Face centered cubic (Al0.32Cr0.68)2O3 thin films have been annealed in the temperature range of 500–1000 °C during 2–8 h. The fcc structure of the film remains intact when annealed at temperatures up to 700 °C for 8 h. X-ray diffraction and transmission electron microscopy show the onset of phase transformation to corundum phase alloys in the sample annealed at 900 °C for 2 h, where annealing at 1000 °C for 2 h results in complete phase transformation to α-(Al0.32Cr0.68)2O3. In-plane and out-of-plane line scans performed in EDX TEM and θ/2θ XRD patterns did not show any phase separation into α-Cr2O3 and Al2O3 prior and after the annealing. The apparent activation energy of this process is 380–480 kJ/mol as determined by the Johnson–Mehl–Avrami model. 相似文献

Californium-252 plasma desorption mass spectrometry (PDMS) hasbeen employed for the characterization of a series of humaninsulin derivatives in order to evaluate the performance ofthis technique as an analytical tool in protein engineering.Several of the characterized modifications result in a 1 a.m.u.mass change. The precision in mass determination obtainableby PDMS analysis is not sufficient for unambiguous verificationof such modifications based on the molecular weight alone. Protein unfolding inside immobilized polymerosomes : One of the most interesting properties of polymeric vesicles is their remarkable stability against extreme temperatures and osmotic stress, and their longevity even under harsh environmental conditions. We have demonstrated, in an application on protein folding, that surface‐tethered polymerosomes are suitable for performing time‐resolved single molecule studies with encapsulated proteins, as illustrated here.

A novel technology to mitigate the climate changes and improve energy security is Pressurized Entrained flow High Temperature Black Liquor Gasification (PEHT-BLG) in combination with an efficient fuel synthesis using the resulting syngas. In order to optimise the technology for use in a pulp and paper mill based biorefinery, it is of great importance to understand how the operational parameters of the gasifier affect the product gas composition. The present paper is based on experiments where gas samples were withdrawn from the hot part of a 3 MW entrained flow pressurized black liquor gasifier of semi industrial scale using a high temperature gas sampling system. Specifically, the influence of process conditions on product gas composition (CO2, CO, H2, CH4, H2S, and COS) were examined by systematically varying the operational parameters: system pressure, oxygen to black liquor equivalence ratio, black liquor flow rate to pressure ratio and black liquor pre-heat temperature. Due to the harsh environment inside the gasification reactor, gas sampling is a challenging task. However, for the purpose of the current study, a specially designed high temperature gas sampling system was successfully developed and used. The results, obtained from two separate experimental campaigns, show that all of the investigated operational parameters have a significant influence on the product gas composition and present valuable information about to the process characteristics. 相似文献

The module of elasticity is one of the most important mechanical properties defining the strength of a material which is a prerequisite to design a component from its early stage of conception to its field of application. When a material is to be thermally sprayed, mechanical properties of the deposited layers differ from the bulk material, mainly due to the anisotropy of the highly textured coating microstructure. The mechanical response of the deposited layers significantly influences the overall performance of the coated component. It is, therefore, of importance to evaluate the effective module of elasticity of the coating. Conventional experimental methods such as microindentation, nanoindentation and four-point bending tests have been investigated and their results vary significantly, mainly due to inhomogeneous characteristics of the coating microstructure. Synchrotron radiation coupled with a tensile test rig has been proposed as an alternative method to determine the coating anisotropic elastic behavior dependence on crystallographic orientations. The investigation was performed on Inconel 718 (IN718) HVOF coatings sprayed on IN718 substrates. Combining these experimental techniques yield a deeper understanding of the nature of the HVOF coating Young’s modulus and thus a tool for Design Practice for repair applications. 相似文献

A tunable microwave phase shifter composed of a low-sintering-temperature, screen-printable barium strontium titanate (BST) film and silver metallization was fabricated on an alumina substrate and co-fired at 900 °C for 3 h. The dielectric properties of the films were characterized in a frequency range of 0.8–8 GHz using scattering parameter measurements and a quasi-static coplanar waveguide transmission line model. The temperature dependency of the films was measured through capacitance measurements in a frequency range of 0.5–2.5 GHz. The figure of merit (phase shift/dB of insertion loss) of the phase shifter was found to be 14.6 at 3 GHz with an applied bias field strength of 2.5 V/μm. The performance of the phase shifter is briefly discussed and compared with other phase shifters fabricated by direct screen-printing of BST films. 相似文献

