1. We compared the binding profiles and contractile mechanisms of putative muscarinic M1 agonists McN-A-343 and AHR-602 with those of carbachol in smooth muscle of guinea-pig taenia caeci. 2. McN-A-343 and AHR-602, as well as carbachol, completely displaced the atropine-sensitive binding of [3H]-quinuclidinyl benzilate to muscarinic receptors present in the membrane preparation. The potency order for the affinity of these agents for muscarinic receptors was carbachol > McN-A-343 > AHR-602. 3. In the presence of 2.2 mM extracellular Ca2+, McN-A-343 and AHR-602 induced contraction corresponding to 79 and 85%, respectively, of the maximal contraction to 0.1 mM carbachol. Contractions induced by these agents were mediated via activation of the muscarinic receptor subtype that had a high affinity for 4-DAMP (M3 selective) but a low affinity for pirenzepine (M1 selective) and AF-DX 116 (M2 selective). These contractions were inhibited by an L-type Ca2+ channel blocker, verapamil. 4. In Ca(2+)-free solution containing 2 mM EGTA, carbachol elicited a transient contraction whereas no contraction was observed in response to McN-A-343 and AHR-602. Application of McN-A-343 or AHR-602 inhibited the carbachol-induced contraction in Ca(2+)-free solution, and this inhibition was surmounted by a higher concentration of carbachol. 5. The EC50 value for carbachol-induced contraction in the presence of extracellular Ca2+ was approximately 175 times lower than that in the absence of Ca2+. After treatment with propylbenzilylcholine mustard, carbachol induced contraction only in the presence of extracellular Ca2+. 6. The results suggest that in the taenia caeci there is a greater receptor reserve for muscarinic M3 receptor-mediated Ca2+ influx than for M3 mediated Ca2+ release. The compounds McN-A-343 and AHR-602 are agonists of the Ca2+ influx pathway, but do not appear to stimulate the Ca2+ release pathway.  相似文献

Planar type Gunn effect devices have been fabricated by sulfur-ion implantation into the Cr doped semi-insulating GaAs substrates. The high doping efficiency as 90% was obtained as a result of long heat treatment. The mobility of the sulfur-ion implanted n layers with average carrier concentration of 4 × 1016cm?3 was 5200 cm2/Vsec at room temperature and 12,000 cm2/Vsec at 77 K. The minimum gate trigger voltage of the Gunn effect digital devices was 100 mV. Sulfur-ion implanted Gunn effect devices have shown superior current drop ratio dependence on doping-depth product, compared to the devices prepared from the epitaxial layer.  相似文献

Diffusionless tetragonal ( t' )  cubic ( c' ) phase transformation in 65-mol%-CeO2-ZrO2 was investigated around the c'-t' equilibrium temperature T0 c'-t' using powder X-ray diffraction, where c' was defined as a tetragonal or cubic phase with an axial ratio of unity. The {400} peak profile of t' -ZrO2 broadened at 860° and 900°C. This indicated that the t' -ZrO2 transformed partially into c' -ZrO2. The {400} peak profile of c' -ZrO2 also broadened at 860° and 900°C, which indicated that the c' -ZrO2 partially transformed to t' -ZrO2. The finish point of t'→c' transformation Tft'c' was investigated by annealing the t' -ZrO2 containing 30–65 mol% CeO2 at various temperatures. The T'ft'→c' line existed in the vicinity of the cubic solubility limit within the t + c two-phase region. This Tft'→c' location, which should have been located in the vicinity of T0c'-t' could not be explained by a simple regular solution model. However, it was described successfully by a thermodynamic model based on Landau's phenomenologic theory.  相似文献

A polymerized complex technique was used to prepare high-purity pyrochlore Y2Ti2O7 powders at 750°C. Heating of a mixed solution of citric acid (CA), ethylene glycol (EG), and yttrium and titanium ions, with a molar ratio of CA:EG:Y:Ti = 5:20:1:1, at 130°C produced a yellowish transparent polymeric gel without any precipitation; this material was used subsequently as a precursor for Y2Ti2O7. Based on the results of 13C-NMR spectroscopy, it was suggested that a mixed-metal (Y,Ti)-CA3 chelate complex formed in a starting CA/EG solution. The formation of pure pyrochlore Y2Ti2O7 occurred when the precursor was heat-treated in a furnace set at 750°C in static air for 4 h.  相似文献

