Xyloglucan is a major hemicellulosic component in plant cell walls. Phytopathogenic fungi secrete cell wall-degrading enzymes on their infection to hosts, while the nature of the cell wall-lytic enzymes of such fungi are yet to be fully understood. Verticillium dahliae is a soil-borne fungus that causes vascular wilt diseases in a variety of commercially important crops worldwide. We purified two types of xyloglucanases, XEG12A and XEG74B, from the culture of naturally isolated Verticillium dahliae strain 2148. XEG12A showed a molecular size of 23 kDa with its maximal activity at pH 7.5. XEG12A specifically hydrolyzed xyloglucan with no activity on other β-glucans. XEG74B had a molecular size of 110 kDa with its optimum pH at 6.0. XEG74B primarily hydrolyzed xyloglucan, with a slight activity on β-1,3-1,4-glucan. Analysis of hydrolytic products of xyloglucanooligasaccharide (XXXGXXXG) by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) revealed that the both enzymes cleaved β-1,4-glucosidic linkage at the position of unbranched chain, while XEG74B showed a little fluctuation with the cleavage site. Both enzymes did not hydrolyzed xyloglucanoheptasaccharide (XXXG) at all. N-Terminal and internal amino acid sequencing of the enzymes revealed that XEG12A and XEG74B belonged to Glycoside Hydrolase (GH) Families 12 and 74, respectively. Based on these results we concluded that V. dahliae XEG12A and XEG74B were xyloglucan-specific endo-β-1,4-glucanases (EC 3.2.1.151).  相似文献

A DNA–cyclodextrin–silica composite was prepared by the sol–gel method. This composite possessed the bi-functions of double-stranded DNA, such as intercalation into DNA, and cyclodextrin, such as inclusion into its intramolecular cavity. Therefore, we demonstrated the accumulation of harmful compounds from an aqueous multi-component solution using a DNA–cyclodextrin–silica composite column. As a result, the DNA–cyclodextrin–silica composite column can effectively accumulate not only planar structure-containing harmful compounds, such as dioxin and polychlorobiphenyl (PCB) derivatives, but also non-planar structure containing compounds, such as bisphenol A and diethylstilbestrol, from an aqueous multi-component solution. The accumulated amount of these harmful compounds was more than 90%. Additionally, the DNA–cyclodextrin–silica composite column was recycled by the application of methanol. Therefore, the DNA–cyclodextrin–silica composite may have the potential to be used as an environmental material for the accumulation of harmful compounds from industrial or experimental waste.  相似文献

Pectin is one of the biopolymers in the cell walls of all plant tissues, but the pectin‐containing materials have been discarded as industrial waste in food‐processing factories. We prepared a water‐insoluble pectin‐inorganic composite material by mixing pectin and a silane coupling reagent, bis(3‐trimethoxysilylpropyl)amine. The mechanical strength of the pectin‐inorganic composite material was higher than that of the pectin material without the addition of an inorganic component. In addition, the thermal stability of the composite material increased with the addition of the inorganic component. Furthermore, when the pectin‐inorganic composite materials were incubated in an aqueous solution of Cu(II), Zn(II), or In(III), these composite materials effectively accumulated not only the heavy metal ions, but also rare‐earth metal ions. Additionally, based on the infrared (IR) measurements, the metal ion accumulative mechanism into the composite material is described. As a result, the IR spectra suggested an electrostatic interaction between the metal ion and carboxy group in the pectin. © 2015 Wiley Periodicals, Inc. J. Appl. Polym. Capuramycins are one of several known classes of natural products that contain an l ‐Lys‐derived l ‐α‐amino‐?‐caprolactam (l ‐ACL) unit. The α‐amino group of l ‐ACL in a capuramycin is linked to an unsaturated hexuronic acid component through an amide bond that was previously shown to originate by an ATP‐independent enzymatic route. With the aid of a combined in vivo and in vitro approach, a predicted tridomain nonribosomal peptide synthetase CapU is functionally characterized here as the ATP‐dependent amide‐bond‐forming catalyst responsible for the biosynthesis of the remaining amide bond present in l ‐ACL. The results are consistent with the adenylation domain of CapU as the essential catalytic component for l ‐Lys activation and thioesterification of the adjacent thiolation domain. However, in contrast to expectations, lactamization does not require any additional domains or proteins and is likely a nonenzymatic event. The results set the stage for examining whether a similar NRPS‐mediated mechanism is employed in the biosynthesis of other l ‐ACL‐containing natural products and, just as intriguingly, how spontaneous lactamization is avoided in the numerous NRPS‐derived peptides that contain an unmodified l ‐Lys residue.  相似文献

