Preovulatory follicles were explanted on the day before ovulation from immature rats given a single injection of Pregnant Mare's Serum gonadotropin (PMS) 2 days earlier. The follicles were incubated for 4 h in modified Krebs bicarbonate buffer containing glucose and albumin in absence or presence of ovine luteinizing hormone (NIH-LH-S18; 0.1-10 mug/ml). The accumulation of progresterone, androstenedione and 17beta-estradiol in the medium was determined by radioimmunoassay. As in indicator of LH exposure the meiotic stage of the follicle-enclosed oocyte was determined at recovery by interference contrast microscopy. The first group of follicles were explanted in the morning, before the endogenous gonadotrophin surge. In hormone-free medium the oocytes remained in the dictyate stage, whereas addition of LH induced oocyte maturation. These follicles, when incubated in hormone-free medium, secreted predominantly androstenedione and estradiol and only low amounts of progesterone. In the presence of LH the secretion of all steroids was enhanced. The second group of follicles were explanted in the evening, 2-4 h after the endogenous gonadotrophin surge. After incubation in hormone-free medium the follicle-enclosed oocytes had matured. The steroid secretion by the follicles was different from that of the first group. In hormone-free medium they secreted predominantly progesterone and low amounts of androstenedione and estradiol. Addition of LH to the medium caused further enhancement of progesterone secretion, but had no effect on androstenedione and estradiol secretion. The third group of follicles were explanted in the evening from rats in which the preovulatory gonadotrophin surge had been prevented by Nembutal treatment. Oocyte maturation and steroid secretion did not differ from that found for the first group of follicles explanted in the morning. The results are compatible with the hypothesis that LH, after a transitory stimulation, inhibits androgen and estrogen secretion and stimulates progesterone secretion by the preovulatory ovarian follicle. 相似文献

Decision Support Systems (DSSs) in the domain of Industrial Engineering and Management can be very powerful tools, but more knowledge about the way users utilize these systems is required. In the current research, we evaluated the effect of previous experience with a task on users’ willingness to accept a suggested decision aid and their performance in a supply chain management system. Participants were randomly assigned to one of three between-participants groups: the No Aid Group, which did not receive any decision aid during the interaction; the Aid Group, which was offered an algorithm, at the beginning of the first session, to help members improve their decisions; and the Mid-term Aid Group, which received, at the beginning of the second session, the same algorithm that was offered to the Aid Group. Participants were tested separately. The results demonstrated that in the second session, the performance of the Aid Group was significantly better in comparison with the No Aid Group. The Mid-term Aid Group’s performance did not show any gain, although both the Aid and Mid-term Aid Groups made similar use of the aid in this session. We concluded that in a DSS, having previous experience with the task before introducing the decision aid compared to having previous experience with the task and with the decision aid is not recommended. 相似文献

Cognitive deficits are core symptoms of schizophrenia but remain poorly addressed by dopamine-based antipsychotic medications. Glutamate abnormalities are implicated in schizophrenia-related cognitive deficits. While the role of the NMDA receptor has been extensively studied, less attention was given to other components that control glutamate homeostasis. Glutamate dynamics at the tripartite synapse include presynaptic and postsynaptic components and are tightly regulated by neuron–astrocyte crosstalk. Here, we delineate the role of glutamate homeostasis at the tripartite synapse in schizophrenia-related cognitive dysfunction. We focus on cognitive domains that can be readily measured in humans and rodents, i.e., working memory, recognition memory, cognitive flexibility, and response inhibition. We describe tasks used to measure cognitive function in these domains in humans and rodents, and the relevance of glutamate alterations in these domains. Next, we delve into glutamate tripartite synaptic components and summarize findings that implicate the relevance of these components to specific cognitive domains. These collective findings indicate that neuron–astrocyte crosstalk at the tripartite synapse is essential for cognition, and that pre- and postsynaptic components play a critical role in maintaining glutamate homeostasis and cognitive well-being. The contribution of these components to cognitive function should be considered in order to better understand the role played by glutamate signaling in cognition and develop efficient pharmacological treatment avenues for schizophrenia treatment-resistant symptoms. 相似文献

Assessment of contamination potential of lettuce by Salmonella enterica serovar Newport added to the plant growing medium 总被引:1,自引:0,他引:1
The capacity of Salmonella enterica serovar Newport to contaminate Romaine lettuce (Lactuca sativa L. cv. Nogal) via the root system was evaluated in 17-, 20-, and 33-day-old plants. Apparent internalization of Salmonella via the root to the above-ground parts was identified in 33- but not 17- or 20-day-old plants and was stimulated by root decapitation. Leaves of lettuce plants with intact and damaged roots harbored Salmonella at 500 +/- 120 and 5,130 +/- 440 CFU/g of leaf, respectively, at 2 days postinoculation but not 5 days later. These findings are first to suggest that Salmonella Newport can translocate from contaminated roots to the aerial parts of lettuce seedlings and propose that the process is dependent on the developmental stage of the plant. 相似文献

Daily stress interacts with trait dissociation to predict sleep-related experiences in young adults.
Building on the previously documented effects of stress and dissociation on sleep and dreaming, we examined their interactive role in general sleep-related experiences (GSEs; e.g., nightmares, falling dreams, hypnagogic hallucinations; see Watson, 2001). Stress, sleep quality, and GSEs were assessed daily for 14 days among young adults. Baseline assessment included life stress, sleep quality, psychopathology, dissociation, and related dimensions. Multilevel analyses indicated that daily stress brings about GSEs among highly dissociative young adults. Additionally, baseline trait dissociation predicted within-subject elevation in GSEs when daily stress was high. Flawed sleep–wake transitions, previously linked to dissociation and sleep-related experiences, might account for this effect. (PsycINFO Database Record (c) 2011 APA, all rights reserved) 相似文献