The aim of this study was to investigate the use of [18F]fluorodeoxyglucose and positron emission tomography (FDG PET) for quantitative evaluation of glucose metabolism in skeletal muscle during walking. Ten young males underwent FDG PET twice during walks, which were done with or without an automated stride assistance system (SAS). Walk ratios were significantly increased by the SAS in seven subjects. Regional glucose metabolism in muscles between the crista iliaca and the planta was clearly visualized in all ten subjects. Glucose utilization increased significantly in the tibialis posterior and medial gastrocnemius muscles of the seven subjects in whom walk ratios were increased by the SAS. FDG PET is useful for analysis of muscle activity during exercise and rehabilitation.  相似文献

Zirconia-yttria (ZrO2-12 mol% YO1.5) powders were prepared via an organic polymerized complex method that was based on the Pechini-type reaction route. A mixed solution of citric acid, ethylene glycol, and zirconium and yttrium ions were polymerized to form a transparent resin, which then became a black powder after charring at ∼450°C. This product was used as a precursor for ZrO2-12 mol% YO1.5. The formation of a metastable tetragonal ( t ´) phase with compositional homogeneity occurred when the precursor was heated at the rate of 10°C/min up to a temperature >560°C. X-ray diffraction peaks became sharp with an increase in the heat-treatment temperature. A t ´-form with cell parameters of a = 5.1265(6) Å and c = 5.1559(9) Å were obtained via heat treatment up to 1400°C. This study shows that the organic polymerized complex method is effective for obtaining the compositionally homogeneous metastable t ´-form in the ZrO2-YO1.5system for short annealing times at relatively lower temperatures.  相似文献

We developed heterojunction-based Schottky solar cells consisting of π-conjugated polymers and n-type GaN. Poly (3,4-ethylendioxythiophene):poly (styrenesulfonate) (PEDOT:PSS) was used as the transparent Schottky contact material and their electrical properties were investigated in comparison with those of a polyaniline (PANI) Schottky contact. The PEDOT:PSS/n-GaN/sapphire (0 0 0 1) sample exhibited high-quality rectifying characteristics with a low reverse leakage current of less than 10−8 A/cm2 at a reverse bias voltage of −3 V. While investigating the photovoltaic performance, it was observed that the open-circuit voltage of the PEDOT:PSS/n-GaN/sapphire (0 0 0 1) sample reached 0.8 V, which was much superior to the photovoltage reported for a conventional metal/GaN Schottky photodetector. We also confirmed that the PEDOT:PSS is as promising a material as PANI for π-conjugated polymer/GaN Schottky solar cells.  相似文献

5-aminolevulinic acid (5-ALA)-induced protoporphyrin IX (PpIX) fluorescence is widely used for the intraoperative detection of malignant tumors. However, the fluorescence emission profiles of the accompanying necrotic regions of these tumors have yet to be determined. To address this, we performed fluorescence and high-performance liquid chromatography (HPLC) analyses of necrotic tissues of squamous cancer after 5-ALA administration. In resected human lymph nodes of metastatic squamous cell carcinoma, we found a fluorescence peak at approximately 620 nm in necrotic lesions, which was distinct from the PpIX fluorescence peak at 635 nm for viable cancer lesions. Necrotic lesions obtained from a subcutaneous xenograft model of human B88 oral squamous cancer also emitted the characteristic fluorescence peak at 620 nm after light irradiation: the fluorescence intensity ratio (620 nm/635 nm) increased with the energy of the irradiation light. HPLC analysis revealed a high content ratio of uroporphyrin I (UPI)/total porphyrins in the necrotic cores of murine tumors, indicating that UPI is responsible for the 620 nm peak. UPI accumulation in necrotic tissues after 5-ALA administration was possibly due to the failure of the heme biosynthetic pathway. Taken together, fluorescence imaging of UPI after 5-ALA administration may be applicable for the evaluation of tumor necrosis.  相似文献
