In this work a sequential multiplex PCR system was designed and validated for the detection of most frequent foodborne pathogen Vibrio species in fish and seafood (Vibrio cholerae, Vibrio parahaemolyticus, Vibrio vulnificus, Vibrio alginoliticus and Vibrio mimicus). The method proposed functions in a hierarchical way, being composed of an end-point multiplex PCR to detect the presence of DNA belonging to the studied species, followed by multiplex PCR and fragment analysis allowing the viability assessment of the detected strains. The final multiplex PCR step of the method may be applied if identification of the serogroup, biotype and/or virulence factor level is necessary. Forty samples of commercial fish and seafood products were used at the method validation stage. Sixty three marine organism samples obtained from various estuarine areas of Spain including shrimps, crabs, bivalve mollusks and fishes were screened for presence of Vibrio species and 2 mussel samples were found positive for V. parahaemolyticus. On the whole, the proposed method is robust and readily adaptable in routine molecular diagnostic laboratories, allowing monitoring and simultaneous detection of all these bacterial pathogens in seafood samples, reducing the expenses and time consumed by other analytical methods. 相似文献

The effect of ultrasound (US) (40 kHz, 200 W, 3 min), blanching (85 °C, 3.5 min), and the combination of both methods was evaluated on the quality of vacuum-packaged potato strips stored at 3 ± 1 °C for up to 10 days. For this study, two cultivars of potatoes were assessed. For blanched Agata samples, the lightness (L*) decreased over 12 % (p < 0.05). Moreover, their hue increased up to 100, obtaining lesser yellow potato strips. In contrast, US did not affect the hue values. The losses of firmness of blanched potato strips were notable (35 % for Agata and 51 % for Agria), whereas US did not change this property (p < 0.05). Nevertheless, no significant differences were found in the total starch content at 10 days. Agata and Agria showed different metabolic behaviors of sucrose in the refrigerated storage. Therefore, Agria cultivar retained better color after frying. These results suggest that US had less impact on color and improve the firmness in vacuum-packaged potato strips with no added chemicals. 相似文献

Germination has been proposed as an economic approach to improve the content of bioactive compounds in pseudocereals. In this work, response surface methodology (RSM) was employed to investigate the impact of germination conditions on the phytochemical content and antioxidant activity of quinoa. The use of desirability methodology showed that the optimum conditions to maximise the content of total phenolic content (TPC) and antioxidant activity in sprouted quinoa were 20 °C for 42 h. Sprouts produced under these conditions exhibited increases of 80% and 30% in TPC and antioxidant activity, respectively, compared to un‐germinated seeds, and contained high γ‐aminobutyric acid (GABA) concentration. The nonsignificant lack‐of‐fit and high determination coefficients obtained confirmed the suitability of the predictive models developed for TPC and antioxidant activity, whilst the one obtained for GABA was not significant (R2 < 0.75) within the conditions studied. Sprouting under optimum conditions enhanced the content of both flavonoid and nonflavonoid compounds, being the increase in flavonoids more pronounced. Kaempferol‐O‐dirhamnosyl‐galactopyranose and quercetin‐O‐glucuronide were the compounds that experienced the most noticeable increase in quinoa after germination. In conclusion, this study provides useful information on the optimum germination conditions to improve the levels of health‐promoting compounds in quinoa. 相似文献

Legumes are the base´s diet in several countries. They hold a high nutritional value, but other properties related to human health are nowadays being studied. The aim of this work was to study the influence of processes (boiling or germination) on the phenolic composition of dark beans (Phaseolus vulgaris L. c.v. Tolosana) and their effect on their antioxidant, neuroprotective and anticancer ability. Phenolic composition of raw and processed dark beans was analysed by HPLC-PAD and HPLC–ESI/MS. The antioxidant activity was evaluated by ORAC. Astrocytes cultures (U-373) have been used to test their neuroprotective effect. Anticancer activities were evaluated on three different cell lines (renal adenocarcinoma (TK-10), breast adenocarcinoma (MCF-7) and melanoma (UACC-62)) by sulphorhodamine B method. Qualitative and quantitative differences in phenolic composition have been observed between raw and processed dark beans that influence the antioxidant activity, mainly for germinated samples which show a decrease of antioxidant capacity. Although every assayed extracts decreased reactive oxygen species release and exhibited cytotoxicity activities on cancer cell lines, raw beans proved to be the most active in neuroprotective and antitumoral effects; this sample is especially rich in phenolic compounds, mainly anthocyanins. This study further demonstrated that phenolic composition of dark beans is related with cooking process and so with their neuroprotective and anticancer activity; cooking of dark beans improves their digestion and absorption at intestinal level, while maintaining its protective ability on oxidative process at cellular level. 相似文献

