We established a differentiation method for homogeneous α7 integrin-positive human skeletal muscle stem cell (α7+hSMSC)-derived osteoblast-like (α7+hSMSC-OB) cells, and found that interleukin (IL)-1β induces matrix metalloproteinase (MMP)-13-regulated proliferation of these cells. These data suggest that MMP-13 plays a potentially unique physiological role in the regeneration of osteoblast-like cells. Here, we examined whether up-regulation of MMP-13 activity by IL-1β was mediated by Wingless/int1 (Wnt) signaling and increased the proliferation of osteoblast-like cells. IL-1β increased the mRNA and protein levels of Wnt16 and the Wnt receptor Lrp5/Fzd2. Exogenous Wnt16 was found to increase MMP-13 mRNA, protein and activity, and interestingly, the proliferation rate of these cells. Treatment with small interfering RNAs against Wnt16 and Lrp5 suppressed the IL-1β-induced increase in cell proliferation. We revealed that a unique signaling cascade IL-1β→Wnt16→Lrp5→MMP-13, was intimately involved in the proliferation of osteoblast-like cells, and suggest that IL-1β-induced MMP-13 expression and changes in cell proliferation are regulated by Wnt16. 相似文献

The formation of OH radicals (OH) for polymer electrolyte fuel cell (PEFC) was investigated with a homemade test cell by means of a fluorescence probe method for several kinds of membrane electrode assembly (MEA) consisting of Nafion. After 12-h operation under open circuit condition, OH was detected at both the anode and the cathode sides but the amount was much larger for the anode side. The formation of OH considerably depended on the amount of Pt/C catalyst loaded in the MEA. Dispersion of Pt particles in Nafion membranes increased the OH formation, but Pt ions did not. Little is known about the development of the olfactory organs of camel. In this study, prenatal development and neuronal differentiation of the vomeronasal organ (VNO) and the olfactory epithelium (OE) of the one‐humped camel were studied by immunohistochemistry and lectin histochemistry. A neuronal marker, protein gene product (PGP) 9.5, but not a marker of fully differentiated olfactory receptor cells, olfactory marker protein, intensely labeled the olfactory receptor cells of the VNO and OE at 395 mm, 510 mm, and 530 mm fetal ages, indicating that the olfactory receptor cells are differentiated, but not fully matured both in the VNO and the OE. In 187 mm and 190 mm fetuses, PGP 9.5 yielded faint immunoreactive signals in the VNO, but not in the OE, although the presence of olfactory receptor cells were demonstrated in both tissues by intense WGA and LEL stainings. We conclude that the camel VNO and OE bear differentiated, but still immature receptor cells; in addition, the onset of neuronal differentiation seems to be somewhat earlier in the VNO than in the OE till half of the prenatal life. Helium irradiation experiments of V–4Ti alloy were conducted in an ECR ion irradiation apparatus by using helium ions with energy of 5 keV. The ion fluence was in the range from 1 × 1017 He/cm2 to 8 × 1017 He/cm2. After the helium ion irradiation, the helium retention was examined by using a technique of thermal desorption spectroscopy (TDS). After the irradiation, the blisters with a size of about 0.1 μm were observed at the surface, and the blister density increased with the ion fluence. Two desorption peaks were observed at approximately 500 and 1200 K in the thermal desorption spectrum. When the ion fluence was low, the retained helium desorbed mainly at the higher temperature regime. As increase of the ion fluence, the desorption at the lower temperature peak increased and the retained amount of helium saturated. The saturated amount was approximately 2.5 × 1017 He/cm2. In previous studies, many fatigue tests have been performed to investigate the fatigue behaviour of out-of-plane gusset welded joints. However, fatigue behaviour of out-of-plane gusset joints with a one-side fillet weld has not yet been clarified sufficiently. In this research, in order to quantitatively clarify the behaviour, welding residual stress at the weld toes of the joint was measured by a cutting method. In addition to this, fatigue tests were carried out on the joints with a one-side weld and turn-around weld. The plate thickness is 9 mm and the weld size is 10 mm. Moreover, finite element stress analyses were conducted on the test specimen models and parametric models with different main plate and gusset plate thickness. Their results show that fatigue cracks for out-of-plane gusset joints with a one-side fillet weld initiate from both weld toes at the start and end points of weld beads and non-penetration weld of the gusset side. Also, the crack initiation points are different depending on the weld arrangement; however, their fatigue strengths are almost same. 相似文献
