Deployable scissor structures are well equipped for temporary and mobile applications since they are able to change their form and functionality. They are structural mechanisms that transform from a compact state to an expanded, fully deployed configuration. A barrier to the current design and reuse of scissor structures, however, is that they are traditionally designed for a single purpose. Alternatively, a universal scissor component (USC)—a generalized element which can achieve all traditional scissor types—introduces an opportunity for reuse in which the same component can be utilized for different configurations and spans. In this article, the USC is optimized for structural performance. First, an optimized length for the USC is determined based on a trade-off between component weight and structural performance (measured by deflections). Then, topology optimization, using the simulated annealing algorithm, is implemented to determine a minimum weight layout of beams within a single USC component.  相似文献

Recent studies showed the superior separation performance of stirred-pulsed columns of different diameters in liquid-liquid extraction processes. Here, an efficient shortcut method will be presented, which is time and resource-efficient as well as cost-effective to determine the operational window of these columns for industrial separation tasks. Savings in time of less experiments and costs of materials consumption can be estimated with up to 30 %. The presented method is particularly suitable before the application of new chemical systems, which are particularly cost-intensive and scarce in material supply.  相似文献

Application Programming Interfaces (API) are exposed to developers in order to reuse software libraries. API directives are natural-language statements in API documentation that make developers aware of constraints and guidelines related to the usage of an API. This paper presents the design and the results of an empirical study on the directives of API documentation of object-oriented libraries. Process development and small‐scale production gets more and more important in fine chemistry and pharmaceutical production. An equipment toolbox assists process synthesis presented in this contribution. A microfluidic calorimeter can measure kinetic and thermodynamic reaction data with commercial plate reactors. A tubular reactor coiled with 90° bends allows for long residence time with low axial dispersion, also known as coiled flow inverter (CFI). A similar setup is used for continuous‐flow cooling crystallization. Small‐scale columns with rotating internals are employed for distillation and liquid‐liquid extraction. Main emphasis will be put on automation and scale‐up in future steps.  相似文献

Advantages of the baculovirus insect cell expression system for production of recombinant proteins include high capacity, flexibility, and glycosylation capability. In this study, this expression system was exploited to produce anti-cancer monoclonal antibody (mAb) CO17-1A, which recognizes the antigen GA733. The heavy chain (HC) and light chain (LC) genes of mAb CO17-1A were cloned under the control of P10 and Polyhedrin promoters in the pFastBac™ dual vector, respectively. Gene expression cassettes carrying the HC and LC genes were transposed into a bacmid in Escherichia coli (DH10Bac). The transposed bacmid was transfected to Sf9 insect cells to generate baculovirus expressing mAb CO17-1A. Confocal immunofluorescence and Western blot analyses confirmed expression of mAb CO17-1A in baculovirus-infected insect cells. The optimum conditions for mAb expression were evaluated at 24, 48, and 72 h after the virus infection at an optimum virus multiplicity of infection of 1. Expression of mAb CO17-1A in insect cells significantly increased at 72 h after infection. HPLC analysis of glycosylation status revealed that the insect-derived mAb (mAbI) CO17-1A had insect specific glycan structures. ELISA showed that the purified mAbI from cell culture supernatant specifically bound to SW948 human colorectal cancer cells. Fluorescence-activated cell sorting analysis showed that, although mAbI had insect specific glycan structures that differed from their mammalian counterparts, mAbI similarly interacted with CD64 (FcγRI) and Fc of IgG, compared to the interactions of mammalian-derived mAb. These results suggest that the baculovirus insect cell expression system is able to express, assemble, and secrete biofunctional full size mAb.  相似文献

