Extracellular β-glucosidase was produced using coffee pulp as a sole carbon source by Penicillium verrucosum by solid state fermentation and 897.36±59 U/g enzyme activity was obtained. Increase in 2.21-fold of enzyme activity on optimizing the bioprocess parameters by response surface methodology based on central composite rotatable design is illustrated. Maximum production level of 1,991.17 U/g was obtained with optimum values of pH 4.2, moisture 66.8%, and fermentation duration of 56 h. The enzyme was partially purified and the enzyme activity was optimum at 50°C temperature and at pH 6. The metal ions such as Mg2+, Zn2+, Ca2+, K+, detergents, and chelator such as EDTA were effective and further increased the β-glucosidase activity. On application of β-glucosidase for simultaneous saccharifiation and fermentation, 3.3% ethanol was obtained. Thus, this study provides insight on exploitation of P. verrucosum for synthesis of of β-glucosidase using coffee pulp which is available abundantly in coffee processing industries.

The enzyme amylase is one of the hydrolyzing enzymes used in detergent formulation in order to remove soil based on polysaccharides. The effectiveness of the enzyme depends on its compatibility with other ingredients of the formulation. Among the studied additives, comprising anionic surfactants sodium dodecyl hydrogen sulfate (SDS) and dioctyl sodium sulfosuccinate, the cationic surfactant cetyl trimethyl ammonium bromide (CTAB), nonionic surfactants polyoxyethylene sorbitan monooleate and polyoxyethylene octyl phenyl ether, carboxy methyl cellulose and sodium sulfate, only the anionic surfactant SDS and cationic surfactant CTAB showed catalytic enhancement of α-amylase. The kinetic parameters, K m and k cat, showed an increase in catalytic activity in the micellar pseudophase. The decrease in optimum temperature from 55 to 30 °C and the shift in optimum pH from 5.5 to 7 on the addition of SDS and CTAB for the hydrolysis of starch are very favorable to enhance the washing characteristics.  相似文献

Five new photocatalysts have been synthesized in order to extend the photo response upto visible range, by adsorbing MC113-MC117 ruthenium complexes on TiO2-Pt composites. Highlight harvesting properties of these ruthenium complexes instigated us to evaluate for photocatalytic activity. The absorption curves of the synthesized photocatalysts extended up to 750 nm. Morphological studies of photocatalysts have been carried out using SEM and powder X-ray crystallography. Among all photocatalysts, MC113PC showed high photocatalytic activity i.e. 9474 TONs. IPCE and fluorescence quenching studies of the catalysts revealed the light harvesting nature and electron injection efficiency. The photocatalytic activity of MC photocatalysts were systematically screened at different pH and employing different sacrificial electron donors (SED) in order to obtain optimal photocatalytic performance.  相似文献
