Real‐time tracking of the dynamics change of self‐assembled nanostructures in physiological environments is crucial to improving their delivery efficiency and therapeutic effects. However, such tracking is impeded by the complex biological microenvironment leading to inhomogeneous distribution. A rotatable fluorescent ratio strategy is introduced that integrates aggregation‐induced emission (AIE) and aggregation‐caused quenching (ACQ) into one nanostructured system, termed AIE and ACQ fluorescence ratio (AAR). Following this strategy, an advanced probe, PEG5k‐TPE4‐ICGD4 (PTI), is developed to track the dynamics change. The extremely sharp fluorescent changes (up to 4008‐fold) in AAR allowed for the clear distinguishing and localization of the intact state and diverse dissociated states. The spatiotemporal distribution and structural dynamics of the PTI micelles can be tracked, quantitatively analyzed in living cells and animal tissue by the real‐time ratio map, and be used to monitor other responsive nanoplatforms. With this method, the dynamics of nanoparticle in different organelles are able to be investigated and validated by transmission electron microscopy. This novel strategy is generally applicable to many self‐assembled nanostructures for understanding delivery mechanism in living systems, ultimately to enhance their performance in biomedical applications.  相似文献

Dopant‐free hole transport materials (HTMs) are essential for commercialization of perovskite solar cells (PSCs). However, power conversion efficiencies (PCEs) of the state‐of‐the‐art PSCs with small molecule dopant‐free HTMs are below 20%. Herein, a simple dithieno[3,2‐b:2′,3′‐d]pyrrol‐cored small molecule, DTP‐C6Th, is reported as a promising dopant‐free HTM. Compared with commonly used spiro‐OMeTAD, DTP‐C6Th exhibits a similar energy level, a better hole mobility of 4.18 × 10?4 cm2 V?1 s?1, and more efficient hole extraction, enabling efficient and stable PSCs with a dopant‐free HTM. With the addition of an ultrathin poly(methyl methacrylate) passivation layer and properly tuning the composition of the perovskite absorber layer, a champion PCE of 21.04% is achieved, which is the highest value for small molecule dopant‐free HTM based PSCs to date. Additionally, PSCs using the DTP‐C6Th HTM exhibit significantly improved long‐term stability compared with the conventional cells with the metal additive doped spiro‐OMeTAD HTM. Therefore, this work provides a new candidate and effective device engineering strategy for achieving high PCEs with dopant‐free HTMs.  相似文献

A specific and reversible method is reported to engineer cell‐membrane function by embedding DNA‐origami nanodevices onto the cell surface. Robust membrane functionalization across epithelial, mesenchymal, and nonadherent immune cells is achieved with DNA nanoplatforms that enable functions including the construction of higher‐order DNA assemblies at the cell surface and programed cell–cell adhesion between homotypic and heterotypic cells via sequence‐specific DNA hybridization. It is anticipated that integration of DNA‐origami nanodevices can transform the cell membrane into an engineered material that can mimic, manipulate, and measure biophysical and biochemical function within the plasma membrane of living cells.  相似文献

The β subunit of adenosine monophosphate (AMP)‐activated protein kinase (AMPK), which exists as two isoforms (β1 and β2) in humans, has a carbohydrate‐binding module (CBM) that interacts with glycogen. Although the β1‐ and β2‐CBMs are structurally similar, with strictly conserved ligand‐contact residues, they show different carbohydrate affinities. β2‐CBM shows the strongest affinity for both branched and unbranched oligosaccharides and it has recently been shown that a Thr insertion into β2‐CBM (Thr101) forms a pocket to accommodate branches. This insertion does not explain why β2‐CBM binds all carbohydrates with stronger affinity. Herein, it is shown that residue 134 (Val for β2 and Thr for β1), which does not come into contact with a carbohydrate, appears to account for the affinity difference. Characterisation by NMR spectroscopy, however, suggests that mutant β2‐Thr101Δ/Val134Thr differs from that of β1‐CBM, and mutant β1‐Thr101ins/Thr134Val differs from that of β2‐CBM. Furthermore, these mutants are less stable to chemical denaturation, relative to that of wild‐type β‐CBMs, which confounds the affinity analyses. To support the importance of Thr101 and Val134, the ancestral CBM has been constructed. This CBM retains Thr101 and Val134, which suggests that the extant β1‐CBM has a modest loss of function in carbohydrate binding. Because the ancestor bound carbohydrate with equal affinity to that of β2‐CBM, it is concluded that residue 134 plays an indirect role in carbohydrate binding.  相似文献

