A spectral element method for the approximate solution of linear elastodynamic equations, set in a weak form, is shown to provide an efficient tool for simulating elastic wave propagation in realistic geological structures in two‐ and three‐dimensional geometries. The computational domain is discretized into quadrangles, or hexahedra, defined with respect to a reference unit domain by an invertible local mapping. Inside each reference element, the numerical integration is based on the tensor‐product of a Gauss–Lobatto–Legendre 1‐D quadrature and the solution is expanded onto a discrete polynomial basis using Lagrange interpolants. As a result, the mass matrix is always diagonal, which drastically reduces the computational cost and allows an efficient parallel implementation. Absorbing boundary conditions are introduced in variational form to simulate unbounded physical domains. The time discretization is based on an energy‐momentum conserving scheme that can be put into a classical explicit‐implicit predictor/multicorrector format. Long term energy conservation and stability properties are illustrated as well as the efficiency of the absorbing conditions. The accuracy of the method is shown by comparing the spectral element results to numerical solutions of some classical two‐dimensional problems obtained by other methods. The potentiality of the method is then illustrated by studying a simple three‐dimensional model. Very accurate modelling of Rayleigh wave propagation and surface diffraction is obtained at a low computational cost. The method is shown to provide an efficient tool to study the diffraction of elastic waves and the large amplification of ground motion caused by three‐dimensional surface topographies. Fragile X syndrome (FXS), the most common form of inherited intellectual disability, is caused by a developmentally regulated silencing of the FMR1 gene, but its effect on human neuronal network development and function is not fully understood. Here, we isolated isogenic human embryonic stem cell (hESC) subclones—one with a full FX mutation and one that is free of the mutation (control) but shares the same genetic background—differentiated them into induced neurons (iNs) by forced expression of NEUROG-1, and compared the functional properties of the derived neuronal networks. High-throughput image analysis demonstrates that FX-iNs have significantly smaller cell bodies and reduced arborizations than the control. Both FX- and control-neurons can discharge repetitive action potentials, and FX neuronal networks are also able to generate spontaneous excitatory synaptic currents with slight differences from the control, demonstrating that iNs generate more mature neuronal networks than the previously used protocols. MEA analysis demonstrated that FX networks are hyperexcitable with significantly higher spontaneous burst-firing activity compared to the control. Most importantly, cross-correlation analysis enabled quantification of network connectivity to demonstrate that the FX neuronal networks are significantly less synchronous than the control, which can explain the origin of the development of intellectual dysfunction associated with FXS. 相似文献

Polyurethane foam (PUF) is known to sorb gas-phase semivolatile organic compounds (SVOCs) from ambient air and is used routinely in conventional high volume filter-sorbent sampling of such pollutants. PUF rings have also been employed as impaction substrates in a high volume cascade impactor (HVCI) used as a sampler for the evaluation of particle toxicity. Though nonvolatile particles (e.g., trace metals, inorganic ions) have been the primary focus, the sampler has also been used to measure particulate SVOC concentrations in ambient air. The aim of this work is to investigate the validity of the latter approach. The results of three sets of experiments conducted in Canada and Denmark are reported herein. Model compounds included native and deuterated polycyclic aromatic hydrocarbons (PAHs). The experiments demonstrated that HVCI PUF substrates sorb gas-phase PAH compounds and that the sorbed mass is subject to mobilization through and out of the sampler. Particulate concentrations of low molecular weight and volatile PAHs are therefore prone to overestimation in samples that have been analyzed after extraction of whole PUF substrates. Sonication of collected particles in water before solvent extraction is effective at dislodging them from the PUF but also acts to redistribute their originally particulate PAH mass back to the PUF and to the sonication water. As a result, the PAH content of particles measured after sonication and subsequent filtration does not accurately represent their true values. These artefacts affect not only measured PAH concentrations but also the results of toxicological assays that are conducted to test the characteristics of particles collected using HVCI PUF samplers. 相似文献

Health risk of coplanar polychlorinated biphenyl congeners in edible fish from the Mediterranean Sea

The muscle tissue of different species of fish was analyzed for polychlorinated biphenyls (PCBs) in order to estimate the daily intake of these contaminants. Contamination levels among the different species varied between 108 and 678 ng/g lipid weight. Isomer-specific analysis in the muscle tissue of the various fish revealed a profile dominated by hexa- and pentachlorobiphenyls, followed by hepta- and tetrachlorobiphenyls. Other congeners, including those with fewer than four or more than nine chlorine atoms, were below the instrumental limit of detection in all samples. PCB dietary intake was below the range of 1 to 4 pg 2,3,7,8-tetrachloro-p-dioxin equivalents per kg body weight per day set by the World Health Organization. In this paper, we are concerned with a generalized Gauss-Seidel approach to sparse linear least-squares problems. Two algorithms, related to those given by Schechter (1959), for the solution of linear systems are presented and their parallel implementation is discussed. In these procedures, which can be viewed as an alternative ordering of the variables in the SOR methods, the variables are divided into nondisjoint groups. We suggest a local hybrid approximation scheme based on polynomials and radial basis functions, and use it to improve the scattered data fitting algorithm of (Davydov, O., Zeilfelder, F., 2004. Scattered data fitting by direct extension of local polynomials to bivariate splines. Adv. Comp. Math. 21, 223–271). Similar to that algorithm, the new method has linear computational complexity and is therefore suitable for large real world data. Mammalian semen chromatin comprises DNA, protamine, and, at lower levels, other proteins. This constitution confers intense compaction to the chromatin, helping to protect the DNA and causing the head of the sperm to be very small, facilitating the safe transport of its genetic contents. It is known that changes in the sperm chromatin compaction lead to fertility problems in bulls, justifying studies of this structure. Although there are theoretical models of sperm chromatin because of its high compaction, there is no morphological evidence of such models. The aim of this study was to demonstrate the ultrastructure of bovine sperm chromatin in an attempt to corroborate the theoretical chromatin models existing today. The isolated bull sperm heads had their chromatin partially unpacked by chemical treatment using sodium dodecyl sulfate (SDS) and dithiothreitol (DTT) and were then embedded in Epon resin. Using an ultramicrotome, ultrathin sections were obtained, which were contrasted with uranyl acetate and lead citrate, and then viewed under transmission electron microscopy. The methodology used allowed the visualization of toroidal structures interconnected by a filamentous nuclear matrix, which is entirely consistent with the most current theoretical models. Microsc.
