RNA G-quadruplex (GQs) sequences in 5′-UTRs of certain proto-oncogenes co-localize with hairpin (Hp) forming sequences resulting in intramolecular Hp-GQ conformational equilibria, which is suggested to regulate cancer development and progression. Thus, regulation of Hp-GQ equilibria with small molecules is an attractive but less explored therapeutic approach. Herein, two tetraphenylethene (TPE) derivatives, TPE−Py and TPE-MePy, were synthesized and their effect on Hp-GQ equilibrium was explored. FRET, CD and molecular docking experiments suggest that cationic TPE-MePy shifts the Hp-GQ equilibrium significantly towards the GQ conformer mainly through π-π stacking and van der Waals interactions. In the presence of TPE-MePy, the observed rate constant values for first and second folding steps were increased up to 14.6 and 2.6-fold, respectively. The FRET melting assay showed a strong stabilizing ability of TPE-MePy (ΔTm=4.36 °C). Notably, the unmethylated derivative TPE−Py did not alter the Hp-GQ equilibrium. Subsequently, luciferase assay analysis demonstrated that the TPE-MePy derivatives suppressed the translation efficiency by ∼5.7-fold by shifting the Hp-GQ equilibrium toward GQ conformers in the 5’-UTR of TRF2. Our data suggests that HpGQ equilibria could be selectively targeted with small molecules to modulate translation for therapy.  相似文献

Thermal history of droplets associated with gas atomization of melt has been investigated. A mathematical model, based on classical theory of heterogeneous nucleation and volume separation of nucleants among droplets size distribution, is described to predict undercooling of droplets. Newtonian heat flow condition coupled with velocity dependent heat transfer coefficient is used to obtain cooling rate before and after nucleation of droplets. The results indicate that temperature profile of droplets in the spray during recalescence, segregated and eutectic solidification regimes is dependent on their size and related undercooling. The interface temperature during solidification of undercooled droplets rapidly approaches the liquidus temperature of the alloy with a subsequent decrease in solid-liquid interface velocity. A comparison in cooling rates of atomized powder particles estimated from secondary dendrite arm spacing measurements are observed to be closer to those predicted from the model during segregated solidification regime of large size droplets.  相似文献

The alkylation of phenol with tert‐butyl alcohol was carried out in a continuous flow reactor over a catalyst synthesized from fly ash. The activity of the synthesized catalyst was compared with those of other conventional zeolite catalysts such as 13X (NaX) and Hβ. Of all the catalysts tested, zeolite Hβ showed the highest activity in phenol conversion followed by the synthesized zeolite (HZOP‐31). The activity of commercial 13X zeolite was found to be same as that of HZOP‐31. Ce‐exchanged catalyst (CeZOP‐31) showed even better performance than 13X in the alkylation of phenol. The effects of different parameters such as reactant mole ratio, temperature and space velocity on phenol conversion and tert‐butyl phenol selectivity were studied. The effect of mass transfer resistance was found to be negligible within the feed rate range and particle size range studied. The apparent activation energy for the reaction of tert‐butyl alcohol over HZOP‐31 was determined as 30.1 kJ mol?1. Copyright © 2006 Society of Chemical Industry  相似文献

The formation of micro-sized droplets in the matrix of the primary phase of Ag–Ge alloys and their undercooling behaviour were investigated. Thermal cycling consisted in equilibration of hypoeutectic alloys near the eutectic temperature in the two-phase liquid–solid region followed by up-quenching of alloys at 30–50°C above the eutectic temperature and their subsequent slow cooling. A wide size range of droplets was generated in the matrix of the silver-rich phase by repeated thermal cycling of the alloy. During slow cooling, droplets of silver-rich liquid exhibited a maximum undercooling of 81°C below the eutectic temperature. Examination of the solidification structure of the undercooled alloys, by X-ray diffraction, polarized light microscopy and electron probe micro analysis techniques, indicated the presence of a metastable phase. The mechanism of formation of droplets and the nature of non-equilibrium solidification of the undercooled melt of silver–germanium alloys, are discussed.  相似文献