Camelina sativa seed is an underutilized oil source that attracts a growing interest, but it requires more research on its composition and processing. Its high omega‐3 content and growing demand for clean food processing technologies make conventional oil extraction less attractive. In this study, the effect of extraction methods on the bioactive lipid composition of the camelina seed lipid was investigated, and its bioactive lipid composition was modified at the extraction stage using ethanol‐modified supercritical carbon dioxide (SC‐CO2). Ethanol‐modified SC‐CO2 extractions were carried out at varying temperatures (50 and 70 °C), pressures (35 and 45 MPa), and ethanol concentrations (0–10%, w/w), and were compared to SC‐CO2, cold press, and hexane extraction. The highest total lipid yield (37.6%) was at 45 MPa/70 °C/10% (w/w) ethanol. Phospholipids and phenolic content increased significantly with ethanol‐modified SC‐CO2 (p < 0.05). SC‐CO2 with 10% (w/w) ethanol concentration selectively increased phosphatidylcholine (PC) content. Apparent solubility of camelina seed lipids in SC‐CO2, determined using the Chrastil model, ranged from 0.0065 kg oil/kg CO2 (35 MPa/50 °C) to 0.0133 kg oil/kg CO2 (45 MPa/70 °C). Ethanol‐modified SC‐CO2 extraction allowed modification of the lipid composition that was not possible with the conventional extraction methods. This is a promising green method for extraction and fractionation of camelina seed lipids to separate and enrich its bioactives.  相似文献

In order to investigate the putative association between chemical contamination in western Lake Ontario and high prevalences of fish tumors, sediments from Hamilton Harbour and Oakville Creek in Lake Ontario and reference sites in non-polluted areas of Ontario, Canada were collected and extracted for organic contaminants. Sediment extracts from Hamilton Harbour had the highest concentrations of polychlorinated biphenyls and organochlorine insecticides (ppb) and contained very high concentrations of polynuclear aromatic hydrocarbons (ppm); although the levels of these compounds varied widely with sampling location in the harbor. A sediment extract from Hamilton Harbour was mutagenic in the Ames bacterial assay, both with and without microsomal activation. High levels of aromatic DNA adducts were induced in cultured mouse C3H1OT1/2 cells after in vitro exposure to Hamilton Harbour sediment extract. In two separate carcinogenicity experiments involving a sac fry microinjection assay with rainbow trout (Oncorhynchus mykiss), Hamilton Harbour sediment extract induced hepatocellular carcinomas in fish. No hepatic neoplasms were observed in fish that had been treated with sediment extract from Oakville Creek, or with extract from a reference sediment. The significance of these results is discussed in relation to the distribution of neoplasms in feral fish within western Lake Ontario.  相似文献

In this paper, testing of radio frequency (RF) devices with mixed-signal testers is discussed. General purpose automatic test equipment (ATE) will be used. In this paper, a more universal test structure utilizing RF building blocks is proposed. A global positioning system (GPS) device is used as an example to illustrate how to develop the RF test plan with this usage. The test plan developed includes fast, cost-effective and dedicated circuitry.

Polycyclic aromatic hydrocarbons (PAHs) can partition from the atmosphere into agricultural crops, contributing to exposure through the dietary pathway. In this study, controlled environmental chamber experiments were conducted to investigate the transfer of PAHs from air into wheat grain, which is a major food staple. A series of PAHs ranging in size from naphthalene to pyrene were maintained at elevated gas-phase concentrations in the chamber housing mature and dry wheat grain both on the plant and with the husk removed. The PAHs did not achieve equilibrium between the air and grain over the 6.5 month monitoring period used in this study. Therefore, PAH uptake under field conditions is expected to be kinetically limited. A clearance study conducted for the grain showed the half-life of clearance was approximately 20 days for all compounds studied. The results suggest that atmospheric contaminants that partition into grain may remain in the grain long enough to contribute to dietary exposure for humans. Mass transfer across the air/grain interface appeared to be limited by grain-side resistance. The grain may act as a multicompartment system with rapid exchange at the surface followed by slower transfer into the grain. A grain/air concentration relationship was derived for the uptake time that is relevant to field conditions.  相似文献

Human mastocytosis is characterized by increased mast cells. It usually occurs as a sporadic disease that is often transient and limited in children and persistent or progressive in adults. The c-KIT protooncogene encodes KIT, a tyrosine kinase that is the receptor for mast cell growth factor. Because mutated KIT can transform cells, we examined c-KIT in skin lesions of 22 patients with sporadic mastocytosis and 3 patients with familial mastocytosis. All patients with adult sporadic mastocytosis had somatic c-KIT mutations in codon 816 causing substitution of valine for aspartate and spontaneous activation of mast cell growth factor receptor (P = 0.0001). A subset of four pediatric onset cases with clinically unusual disease also had codon 816 activating mutations substituting valine, tyrosine, or phenylalanine for aspartate. Typical pediatric patients lacked 816 mutations, but limited sequencing showed three of six had a novel dominant inactivating mutation substituting lysine for glutamic acid in position 839, the site of a potential salt bridge that is highly conserved in receptor tyrosine kinases. No c-KIT mutations were found in the entire coding region of three patients with familial mastocytosis. We conclude that c-KIT somatic mutations substituting valine in position 816 of KIT are characteristic of sporadic adult mastocytosis and may cause this disease. Similar mutations causing activation of the mast cell growth factor receptor are found in children apparently at risk for extensive or persistent disease. In contrast, typical pediatric mastocytosis patients lack these mutations and may express inactivating c-KIT mutations. Familial mastocytosis, however, may occur in the absence of c-KIT coding mutations.  相似文献