The chain stiffness and local chain conformation of atactic poly(α-methylene-γ-butyrolactone) (PMBL), which is a side chain cyclic structural analog of poly(methyl methacrylate) (PMMA), with a weight-average molecular weight (Mw) ranging from 2.8 × 103 to 2.6 × 106 in N,N-dimethylformamide (DMF) and γ-butyrolactone (GBL) were characterized by size exclusion chromatography with a multi-angle light-scattering detector (SEC-MALS) and synchrotron radiation small-angle X-ray scattering (SAXS). Based on the Kratky-Porod worm-like chain model, the scattering functions and the Mw dependence of z-average root-mean-square radius of gyration <S2>z1/2 yielded the Kuhn segment lengths λ−1, the diameter of the PMBL chains d, and the excluded-volume strengths in DMF and GBL. The local conformation of atactic PMBL in DMF and GBL were slightly larger than those of atactic PMMA, due to the presence of the conformationally rigid lactone ring structure.  相似文献

Developmental ability of cloned embryos from neural stem cells   总被引:2,自引:0,他引:2  
The success rate is generally higher when cloning mice from embryonic stem (ES) cell nuclei than from somatic cell nuclei, suggesting that the embryonic nature or the undifferentiated state of the donor cell increases cloning efficiency. We assessed the developmental ability of cloned embryos derived from cultured neural stem cell (NSC) nuclei and compared the success rate with that of embryos cloned from other donor cells such as differentiated NSCs, cumulus cells, Sertoli cells and ES cells in the mouse. The transfer of two-cell cloned embryos derived from cultured NSC nuclei into surrogate mothers produced five live cloned mice. However, the success rate (0.5%) was higher in embryos cloned from cultured NSC nuclei than from differentiated NSCs (0%), but lower than that obtained by cloning mice from other cell nuclei (2.2-3.5%). Although the in vitro developmental potential to the two-cell stage of the cloned embryos derived from NSC nuclei (73%) was similar to that of the cloned embryos derived from other somatic cell nuclei (e.g., 85% in Sertoli cells and 75% in cumulus cells), the developmental rate to the morula-blastocyst stage was only 7%. This rate is remarkably lower than that produced from other somatic cells (e.g., 50% in Sertoli cells and 54% in cumulus cells). These results indicate that the undifferentiated state of neural cells does not enhance the cloning efficiency in mice and that the arrest point for in vitro development of cloned embryos depends on the donor cell type.  相似文献

We have developed an analytical method for terpene resins in chewing gum. The fraction including terpene resins was prepared by means of hexane extraction and two silica gel column chromatography treatments (hexane and ethyl acetate) from chewing gum. The terpene resin fraction was analyzed with LC/MS and IR. The terpene resins are mixtures of polymeric pinene and/or limonene, which have a monomer molecular weight of 136. The MS spectrum of the terpene resin peak on the LC/MS total ion chromatogram showed protonated molecular ion (M + H)+ peaks at intervals of m/z 136, characteristic of a complex mixture of polyterpenes. IR spectroscopy is a suitable technique to identify the terpene resin type, ie., pinene or limonene. When the method was applied to imported chewing gum sold in Japan, terpene resins were clearly detected.  相似文献

We investigated the effect of UV-A light (wavelength 315 to 400 nm) irradiation during storage on tomato fruit injury. Mature green tomato fruit (cv. House Momotaro) were exposed to UV-A at doses of 0.02, 0.5, and 2 mW x cm(-2) throughout storage at 25 degrees C. The physiological disorders, fruit ripening, superoxide dismutase (SOD) activity, and increases in fruit temperature were evaluated. All UV-A-irradiated and nonirradiated tomatoes developed a full red color at the same time (2 weeks). Irradiated fruit ripened normally, and exposure of tomato fruits to UV-A did not lead to the discoloration of ripe tomato fruit at any dosage. The fruit temperature did not increase in response to various UV-A light doses and exposure times, and none of the UV-irradiated fruits showed physiological disorders (dull skin blemish, pitting). The SOD activity of UV-A-irradiated fruit exposed to the various UV-A doses did not significantly (P = 0.05) differ from that of fruit stored in dark conditions. The SOD results imply that UV-A light might not induce reactive oxygen species in UV-A-irradiated fruit.  相似文献

Yttria-ceria-doped tetragonal zirconia (Y,Ce)-TZP)/alumina (Al2O3) composites were fabricated by hot isostatic pressing at 1400° to 1450°C and 196 MPa in an Ar–O2 atmosphere using the fine powders prepared by hydrolysis of ZrOCl2 solution. The composites consisting of 25 wt% Al2O3 and tetragonal zirconia with compositions 4 mol% YO1.5–4 mol% CeO2–ZrO2 and 2.5 mol% YO1.5–5.5 mol% CeO2–ZrO2 exhibited mean fracture strength as high as 2000 MPa and were resistant to phase transformation under saturated water vapor pressure at 180°C (1 MPa). Postsintering hot isostatic pressing of (4Y, 4Ce)-TZP/Al2O3 and (2.5Y, 5.5Ce)-TZP/Al2O3 composites was useful to enhance the phase stability under hydrothermal conditions and strength.  相似文献