Tandem cycloisomerization reactions of functionalized 1,6-enynes under indium(III) catalysis are described. This atom-economic transformation proceeds smoothly with 5-exo-dig regioselectivity using commercial In(III) halides and 1,6-enynes furnished with alcohol, carboxylic acid or amine functional groups to give bicyclic structures in good yields and diastereoselectivities. The reaction with enynals involves a three–step mechanism to give an oxatricycle and a conjugated 1,3-diene. In the absence of the internal nucleophile the enyne cycloisomerization evolves through a skeletal rearrangement or a cyclopropanation reaction after the regioselective 5-exo-dig cyclization. The 1,6-enyne cycloisomerization is stereospecific and the stereoselectivity appears to be independent of the internal nucleophile. Experimental data support a common reaction mechanism involving an initial alkyne electrophilic π-coordination of In(III) followed by Markovnikov electrophilic alkene addition and ring-closure by nucleophilic attack. DFT studies hold up a stepwise mechanism involving the formation of a chiral non-classical carbocation intermediate that determines the diastereoselectivity of this tandem cycloisomerization reaction. 相似文献

Various methods of preparation of epoxy resin/clay mixtures, before the addition of the crosslinking agent and curing to form epoxy‐based polymer layered silicate (PLS) nanocomposites, have been investigated to determine their effect on the nanostructure. Organically modified montmorillonite clay was used, and the mixtures were prepared by both simple mixing and solvent‐based methods. X‐ray diffraction shows that intercalation of the resin into the clay galleries occurs for all clay loadings up to 25 wt % and for both preparation methods, but the dispersion of the clay in the resin, observed by optical microscopy, is significantly better for the solvent preparation method. Differential scanning calorimetry (DSC) shows that the intercalated resin has the same molecular mobility as the extra‐gallery resin, but suggests that the intercalated resin does not penetrate completely into the galleries. Prolonged storage of the resin/clay mixtures at room temperature leads to changes in the DSC response, as well as in the response to thermogravimetry, which are interpreted as resulting from homopolymerization of the epoxy resin, catalyzed by the onium ion in the modified clay. This confirms and explains the earlier observation of Benson Tolle and Anderson (J Appl Polym Sci 2004, 91, 89) that “conditioning” of the resin/clay mixtures at ambient temperature has a significant effect when the crosslinking agent is subsequently added, and indicates that the preparation method has important consequences for the nanostructure development in the PLS nanocomposites. © 2006 Wiley Periodicals, Inc. The effect of a reactive diluent (RD) on the kinetics of the curing of an epoxy resin, based on diglycidyl ether of bisphenol A (DGEBA), with a carboxylic anhvdride derived from methyl-tetrahydrophthalic anhydride (MTHPA) catalyzed by a tertiary amine has been studied. The reactive diluent was a low-viscosity aliphatic diglycidyl ether, and the compositions per 100 parts by weight (pbw) of DGEBA were 10, 30, and 50 pbw of RD with the stoichiometric quantity of MTHPA and 1 pbw of catalyst. The curing kinetics was monitored by differential scanning calorimetry (DSC), and the kinetic parameters were determined from the nonisothermal DSC curves by the method described by Málek. The kinetic analysis suggests that the two-parameter autocatalytic model is the more appropriate to describe the kinetics of the curing reaction of this epoxy-anhydride system. The kinetic parameters thus derived satisfactorily simulate both the nonisothermal DSC curves and the isothermal conversion-time plots. Increasing the RD content leads to a small increase in both the nonisothermal and the isothermal heats of curing and has a slight effect on the kinetic parameters E, ln A, m, and n, and, consequently, on the overall reactivity of the system. On the other hand, the increase of the RD content significantly affects the structure of the crosslinked epoxy. It is confirmed that the introduction of aliphatic chains in the structure of the epoxy increases the mobility of the segmental chains in the glass transition region. A location used for 10 years as an uncontrolled olive oil mill wastewater disposal site is studied in this work. Once it was closed the sedimented solid waste on the soil surface was removed. In order to evaluate the influence upon soil characteristics, morphological aspects and analytical parameters of a soil profile from the affected zone are compared to those of a control soil located near the landfill. The residual contamination levels in the underlying soil are determined. The results show that the wastewater infiltration in the soil has caused carbonate dissolution and redistribution and modifications in pH values, electrical conductivity, nutrient contents, phenolic compounds and biological activity of the horizons. Removal of waste, natural leaching and biological activity, in time, led to an effective decrease in electrical conductivity and phenolic compounds, although residual levels can be important even 2 years later. 相似文献
