Mammalian semen chromatin comprises DNA, protamine, and, at lower levels, other proteins. This constitution confers intense compaction to the chromatin, helping to protect the DNA and causing the head of the sperm to be very small, facilitating the safe transport of its genetic contents. It is known that changes in the sperm chromatin compaction lead to fertility problems in bulls, justifying studies of this structure. Although there are theoretical models of sperm chromatin because of its high compaction, there is no morphological evidence of such models. The aim of this study was to demonstrate the ultrastructure of bovine sperm chromatin in an attempt to corroborate the theoretical chromatin models existing today. The isolated bull sperm heads had their chromatin partially unpacked by chemical treatment using sodium dodecyl sulfate (SDS) and dithiothreitol (DTT) and were then embedded in Epon resin. Using an ultramicrotome, ultrathin sections were obtained, which were contrasted with uranyl acetate and lead citrate, and then viewed under transmission electron microscopy. The methodology used allowed the visualization of toroidal structures interconnected by a filamentous nuclear matrix, which is entirely consistent with the most current theoretical models. Microsc. Epidemiological studies routinely use central-site particulate matter (PM) as a surrogate for exposure to PM of ambient (outdoor) origin. Below we quantify exposure errors that arise from variations in particle infiltration to aid evaluation of the use of this surrogate, rather than actual exposure, in PM epidemiology. Measurements from 114 homes in three cities from the Relationship of Indoor, Outdoor and Personal Air (RIOPA) study were used. Indoor PM2.5 of outdoor origin was calculated as follows: (1) assuming a constant infiltration factor, as would be the case if central-site PM were a "perfect surrogate" for exposure to outdoor particles; (2) including variations in measured air exchange rates across homes; (3) also incorporating home-to-home variations in particle composition, and (4) calculating sample-specific infiltration factors. The final estimates of PM2.5 of outdoor origin take into account variations in building construction, ventilation practices, and particle properties that result in home-to-home and day-to-day variations in particle infiltration. As assumptions became more realistic (from the first, most constrained model to the fourth, least constrained model), the mean concentration of PM2.5 of outdoor origin increased. Perhaps more importantly, the bandwidth of the distribution increased. These results quantify several ways in which the use of central site PM results in underestimates of the ambient PM2.5 exposure distribution bandwidth. The result is larger uncertainties in relative risk factors for PM2.5 than would occur if epidemiological studies used more accurate exposure measures. A highly efficient ZnO photoanode for dye‐sensitized solar cells was successfully grown by a simple, low cost, and scalable method. A nanostructured coral‐shaped Zn layer was deposited by sputtering onto fluorine‐doped tin oxide/glass slices at room temperature and then thermally oxidized in ambient atmosphere. Stoichiometry, crystalline phase, quality, and morphology of the film were investigated, evidencing the formation of a highly porous branched nanostructure, with a pure wurtzite crystalline structure. ZnO‐based dye‐sensitized solar cells were fabricated with customized microfluidic architecture. Dye loading on the oxide surface was analyzed with ultraviolet‐visible spectroscopy, and the dependence of the cell efficiency on sensitizer incubation time and film thickness was studied by current‐voltage electrical characterization, incident photon‐to‐electron conversion efficiency, and impedance spectroscopy measurements, showing the promising properties of this material for the fabrication of dye‐sensitized solar cell photoanodes with a solar conversion efficiency up to 4.58%. Aluminium nitride thin films were deposited on titanium fixtures, while silicon oxide thin films were deposited on titanium fixtures and chromium-cobalt substrates, using the RF magnetron sputtering technique. Each coating showed different corrosion behaviours when electrochemical characterisations were made in a simulated biological environment, i.e. Hank's solution. Although aluminium nitride was more effective in reducing the localised corrosion on titanium implants, silicon oxide showed the best behaviour against general corrosion and results were consistent for the two kinds of substrates. Moreover the application of a SiO2/AlN bilayer onto the Ti fixtures improved the performances of each separate coating. When the coated surfaces were compared to uncoated substrates, there was no statistically significant difference in cell viability in the response of two different human osteoblastic cell lines — Saos-2 and MG-63. Field emission scanning electron microscopy assessed the surface morphology of coated and uncoated samples, whilst Fourier transform infrared spectroscopy was applied to investigate the bonding structure of barrier layers on silicon samples. On the basis of the data obtained, we are of the opinion that the tested materials show some promising characteristics for the fabrication of novel dental implants with reduced ion release properties. 相似文献