We report on the fabrication and characterization of nanoscale memory elements based on solid electrolytes. When combined with silver, chalcogenide glasses such as Se-rich Ge-Se are good solid electrolytes, exhibiting high Ag ion mobility and availability. By placing an anode that has oxidizable Ag and an inert cathode (e.g., Ni) in contact with a thin layer of such a material, a device is formed that has an intrinsically high resistance, but which can be switched to a low-resistance state at small voltage via reduction of the silver ions. An opposite bias will return the device to a high-resistance state, and this reversible switching effect is the basis of programmable metallization cell technology. In this paper, electron beam lithography was used to make sub-100-nm openings in polymethylmethacrylate layers used as the dielectric between the device electrodes. The solid electrolyte film was formed in these via-holes so that their small diameter defined the active switching area between the electrodes. The Ag-Ge-Se electrolyte was created by the photodiffusion, with or without thermal assistance, of an Ag layer into the Ge-Se base glass. Combined thermal and photodiffusion leads to a nanophase separated material with a dispersed Ag ion-rich material with an average crystallite size of 7.5 nm in a glassy insulating Ge-rich continuous phase. The nanoscale devices write at an applied bias as low as 0.2 V, erase by -0.5 V, and fall from over 10/sup 7/ /spl Omega/ to a low-resistance state (e.g., 10/sup 4/ /spl Omega/ for a 10-/spl mu/A programming current) in less than 100 ns. Cycling appears excellent with projected endurance well beyond 10/sup 11/ cycles.  相似文献

This study aimed to investigate the progress of wound healing around silicone expander with particular emphasis on fibroblasts, myofibroblasts and collagen in the repair phase. Semi-quantitative evaluation of inflammatory cells and their cytokines, fibroblasts and myofibroblasts at the tissue–material interface was carried out. Commercially available silicone expander was implanted in gluteus muscle of young female Wistar rats for 3, 7, 14, 30, 90 and 180 days. Ultra high molecular weight polyethylene served as control. The cellular response was studied by immunohistochemistry and Transmission Electron Microscopy. A thick collagenous fibrous capsule was observed around the silicone expander at 180 days, with persistent myofibroblasts, lymphocytes and macrophages as compared to the thin fibrous encapsulation around the UHMWPE implants. The regulatory role of cytokines and immune cells in myofibroblast persistence in tissue–implant interface around silicone expander has been extensively studied. Results of this study indicate the need to elucidate the signaling molecules in the transition of fibroblast to myofibroblast around silicone expander implants.  相似文献

ABSTRACT: Three antioxidant flavone C -glucosides, isoorientin, vitexin, and isovitexin, were identified for the 1st time from Atractylodes japonica leaves by an activity-guided fractionation with various analytical techniques including column chromatography, high-performance liquid chromatography (HPLC), fast atom bombardment mass spectrometry (FAB-MS), and nuclear magnetic resonance (NMR). Hydroxyl and superoxide anion radicals scavenging activities of these compounds were examined using electron spin resonance (ESR). Inhibitory activities of these compounds on human low-density lipoprotein (LDL) oxidation were evaluated by malondialdehyde (MDA) and thiobarbituric acid-reactive substances (TBARS), both representing intermediates of lipid peroxidation. These flavone C -glucosides displayed about 50% of scavenging activity against hydroxyl radicals at the concentrations below 10 μ M . The superoxide dismutase (SOD)-equivalent activities of isoorientin, vitexin, and isovitexin at 1 m M were 31.37, 2.71, and 2.63 unit/mL, respectively. Isoorientin at 1 μ M exhibited over 60% inhibition of MDA formed during copper-mediated human LDL oxidation. Amounts of free MDA in LDL treated with isoorientin, vitexin, isovitexin, and control were 20.06, 40.73, 34.08, and 48.03 n M /mg protein, respectively. These compounds also prolonged the lag phase time of the conjugated diene formation. There was a positive correlation between the free radical scavenging activities and the inhibitory effects on the LDL oxidation of these compounds. These results suggest that the flavone C -glucosides isolated from the leaves of A japonica possess beneficial antioxidant properties against free radicals as well as LDL oxidation.  相似文献
