BACKGROUND: Grape skin cell walls form a barrier against the diffusion of colour. The colour of red wines is mainly due to anthocyanins, although their concentration in wine is not always correlated with the anthocyanin content of grape skins. The cell wall composition changes during fruit ripening, so it was thought that a study of the ripening behaviour of four premium varieties might provide information on how the composition changes during ripening and explain the technological differences. RESULTS: The largest quantities of skin cell wall material (CWM) were isolated from Monastrell grapes, probably owing to the greater number of cell layers they contain and the thicker cell walls. During ripening, a loss of skin CWM (e.g. 120–60 and 65–45 mg CWM g?1 fresh skin for Monastrell J and Cabernet Sauvignon respectively) and galactose (e.g. 36–20 and 55–30 mg g?1 CWM for Cabernet Sauvignon and Monastrell J respectively) was observed, together with a decrease in the degree of pectin methylation and acetylation, except in Syrah, in which pectin methylation remained unchanged. These changes were accompanied by the accumulation of glucose, while other neutral sugars showed no significant variations. A correlation was found between the degree of ripening and those constituents changing in the grape skin cell wall. CONCLUSION: The cell wall composition differs sufficiently between grape varieties to allow discrimination between them. Ripening was associated with a decrease in the quantity of CWM and galactose in the cell walls. Monastrell showed the largest quantities of both throughout ripening. In this work, we have investigated by differential scanning calorimetry the enthalpy relaxation of two poly[methyl(α-n-alkyl)acrylates] in which it is possible to change the length of the two alkyl chains. In particular, we have evaluated the Narayanaswamy parameter, which controls relative contribution of temperature and of structure to the relaxation times, by two methods: Grenet's method (GM) and the peak-shift method (PSM). The data obtained show that both methods lead to equivalent results. Nevertheless, PSM requires fewer experiments than GM, and PSM appears to be more practical. The results obtained on the two acrylates show that the parameter x increases with the lateral chain length, that is to say, that the temperature effects increase as the length of the alkyl chain is increased.  相似文献

In goats, embryo oocyte competence is affected by follicle size regardless the age of the females. In previous studies we have found differences in blastocyst development between oocytes coming of small (<3 mm) and large follicles (>3 mm) in prepubertal (1–2 months-old) goats. Oocyte competence and Follicular Fluid (FF) composition changes throughout follicle growth. The aim of this study was to analyze Fatty Acids (FAs) composition and metabolomic profiles of FF recovered from small and large follicles of prepubertal goats and follicles of adult goats. FAs were analyzed by chromatography and metabolites by 1H-Nuclear Magnetic Resonance (1H-NMR) Spectrometry. The results showed important differences between adult and prepubertal follicles: (a) the presence of α,β-glucose in adult and no detection in prepubertal; (b) lactate, -N-(CH3)3 groups and inositol were higher in prepubertal (c) the percentage of Linolenic Acid, Total Saturated Fatty Acids and n-3 PUFAs were higher in adults; and (d) the percentage of Linoleic Acid, total MUFAs, PUFAs, n-6 PUFAs and n-6 PUFAs: n-3 PUFAs ratio were higher in prepubertal goats. Not significant differences were found in follicle size of prepubertal goats, despite the differences in oocyte competence for in vitro embryo production.  相似文献

The TaqMan real-time PCR has the highest potential for automation, therefore representing the currently most suitable method for screening, allowing the detection of fraudulent or unintentional mislabeling of species. This work describes the development of a real-time polymerase chain reaction (RT-PCR) system for the detection and identification of common octopus (Octopus vulgaris) and main substitute species (Eledone cirrhosa and Dosidicus gigas). This technique is notable for the combination of simplicity, speed, sensitivity and specificity in an homogeneous assay.  相似文献

The antioxidant activity and phytochemical composition (ascorbic acid, free sugars and phenolic compounds) of decoctions and infusions of wild and commercial samples of Tuberaria lignosa (Sweet) Samp. Aerial parts were evaluated and compared. Among wild samples, the effects of the drying method (freeze or shade-drying) on those parameters were studied. Infusion of the freeze-dried wild sample gave the highest levels of sugars, while infusion of shade-dried wild sample and decoction of the freeze-dried sample presented higher ascorbic acid and phenolic compounds content (including ellagitannins and flavonoids) than the other samples. The last two samples also revealed higher antioxidant activity, in some cases even higher than Trolox. Decoctions gave lower amounts of disaccharides than infusions, which seemed to be hydrolysed, increasing the content of monosaccharides. Commercial samples showed the lowest content in phenolic compounds, mainly in ellagitannins and flavonoids, and also the lowest antioxidant activity. This work gives scientific evidence to the traditional medicinal uses of wild Tuberaria lignosa, highlighting the interest of its decoctions and infusions as a source of bioactive compounds and functional beverages.  相似文献

Size has been related with pea seed growth and maturation [1], and it is also considered an important parameter when peas are chosen for canning [2]. Some changes occur in the composition of vegetables during blanching, which are the partial loss of major components [3], that are further reduced during canning, mainly due to leaching into the water used for processing [4]. This leaching can be significant if the brine is discarded and not used as a part of the food consumed, as it is often done with most of canned vegetables [5]. Pea seed is an important source of protein for humans, and it has been suggested [6] that when considering pea seeds protein quality more emphasis should be given to the study of the albumin fraction, because it is the fraction rich in the sulphur amino acids. Although electrophoretic analysis of heat-treated proteins can be considered as time consuming it has been used to monitor changes in specific proteins and mixtures of protein subjected to a variety of heat treatments [7]. In general, heat treatments result in a drastic change in polyacrylamide gel electrophoresis (PAGE) patterns in the presence of the detergent sodium dodecyl sulfate (SDS) by reducing the number of bands compared to untreated samples [8]. The purpose of this study was to monitor changes in the albumin fraction of the protein of 4 sizes of peas in the raw, blanched and canned products, and the can brine solutions, using SDS-PAGE.  相似文献

The fullerene family, and especially C(60), has delighted the scientific community during the last 25 years with perspective applications in a wide variety of fields, including the biological and the biomedical domains. Several biomedical uses have been explored using water-soluble C(60)-derivatives. However, the employment of fullerenes for drug delivery is still at an early stage of development. The design and synthesis of multifunctionalized and multimodal C(60) systems able to cross the cell membranes and efficiently deliver active molecules is an attracting challenge that involves multidisciplinary strategies. Promising results have emerged in the last years, bringing fullerenes again to the front of interest. Herein, the state of the art of this emerging field is presented and illustrated with some of the most representative examples.  相似文献

A series of colour assays were conducted taking into account old bibliographic recommendations and oral testimony regarding traditional lime painting technology from the Alentejo (South Portugal) region. The main objectives were: (a) the colorimetric analysis of paintings made with different limewash preparation methods; (b) the comparison between natural processed red and yellows ochre pigments (“home made” and from industrial origin) and synthetic iron oxides in different concentrations and lime proportions; (c) the evaluation of the colorimetric CIELAB coordinates during the lime carbonation process in coloured mortars and limewashes. The results shows that the evolution of L*, a*, and b* during the carbonation process is independent of pigment concentration and paint thickness but dependent of pigment composition and finishing technique (lime painting or coloured mortar).
