- 0.3 nm strands although strand diameters ranged from 0.6-2.5 nm. The strands sometimes appear to be wrapped around the filament axis; less often, they appear to be roughly parallel to the PHF axis, and otherwise appear to be randomly oriented. Images of pronase-digested PHF core images are discussed in relation to the core's biochemical composition, its proposed beta structure, and structural subunit models. Images of the untreated and the pronase-digested PHF support a helical ribbon morphology.  相似文献

Due to their interesting size-dependent magnetic characteristics and relative biocompatibility, magnetic superparamagnetic iron oxide (SPIO) nanoparticles have been widely exploited as probes for cell and subcellular structure identification, as well as medication and gene delivery. A thorough understanding of the mechanics of the interaction between nanoparticles and macrophages is vital in managing dynamic processes in nanomedicine. In this study, the interaction behavior and uptake of SPIO nanoparticles by M1- and M2-type macrophages were investigated. Mice monocytes were differentiated into M1 and M2 macrophages, and the uptake of SPIO nanoparticles was studied using a TEM microscope. A high resolution image of 1 nm resolution, an image processing technique, was developed to extract the SPIO-NPs from tomographic TEM microscopic images. Lysosomes appear to be the zones of high concentrations of SPIO inside macrophages. Lysosomes were first selected in each image, and then segmentation by the Otsu thresholding method was used to extract the SPIO-NPs. The Otsu threshold method is a global thresholding technique used to automatically differentiate SPIOs from the background. The SPIO-NPs appear in red colors, and the other pixels in the image are considered background. Then, an estimation of the SPIO-NP uptakes by lysosomes is produced. Higher uptake of all-sized nanoparticles was observed in M1- and M2-type macrophages. An accurate estimation of the number of SPIO-NPs was obtained. This result will help in controlling targeted drug delivery and assessing the safety impact of the use of SPIO-NPs in nanomedicine for humans.  相似文献

This paper considers the problem of optimizing the operation of Indirect Adiabatic Cooling (IAC) systems with application to data centers. Optimal operation is achieved when the required cooling demand is satisfied at the minimum energy cost. For this purpose, we design a supervisory control system, where the higher layer determines the optimal set-points for the local controllers by employing an Extremum Seeking Control (ESC) scheme. In particular, we consider a Newton-like phasor ESC, which augments the derivative estimator underlying the phasor approach to capture also the Hessian of the plant index and then it uses these estimates to steer the system along a Newton-like direction. The effectiveness of the considered approach is tested in simulation by exploiting a Matlab-based simulation environment including an IAC system and a computer room.  相似文献

Nanoengineered periodic array of holes on superconducting thin films have a great interest due to their excellence for the studies of the vortex pinning mechanisms in the type-II superconductors. Rectangular array of holes has been fabricated over a microbridge of Nb superconducting thin film by e-beam lithography. Rectangular array of holes have two type of scenario, at low magnetic fields matching effects are sharp and narrow while at high fields wide and shallower. In this work, we study the matching pinning effect by the artificial hole array in superconducting Nb thin films. We observed that as the inter distance between holes is decreased; the sharp matching effects become dominant and vice versa.  相似文献

In this article, a ternary WO3/g‐C3N4@ BiVO4 composites were prepared using eco‐friendly hydrothermal method to produce efficient hydrogen energy through water in the presence of sacrificial agents. The prepared samples were characterized by scanning electron microscopy (SEM), scanning transmission electron microscopy (STEM), transmission electron microscopy (TEM), X‐ray diffraction (XRD), ultraviolet‐visible (UV‐vis), Brunauer‐Emmett‐Teller (BET) surface area, and photoluminescence spectroscopy (PL) emission spectroscopy. The experimental study envisages the formation of 2‐D nanostructures and observed that such kinds of nanostructures could provide more active sites for photocatalytic reduction of water and their inherent reactive‐species mechanism. The results showed the excellent photocatalytic performance (432 μmol h?1 g?1) for 1.5% BiVO4 nanoparticles in WO3/g‐C3N4 composite when compared with pure WO3 and BiVO4. The optical properties and photocatalytic activity measurement confirmed that BiVO4 nanoparticles in WO3/g‐C3N4 photocatalyst inhibited the recombination of photogenerated electron and holes and enhanced the reduction reactions for H2 production. The enhanced photocatalytic efficiency of the composite nanostructures may be attributed to wide absorption region of visible light, large surface area, and efficient separation of electrons/holes pairs owing to synergistic effects between BiVO4 and WO3/g‐C3N4. The prepared samples would be a precise optimal photocatalyst to increase their suppliers for worldwide applications especially in energy harvesting.  相似文献

Mobile app reviews by users contain a wealth of information on the issues that users are experiencing. For example, a review might contain a feature request, a bug report, and/or a privacy complaint. Developers, users and app store owners (e.g. Apple, Blackberry, Google, Microsoft) can benefit from a better understanding of these issues – developers can better understand users’ concerns, app store owners can spot anomalous apps, and users can compare similar apps to decide which ones to download or purchase. However, user reviews are not labelled, e.g. we do not know which types of issues are raised in a review. Hence, one must sift through potentially thousands of reviews with slang and abbreviations to understand the various types of issues. Moreover, the unstructured and informal nature of reviews complicates the automated labelling of such reviews. In this paper, we study the multi-labelled nature of reviews from 20 mobile apps in the Google Play Store and Apple App Store. We find that up to 30 % of the reviews raise various types of issues in a single review (e.g. a review might contain a feature request and a bug report). We then propose an approach that can automatically assign multiple labels to reviews based on the raised issues with a precision of 66 % and recall of 65 %. Finally, we apply our approach to address three proof-of-concept analytics use case scenarios: (i) we compare competing apps to assist developers and users, (ii) we provide an overview of 601,221 reviews from 12,000 apps in the Google Play Store to assist app store owners and developers and (iii) we detect anomalous apps in the Google Play Store to assist app store owners and users.  相似文献

Tuning of porosity and surface properties of nanoparticles especially on carbon-based nanomaterials, adopting a ‘greener’ or self-activation synthesis technique for electrical charge storage, is progressing. Herein, we report the self-activation of Teak wood sawdust in a nitrogen atmosphere at different activation temperatures to synthesize carbon nanoparticles. The activated carbon nanoparticles synthesized at 900 °C exhibits a maximum?~?360 m2 g?1 surface area with?~?2 nm average pore size diameter. Five electrolytes viz. KOH, KCl, Na2SO4, NaCl, and H3PO4 are used for studying the supercapacitance nature of the activated carbon nanoparticles in a 3-electrode configuration. A maximum specific capacitance of?~?208 F g?1 @ 0.25 A g?1 is obtained in 1 M KOH as the electrolyte. Two symmetric supercapacitors, aqueous (1 M KOH) and solid-state (PVA/KOH), are fabricated, and their performance difference is compiled. The solid-state symmetric supercapacitor performs in a wider voltage window (1.7 V) with a superior energy density of 27.1 Wh kg?1 at a power density of 178 W kg?1.
