Time domain analysis of heart period variability in patients without structural heart disease demonstrated increased parasympathetic modulation before paroxysmal atrial fibrillation (AF) occurring predominantly at night. However, diurnal differences in autonomic activity preceding AF episodes in a diverse patient population have not been assessed. Accordingly, we performed spectral analysis of heart period variability on Holter recordings during sinus rhythm preceding AF in 29 patients, 17 with night and 12 with day episodes. Samples taken 5, 10, and 20 minutes before AF onset were compared. Normalized high-frequency (HF) spectral power change was greater when comparing the interval 10 to 5 minutes with 20 to 10 minutes preceding AF in 26 of 29 patients (0.09 +/- 0.07 vs 0.03 +/- 0.02; p < 0.0001). HF spectral power increased before 3 of 12 AF episodes during the day compared with 15 of 17 AF episodes during the night (p = 0.001). Nocturnal AF episodes were preceded by increased HF spectral power in the 5- versus the 20-minute sample expressed as natural logarithm-transformed values (5.6 +/- 4.8 vs 4.2 +/- 4.0; p < 0.005) and normalized values (0.19 +/- 0.09 vs 0.10 +/- 0.07; p < 0.02), a decrease in low-frequency/HF ratio (1.05 +/- 0.61 vs 2.21 +/- 1.75; p < 0.05) and heart rate (60 +/- 13 vs 71 +/- 13 beats/min; p = 0.06). Structural heart disease was more common with daytime than nocturnal AF episodes (58% vs 18%, p < 0.05). In conclusion, HF spectral power change was increased preceding most AF episodes. However, diurnal differences were demonstrated. Contrary to daytime AF, increased parasympathetic activity preceded predominantly nocturnal AF, mostly in younger patients with structurally normal hearts.  相似文献

Mutations in the apical Na-K-2Cl co-transporter, NKCC2, cause type I Bartter syndrome (BS1), a life-threatening kidney disease. We have previously demonstrated that the BS1 variant Y998X, which deprives NKCC2 from its highly conserved dileucine-like motifs, compromises co-transporter surface delivery through ER retention mechanisms. However, whether these hydrophobic motifs are sufficient for anterograde trafficking of NKCC2 remains to be determined. Interestingly, sequence analysis of NKCC2 C-terminus revealed the presence of consensus di-acidic (D/E-X-D/E) motifs, 949EEE951 and 1019DAELE1023, located upstream and downstream of BS1 mutation Y998X, respectively. Di-acidic codes are involved in ER export of proteins through interaction with COPII budding machinery. Importantly, whereas mutating 949EEE951 motif to 949AEA951 had no effect on NKCC2 processing, mutating 1019DAE1021 to 1019AAA1021 heavily impaired complex-glycosylation and cell surface expression of the cotransporter in HEK293 and OKP cells. Most importantly, triple mutation of D, E and E residues of 1019DAELE1023 to 1019AAALA1023 almost completely abolished NKCC2 complex-glycosylation, suggesting that this mutant failed to exit the ER. Cycloheximide chase analysis demonstrated that the absence of the terminally glycosylated form of 1019AAALA1023 was caused by defects in NKCC2 maturation. Accordingly, co-immunolocalization experiments revealed that 1019AAALA1023 was trapped in the ER. Finally, overexpression of a dominant negative mutant of Sar1-GTPase abolished NKCC2 maturation and cell surface expression, clearly indicating that NKCC2 export from the ER is COPII-dependent. Hence, our data indicate that in addition to the di-leucine like motifs, NKCC2 uses di-acidic exit codes for export from the ER through the COPII-dependent pathway. We propose that any naturally occurring mutation of NKCC2 interfering with this pathway could form the molecular basis of BS1.  相似文献