Breath alcohol analyser is used to detect alcohol content in end-expiratory breaths in order to enforce driving regulations under the influence of alcohol legislation. The accuracy and reliability of the routine measurements of alcohol content performed with breath alcohol analyser can be achieved by the calibration of the breath alcohol analyser using standards traceable to SI reference material. Proper calibration is essential for transparency in legal verification for which reference material is needed. At international level, a number of NMIs are active to address this important measurement issue of providing accurate measurements. Several international key comparison programs have been organized so far for the determination of ethanol content in aqueous and in nitrogen/air matrix. NIST, USA; BAM, Germany; IRMM, Belgium, Portugal, INMETRO, Brazil, LGC, UK etc. have developed certain reference materials of ethanol in water solution/air with different concentration ranges. However, no such reference material is introduced in India as an indigenous standard, rather, being procured from abroad or using high purity alcohol for calibration purposes. CSIR-NPL, India, being the NMI is now focusing on establishing the calibration facility and development of SI traceable aqueous alcohol standard to provide test reliability for the testing in breath alcohol analyser. This program has a societal impact which contributes to human health and regulatory needs for the nation.  相似文献

Carcinomas of unknown primary site are microscopically confirmed metastatic epithelial malignancies with no identified primary site at the onset of treatment. Their incidence is about 5% among all cancer patients. They represent a group of heterogeneous tumors with low chemosensitivity and poor outcome: the overall median survival is about 6 months. The search for primary tumor should be limited to the identification of subgroups of patients requiring specific therapies: 1) patients with cervical lymph nodes containing squamous carcinoma; 2) women with axillary lymph nodes containing adenocarcinoma; 3) women with peritoneal adenocarcinomatosis; 4) young men with poorly differentiated carcinoma of the midline; 5) patients with neuroendocrine metastasis. Other patients are to be managed with symptomatic procedures and possibly chemotherapy. Simple prognostic factors such as performance status, histology and serum levels of alkaline phosphatase may help the physician to select treatment strategies with the aim of preserving an optimal quality of life.  相似文献

The spray deposition process has been employed in synthesis of Al-4.5Cu-10Al2O3 and Al-4.5Cu-10Al2O3-10Pb based composites. The microstructure and wear characteristics of composites were investigated. The rapid solidification inherent in spray deposition processing resulted in a uniform dispersion of Al2O3 and Pb particles co-existing in the matrix of the- primary α-phase. The grain size of the Al-4.5Cu-10Al2O3-Pb composite was observed to be higher than that of the Al-4.5Cu-10Al2O3 composite in various sections of the spray deposit. The wear rate of composite materials decreased with addition of Pb phase. This behavior is discussed in the light of the microstructural modification induced by spray deposition and the morphology of debris particles on the wear track surfaces. The wear characteristics of the composites are compared with that of the liquid immiscible Al-4.5Cu-10Pb alloy.  相似文献

In the present work a thermodynamic model is presented for the nucleation of metastable Ag3Ge phase from the undercooled melt in Ag-Ge alloy system. It is shown that under kinetically induced favourable condition, nucleation of a single phase compound with an approximate stoichiometry Ag3Ge has greater affinity for nucleation over Ag and Ge based phases requiring diffusion in the liquid.  相似文献

This paper deals with an analysis of a normally impinging micropolar fluid jet on a curved surface. The flow near the stagnation point in the impingement region is divided into inviscid and viscous flow regions. The inviscid flow solution is governed by Euler's equations of motion expressed in curvilinear coordinate system. The viscous flow solution is governed by the zeroth and the first order boundary layer equations. These boundary layer equations are solved by assuming power series expansions for both velocity and microrotation fields which give rise to two systems of ordinary coupled differential equations. The effects of surface curvature and material parameters on boundary layer characteristics have been studied and presented graphically. The gradients of zeroth order velocity and microrotation at the wall decrease and the zeroth order displacement and momentum thicknesses increase with decrease in the value of surface curvature. The reduction in curvature results in the reduction in the gradients of first order velocity and microrotation at the wall as well as first order displacement and momentum thicknesses.  相似文献
