CD40 engagement induces a variety of functional outcomes following association with adaptor molecules of the TNF receptor-associated factor (TRAF) family. Whereas TRAF2, -5, and -6 initiate NF-kappaB activation, the outcomes of TRAF3-initiated signaling are less characterized. To delineate CD40-induced TRAF3-dependent events, Ramos B cells stably transfected with a dominant negative TRAF3 were stimulated with membranes expressing recombinant CD154/CD40 ligand. In the absence of TRAF3 signaling, activation of p38 and control of Ig production were abrogated, whereas Jun N-terminal kinase activation and secretion of IL-10, lymphotoxin-alpha, and TNF-alpha were partially blocked. By contrast, induction of apoptosis, activation of NF-kappaB, generation of granulocyte-macrophage CSF, and up-regulation of CD54, MHC class II, and CD95 were unaffected by the TRAF3 dominant negative. Together, these results indicate that TRAF3 initiates independent signaling pathways via p38 and JNK that are associated with specific functional outcomes.  相似文献

Search Engine Marketing (SEM) is the practice of manipulating and/or paying the search engines (Google, Yahoo!, Bing, etc.) to drive traffic to websites. The SEM community has developed techniques that can channel visitors to websites; however, little work has been done to develop models able to estimate the amount of traffic generated by SEM. In this paper, we develop formulae that can be used to estimate traffic resulting from SEM campaigns that can be used by search marketing agencies for competition analysis and by web hosting providers for performance analysis and capacity planning. Our experimental results show that our models work best for targeted marketing campaigns, but the formulae presented can be generalized to broader marketing domains.  相似文献

New heuristics and strategies have enabled major advancements in SAT solving in recent years. However, experimentation has shown that there is no winning solution that works in all cases. A degradation of orders of magnitude can be observed if the wrong heuristic is chosen. The problem is that it is impossible to know, in advance, which heuristics are best for a given problem. Consequently, many ideas - those that turn out to be useful for a small subset of the cases, but significantly increase run times on most others - are discarded.We propose the notion of Adaptive Solving as a possible solution to this problem. In our framework, the SAT solver monitors the effectiveness of the search on-the-fly using a Performance Metric. The metric gives a score according to its assessment of the search progress. Based on this score, one or more heuristics are turned on or off. The goal is to use a specific heuristic or strategy when it is advantageous, and turn it off when it is not, before it does too much damage. We suggest several possible metrics, and compare their effectiveness. Our adaptive solver achieves significant speedups on a large set of examples. We also show that applying different heuristics on different parts of the search space can improve run times even beyond what can be achieved by the best heuristic on its own.  相似文献

During space missions, such as the prospective Mars mission, crew labor time is a strictly limited resource. The diet for such a mission (based on crops grown in a bioregenerative life support system) will require astronauts to prepare their meals essentially from raw ingredients. Time spent on food processing and preparation is time lost for other purposes. Recipe design and diet planning for a space mission should therefore incorporate the time required to prepare the recipes as a critical factor. In this study, videotape analysis of an experienced chef was used to develop a database of recipe preparation time. The measurements were highly consistent among different measurement teams. Data analysis revealed a wide variation between the active times of different recipes, underscoring the need for optimization of diet planning. Potential uses of the database developed in this study are discussed and illustrated in this work.  相似文献

OBJECTIVE: To ascertain the prevalence of fibromyalgia syndrome (FMS) in systemic lupus erythematosus (SLE) and to evaluate its clinical impact and relationship to SLE disease activity. METHODS: A cross-sectional analysis of 102 patients from a public hospital SLE clinic. Information was obtained on symptoms of FMS, disability, tender points, pain thresholds, and SLE disease activity. RESULTS: Twenty-two SLE patients (22%) met the American College of Rheumatology criteria for FMS, and another 24 (23%) had clinical FMS but did not meet the classification criteria. The patients who met the criteria for FMS had a significantly increased frequency and severity of symptoms commonly associated with FMS, and were much more likely to be unable to perform daily activities. The FMS patients also were less likely to be employed, and more likely to be divorced and to be receiving welfare or medical disability benefits. However, patients with and those without FMS did not differ in measures of SLE activity. CONCLUSION: FMS is very common in SLE patients, and accounts for many of the symptoms and much of the disability in these patients.  相似文献

