Examined participation in health behaviors, health locus of control, and negotiation of developmental tasks of adulthood with 75 patients with cystic fibrosis (CF), aged 18–42 yrs old. Ss completed the Multidimensional Health Locus of Control Scale. Results indicate there were a number of health behaviors for which Ss had not yet assumed responsibility, such as managing their medical insurance correspondence and monitoring nutrition. Ss reported that they were more compliant with their medical regimen currently than when first assuming responsibility for their health as adolescents. Multidimensional health locus of control profiles were similar to those of other chronic illness populations. Most Ss appeared to have successfully mastered adult developmental tasks, such as living independently, getting married, and attaining educational or vocational goals. A biobased polymer derived from cashew nut shell liquid (CNSL) as a renewable resource was investigated for use as an antibacterial material. CNSL is a mixture of aromatics containing cardanol as the main component and cardol and 2‐methylcardol as minor components. CNSL composition analyses showed that the minor components (i.e., cardol and 2‐methylcardol) in CNSL had higher contents of unsaturated structures than cardanol. These higher unsaturated contents promoted the thermal polymerization in the preparation of an epoxy CNSL prepolymer (ECNP). The biobased polymer film was fabricated by the reaction of amine compounds and ECNP without any organic solvent. The ECNP film took less than 2.0 h to reach a hardened dry condition at room temperature because of the crosslinking reaction between epoxy and amine groups. The antibacterial activities of the biobased polymer against Escherichia coli and Staphylococcus aureus were evaluated. CNSL showed antibacterial activity against S. aureus, whereas epoxy CNSL and ECNP alone showed no significant antibacterial activity against E. coli or S. aureus. This indicated that the antibacterial activity was based on the phenolic and catechol hydroxyl groups of CNSL. In addition, a biobased polymer film derived from CNSL and diamine showed antibacterial activity against both E. coli and S. aureus, even with alcohol conditioning. This suggested that the antibacterial activity was certainly fixed in the structure of the ECNP‐based polymers after the standard antisepsis treatment in medical facilities. After viewing 2 sexually explicit films, 52 sexually functional participants were given bogus feedback indicating a low erectile response. The men were given either an external, fluctuating attribution (i.e., poor films) or an internal, stable attribution (i.e., problematic thoughts about sex) for the low arousal. As hypothesized, participants in the external, fluctuating group evidenced greater erectile response and subjective arousal during a 3rd film than did participants given the internal, stable attribution. This may indicate that after an occasion of erectile difficulty, the cause to which the difficulty is attributed plays an important role in future sexual functioning. The effects of bone morphogenetic proteins (BMPs), members of the transforming growth factor-β (TGF-β) family, in endometrial cancer (EC) have yet to be determined. In this study, we analyzed the TCGA and MSK-IMPACT datasets and investigated the effects of BMP2 and of TWSG1, a BMP antagonist, on Ishikawa EC cells. Frequent ACVR1 mutations and high mRNA expressions of BMP ligands and receptors were observed in EC patients of the TCGA and MSK-IMPACT datasets. Ishikawa cells secreted higher amounts of BMP2 compared with ovarian cancer cell lines. Exogenous BMP2 stimulation enhanced EC cell sphere formation via c-KIT induction. BMP2 also induced EMT of EC cells, and promoted migration by induction of SLUG. The BMP receptor kinase inhibitor LDN193189 augmented the growth inhibitory effects of carboplatin. Analyses of mRNAs of several BMP antagonists revealed that TWSG1 mRNA was abundantly expressed in Ishikawa cells. TWSG1 suppressed BMP7-induced, but not BMP2-induced, EC cell sphere formation and migration. Our results suggest that BMP signaling promotes EC tumorigenesis, and that TWSG1 antagonizes BMP7 in EC. BMP signaling inhibitors, in combination with chemotherapy, might be useful in the treatment of EC patients.  相似文献