Previously, we developed a simple procedure of intracameral injection of silicone oil (SO) into mouse eyes and established the mouse SOHU (SO-induced ocular hypertension under-detected) glaucoma model with reversible intraocular pressure (IOP) elevation and significant glaucomatous neurodegeneration. Because the anatomy of the non-human primate (NHP) visual system closely resembles that of humans, it is the most likely to predict human responses to diseases and therapies. Here we tried to replicate the mouse SOHU glaucoma model in rhesus macaque monkeys. All six animals that we tested showed significant retinal ganglion cell (RGC) death, optic nerve (ON) degeneration, and visual functional deficits at both 3 and 6 months. In contrast to the mouse SOHU model, however, IOP changed dynamically in these animals, probably due to individual differences in ciliary body tolerance capability. Further optimization of this model is needed to achieve consistent IOP elevation without permanent damage of the ciliary body. The current form of the NHP SOHU model recapitulates the severe degeneration of acute human glaucoma, and is therefore suitable for assessing experimental therapies for neuroprotection and regeneration, and therefore for translating relevant findings into novel and effective treatments for patients with glaucoma and other neurodegenerations.  相似文献

The electrothermal process of magnesium metal production is a promising route, where large sized internally heated reactor is used for magnesium production resulting in less energy and labour intensive and high space-time yield process. However, the dissolution behavior of dolime in the electrothermal slag has been found critical for the process optimization. In this paper, the dissolution kinetics of the dolime in the slag was discussed. Quaternary slag (CaO-Al2O3-SiO2-MgO) was prepared having basicity CaO/SiO2 ≥ 1.8 and Al2O3/SiO2 ≥ 0.26 for dolime dissolution studies by static hot dip method. Prior to the experiments, FactSage calculations were carried out varying temperatures and slag compositions. In the kinetic studies, dolime particles 10–15 mm size was added in slag melted at 1450, 1500 and 1550°C and samples were taken at various time intervals. The chemical analysis of slag sample was carried out to investigate the dissolution kinetics to establish the rate expression. The activation energy for the process was calculated for different models used in study and was found to be in the range of 130–270 kJ/mol. SEM analysis was done for surface analysis of reacted particles. This study would be helpful in optimizing the dolime charging rate during pilot scale trials for electrothermal magnesium production at CSIR-NML, Jamshedpur.  相似文献

In galvanising line of cold rolling mill, mechanical properties, i.e. yield strength (YS) and ultimate tensile strength (UTS), are achieved by controlling the key process parameters within specified limits. In this paper, a feed-forward back-propagation artificial neural network (ANN) is proposed to predict the mechanical properties of a coil from its chemical composition, thickness, width and key galvanising process parameters. Principal component analysis is used to avoid redundancy and collinearity effects in input variables for the ANN. The model predicted the YS and UTS with an accuracy of ±10?megapascal (MPa) for 90% of the data. The model was implemented in the continuous galvanising line of Tata Steel, India. An online quality monitoring system was developed to monitor the predicted mechanical properties and process parameters of a galvanised coil. In this paper, we propose a speech enhancement method where the front-end decomposition of the input speech is performed by temporally processing using a filterbank. The proposed method incorporates a perceptually motivated stationary wavelet packet filterbank (PM-SWPFB) and an improved spectral over-subtraction (I-SOS) algorithm for the enhancement of speech in various noise environments. The stationary wavelet packet transform (SWPT) is a shift invariant transform. The PM-SWPFB is obtained by selecting the stationary wavelet packet tree in such a manner that it matches closely the non-linear resolution of the critical band structure of the psychoacoustic model. After the decomposition of the input speech, the I-SOS algorithm is applied in each subband, separately for the estimation of speech. The I-SOS uses a continuous noise estimation approach and estimate noise power from each subband without the need of explicit speech silence detection. The subband noise power is estimated and updated by adaptively smoothing the noisy signal power. The smoothing parameter in each subband is controlled by a function of the estimated signal-to-noise ratio (SNR). The performance of the proposed speech enhancement method is tested on speech signals degraded by various real-world noises. Using objective speech quality measures (SNR, segmental SNR (SegSNR), perceptual evaluation of speech quality (PESQ) score), and spectrograms with informal listening tests, we show that the proposed speech enhancement method outperforms than the spectral subtractive-type algorithms and improves quality and intelligibility of the enhanced speech.  相似文献

We give a family of simultaneous confidence bands for the regression line α + βX for all X under the random intercept regression model. These types of models frequently arise in engineering and econometrics. The shape of the bands is controlled by the user, so many possible bands are obtainable. As an illustration, an engineering application involving a wind tunnel test of an aircraft design is described.