A model was developed to simulate drainage of compressible particle suspensions, and study how cake compression and volumetric load influence the process. The input parameters were settling velocity, cake resistance and compressibility. These parameters were found using a new experimental method. Dextran‐MnO2 particle suspensions were drained as these resemble organic waste slurries with respect to settling and compressibility. It was demonstrated that cake compressibility must be taken into account to obtain adequate simulations. This implies that pressurized filtration resistances cannot be used for drainage simulations. In the filtration step, a distinct increase of dry matter from top to bottom of the cake was observed. During the subsequent consolidation, the cake compressed and a uniform dry matter profile was found. The final dry matter content of the cake increased with feed concentration and volumetric load. Quantitative measurement of intramolecular and intermolecular interactions in protein structure is an elusive task, not easy to address experimentally. The phenomenon denoted ‘energetic coupling’ describes short- and long-range interactions between two residues in a protein system. A powerful method to identify and quantitatively characterize long-range interactions and allosteric networks in proteins or protein–ligand complexes is called double-mutant cycles analysis. In this review we describe the thermodynamic principles and basic equations that underlie the double mutant cycle methodology, its fields of application and latest employments, and caveats and pitfalls that the experimentalists must consider. In particular, we show how double mutant cycles can be a powerful tool to investigate allosteric mechanisms in protein binding reactions as well as elusive states in protein folding pathways. 相似文献

The lysosomal storage disease Niemann–Pick type C (NPC) is caused by impaired cholesterol efflux from lysosomes, which is accompanied by secondary lysosomal accumulation of sphingomyelin and glucosylceramide (GlcCer). Similar to Gaucher disease (GD), patients deficient in glucocerebrosidase (GCase) degrading GlcCer, NPC patients show an elevated glucosylsphingosine and glucosylated cholesterol. In livers of mice lacking the lysosomal cholesterol efflux transporter NPC1, we investigated the expression of established biomarkers of lipid-laden macrophages of GD patients, their GCase status, and content on the cytosol facing glucosylceramidase GBA2 and lysosomal integral membrane protein type B (LIMP2), a transporter of newly formed GCase to lysosomes. Livers of 80-week-old Npc1−/− mice showed a partially reduced GCase protein and enzymatic activity. In contrast, GBA2 levels tended to be reciprocally increased with the GCase deficiency. In Npc1−/− liver, increased expression of lysosomal enzymes (cathepsin D, acid ceramidase) was observed as well as increased markers of lipid-stressed macrophages (GPNMB and galectin-3). Immunohistochemistry showed that the latter markers are expressed by lipid laden Kupffer cells. Earlier reported increase of LIMP2 in Npc1−/− liver was confirmed. Unexpectedly, immunohistochemistry showed that LIMP2 is particularly overexpressed in the hepatocytes of the Npc1−/− liver. LIMP2 in these hepatocytes seems not to only localize to (endo)lysosomes. The recent recognition that LIMP2 harbors a cholesterol channel prompts the speculation that LIMP2 in Npc1−/− hepatocytes might mediate export of cholesterol into the bile and thus protects the hepatocytes. 相似文献

Radionuclide imaging of HER2 expression in tumours may enable stratification of patients with breast, ovarian, and gastroesophageal cancers for HER2-targeting therapies. A first-generation HER2-binding affibody molecule [99mTc]Tc-ZHER2:V2 demonstrated favorable imaging properties in preclinical studies. Thereafter, the affibody scaffold has been extensively modified, which increased its melting point, improved storage stability, and increased hydrophilicity of the surface. In this study, a second-generation affibody molecule (designated ZHER2:41071) with a new improved scaffold has been prepared and characterized. HER2-binding, biodistribution, and tumour-targeting properties of [99mTc]Tc-labelled ZHER2:41071 were investigated. These properties were compared with properties of the first-generation affibody molecules, [99mTc]Tc-ZHER2:V2 and [99mTc]Tc-ZHER2:2395. [99mTc]Tc-ZHER2:41071 bound specifically to HER2 expressing cells with an affinity of 58 ± 2 pM. The renal uptake for [99mTc]Tc-ZHER2:41071 and [99mTc]Tc-ZHER2:V2 was 25–30 fold lower when compared with [99mTc]Tc-ZHER2:2395. The uptake in tumour and kidney for [99mTc]Tc-ZHER2:41071 and [99mTc]Tc-ZHER2:V2 in SKOV-3 xenografts was similar. In conclusion, an extensive re-engineering of the scaffold did not compromise imaging properties of the affibody molecule labelled with 99mTc using a GGGC chelator. The new probe, [99mTc]Tc-ZHER2:41071 provided the best tumour-to-blood ratio compared to HER2-imaging probes for single photon emission computed tomography (SPECT) described in the literature so far. [99mTc]Tc-ZHER2:41071 is a promising candidate for further clinical translation studies. 相似文献