Perlecan (HSPG2), a basement membrane-type heparan sulfate proteoglycan, has been implicated in the development of aortic tissue. However, its role in the development and maintenance of the aortic wall remains unknown. Perlecan-deficient mice (Hspg2−/−-Tg: Perl KO) have been found to show a high frequency (15–35%) of aortic dissection (AD). Herein, an analysis of the aortic wall of Perl KO mice revealed that perlecan deficiency caused thinner and partially torn elastic lamina. Compared to the control aortic tissue, perlecan-deficient aortic tissue showed a significant decrease in desmosine content and an increase in soluble tropoelastin levels, implying the presence of immature elastic fibers in Perl KO mice. Furthermore, the reduced expression of the smooth muscle cell contractile proteins actin and myosin in perlecan-deficient aortic tissue may explain the risk of AD. This study showed that a deficiency in perlecan, which is localized along the elastic lamina and at the interface between elastin and fibrillin-1, increased the risk of AD, largely due to the immaturity of extracellular matrix in the aortic tissue. Overall, we proposed a new model of AD that considers the deficiency of extracellular molecule perlecan as a risk factor.  相似文献

Asymmetric anionic polymerizations of a prochiral monomer, 7-cyano-7-phenyl-1,4-benzoquinone methide (3), using chiral initiators were performed under various conditions, and optically active polymers having configurational chirality in the main chain were obtained though their specific rotation values are quite small. The optically active polymer with a negative specific rotation value of −4.4° was obtained by polymerization of 3 with a lithium isopropylphenoxide (iPrPhOLi)/(−)-sparteine ((−)-Sp) in a mixture solution of dichloromethane/toluene ratio of 30/70 (in vol%) at −40 °C. Stereostructures of 1-mer, 2-mer, and oligomers obtained by asymmetric anionic oligomerization of 3 with iPrPhOLi/(−)-Sp were examined in detail. It was found that the stereoselectivity turned out to be the opposite between the initiation reaction and the propagation one.  相似文献

Claudin-2 (CLDN2), a tight junctional protein, is involved in the chemoresistance in a three-dimensional spheroid culture model of human lung adenocarcinoma A549 cells. However, the mechanism has not been fully clarified. We found that the knockdown of CLDN2 expression by siRNA in the spheroid reduces the expression of glucose transporters and metabolic enzymes. In a two-dimensional culture model, the expression of these proteins was increased by glucose deprivation or fasentin, an inhibitor of glucose transporter. In addition, the expression levels of nuclear factor erythroid 2-related factor 2 (Nrf2) and antioxidant enzymes including heme oxygenase-1, NAD(P)H:quinone oxidoreductase-1, and a glutamate–cysteine ligase modifier subunit were increased by fasentin. The fluorescence intensities of JC-1, a probe of mitochondrial membrane potential, and MitoROS 580, a probe of mitochondrial superoxide production, were increased by fasentin. These results suggest that mitochondrial production of reactive oxygen species is increased by glucose deficiency. The knockdown of CLDN2 enhanced the flux of 2-deoxy-2-[(7-nitro-2,1,3-benzoxadiazol-4-yl)amino]-D-glucose (2-NBDG), a fluorescent deoxyglucose derivative, in a transwell assay, and the accumulation of glucose and 2-NBDG in spheroid cells. The expression of Nrf2 was decreased by CLDN2 knockdown, which was inhibited by fasentin and sulforaphane, a typical Nrf2 activator, in spheroid cells. The sensitivity of spheroid cells to doxorubicin, an anthracycline antitumor antibiotic, was enhanced by CLDN2 knockdown, which was inhibited by fasentin and sulforaphane. We suggest that CLDN2 induces chemoresistance in spheroid cells mediated through the inhibition of glucose transport and activation of the Nrf2 signal.  相似文献

The objectives of this study were to describe the operating conditions of dry chemistry instruments in primary care, as well as to elucidate financial aspects in general practice fee-for-service settings. We used questionnaires mailed to all users of the two most used dry chemistry instruments in Norway, as well as to a 14% random sample of Norwegian GPs. The overall response rate was 79%. The mean number of dry chemistry analyses varied considerably between individual users, but in general a substantial number of analyses were carried out. Even though most analyses on the instruments' repertoire were available in all user groups, a total of 13 additional constituents were suggested to be included in the repertoire. In occupational health care most results were ready when the client was present; this was not the case in general practice. The instruments were more profitable when more constituents were analysed per sample, although profitability varied substantially in the period studied (1986-1989). A discrete time history event analysis revealed that net profit earned, lower instrument price, available information about the technology and being in solo practice significantly influenced the decision to buy an instrument in fee-for-service practices.  相似文献