To determine whether part of the anti-inflammatory effects of prostaglandin E2 (PGE2) was related to inhibition of T cell interactions with endothelial cells (EC), the effects of PGE2 and other cAMP-elevating agents on the transendothelial migration of human T cells was examined. Although PGE2 did not effect T cell binding to EC, concentration-dependent inhibition of the transendothelial migration of T cells through unstimulated or IL-1-activated EC was observed. PGE2 inhibited the function of both T cells and EC, with maximal inhibition observed when both T cells and EC were treated with PGE2. However, the inhibitory action of PGE2 could not be ascribed to an effect on the adhesion receptor pair, CD11a/CD18-CD54. The inhibitory effect of PGE2 seemed to relate to its capacity to elevate cellular cAMP levels, because 3-isobutyl-1-methylxanthine enhanced PGE2 activity and dibutyryl cAMP and forskolin also inhibited transendothelial migration. The inhibitory effect of PGE2 and the other cAMP-elevating agents on the function of T cells related in part to suppression of their intrinsic locomotory behavior as random migration in the absence of EC was blocked. In EC, PGE2 and the other cAMP-elevating agents increased the barrier function of EC as evidenced by a decrease in the diffusion of [3H]mannitol through the endothelium. These results indicate that part of the anti-inflammatory action of PGE2 relates to its capacity to suppress the transendothelial migration of T cells by cAMP-mediated alterations in the function of both T cells and EC.  相似文献

Cookstoves are a major source of black carbon (BC) particles and associated organic compounds, which influence the atmospheric radiative balance. We present results from experiments that characterize BC emissions from a rocket stove coated with secondary organic aerosol. Optical properties, namely, BC mass absorption cross-section (MACBC) and mass scattering cross-section (MSC), as a function of the organic-to-black carbon ratio (OA:BC) of fresh and aged cookstove emissions were compared with Mie and Rayleigh–Debye–Gans (RDG) calculations. Mie theory reproduced the measured MACBC across the entire OA:BC range. However, Mie theory failed to capture the MSC at low OA:BC, where the data agreed better with RDG, consistent with a fractal morphology of fresh BC aggregates. As the OA:BC increased, the MSC approached Mie predictions indicating that BC-containing particles approach a core–shell structure as BC cores become heavily coated. To gain insight into the implications of our findings, we calculated the spectral simple forcing efficiency (dSFE) using measured and modeled optical properties as inputs. Good agreement between dSFE estimates calculated from measurements and Mie-modeled dSFE across the entire OA:BC range suggests that Mie theory can be used to simulate the optical properties of aged cookstove emissions.

Land use and landscape structure changes were investigated using the example of an agricultural landscape in central Bohemia. Attention was paid particularly to the great changes which occurred during the 40 years of socialist collectivism. The analysis of landscape development shows that statistics about land use can give only general information about landscape macrostructure and cannot provide a perfect idea of the actual spatial composition of landscape elements. Landscape microstructure expressed in spatial arrangements, shape, size, quality and connectivity of patches, lines and small interactive elements plays the main role in landscape dynamics and is the principal influence in landscape stability.  相似文献

The Information Bottleneck is an information theoretic framework that finds concise representations for an ‘input’ random variable that are as relevant as possible for an ‘output’ random variable. This framework has been used successfully in various supervised and unsupervised applications. However, its learning theoretic properties and justification remained unclear as it differs from standard learning models in several crucial aspects, primarily its explicit reliance on the joint input–output distribution. In practice, an empirical plug-in estimate of the underlying distribution has been used, so far without any finite sample performance guarantees. In this paper we present several formal results that address these difficulties. We prove several finite sample bounds, which show that the information bottleneck can provide concise representations with good generalization, based on smaller sample sizes than needed to estimate the underlying distribution. The bounds are non-uniform and adaptive to the complexity of the specific model chosen. Based on these results, we also present a preliminary analysis on the possibility of analyzing the information bottleneck method as a learning algorithm in the familiar performance-complexity tradeoff framework. In addition, we formally describe the connection between the information bottleneck and minimal sufficient statistics.  相似文献