A rural producer who wishes to market organic products must obtain the Brazilian Organic Conformity Assessment System seal and consider a series of criteria for certification. The objective of this article is to identify which criteria should take priority and which organic conformity assessment mechanism (certification by Audit or by the Participatory Guarantee System—SPG) is the most appropriate option for a rural producer who wishes to obtain the seal of organic certification. After a bibliometric analysis, the researchers used the Analytic Hierarchy Process multicriteria method. A sensitivity analysis was also carried out to investigate variations in the priorities of the criteria and certification alternatives. The results indicated the following order of priority of the criteria: cost, market access, and producer availability. SPG was the most relevant assessment mechanism. 相似文献

Accurate estimation of human exposures to volatile organic compounds (VOCs) is a key element of strategies designed to protect public health from the adverse effects of hazardous air pollutants. The focus here is on examining the capability of three different exposure metrics (outdoor community concentrations, indoor residential concentrations, and a simple time-weighted model) to estimate observed personal exposures to 14 VOCs. The analysis is based on 2-day average concentrations of individual VOCs measured concurrently in outdoor (O) air in three urban neighborhoods, indoor (I) air in participant's residences, and personal (P) air near the breathing zone of 71 healthy, nonsmoking adults. A median of four matched P-I-O samples was collected for each study participant in Minneapolis/St. Paul over three seasons (spring, summer, and fall) in 1999 using charcoal-based passive air samplers (3M model 3500 organic vapor monitors). Results show a clear pattern for the 14 VOCs, with P > I > O concentrations. Intra-individual variability typically spanned at least an order of magnitude, and inter-individual variability spanned 2 or more orders of magnitude for each of the 14 VOCs. Although both O and I concentrations generally underestimated personal exposures, I concentrations provided a substantially better estimate of measured P concentrations. Mean squared error (MSE) as well as correlation measures were used to assess estimator performance at the subject-specific level, and hierarchical, mixed effects models were used to estimate the bias and variance components of MSE by tertile of personal exposure. Bias and variance both tended to increase in the upper third of the P exposure distribution for O versus P and I versus P. A simple time-weighted model incorporating measured concentrations in both outdoor community air and indoor residential air provided no improvement over I concentration alone for the estimation of P exposure. 相似文献

We used low-voltage transmission electron holography to probe surface electrostatic potentials in graphene membranes and carbon nanotubes, as the number of graphenes varies. Further, we measured the phase shift induced by an individual graphene, and mapped the phase shift variation throughout a whole few-graphene-crystal as a function of the local number of layers. We found a size/surface effect as the ratio between the surface and the total number of atoms increases for an individual nanotube or graphene membrane. This surface phase term can be related to a fine electrostatic potentials redistributions occurring at the outer layers in carbon nanotubes and graphene membranes. 相似文献

The main goals of our study were to evaluate: 1) the annual risk of tuberculosis infection (ARTI) and its annual decrease in Uganda; 2) the expected incidence of new tuberculosis cases and the notification rate; and 3) the role of incentives given to children tested in increasing compliance with the survey procedures. The methodology is based on performing the standard World Health Organization (WHO) tuberculin test on children of the same age groups at intervals of 10-15 yrs, identifying infected persons by induration distribution analysis, and converting the prevalence rates detected into risk rates according to the ARTI model. Two thousand six hundred and twenty one school children aged 10 yrs old and bacilli Calmette-Guérin (BCG) nonvaccinated, in six study areas, were injected with two tuberculin units (TU) of purified protein derivative (PPD) RT 23 Copenhagen. The detected prevalence was 14 +/- 1.4% (prevalence +/- 95% confidence interval (95% CI)) and the ARTI value 1.2 +/- 0.9%, with an estimated annual decrease of 0.83% from 1958 to 1970 and 2.9% in the 1970-1987 period. The estimated expected incidence of new cases in Uganda was 59 smear positive and 75 smear negative/extrapulmonary cases per 100,000 population in 1987, and 53 and 65, respectively, in 1990, with an overall 68% notification coverage. No significant improvement in children returning for reading was observed in the group receiving incentives. We conclude that the average decrease (2.9%) probably represents the natural decline of tuberculosis in Uganda. The coverage appears encouraging, although the ARTI detected could be underestimated, since the existing ARTI model was developed and validated before the human immunodeficiency virus (HIV) era.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

Participants asked to judge the final position of a moving target typically indicate a position shifted forward. In the 6 experiments reported here, participants were asked to indicate both the starting position (SP) and the vanishing position (VP) at the onset and offset of a moving target. Results confirmed the forward displacement of the VP and showed a backward displacement of the SP. To test whether perceptual estimation was influenced by curvature of the trajectory, curvilinear motions were also used. Results showed that apparent displacements are along the geometrical tangents to the SP and VP. Relationships between the results and other findings such as the flash-lag effect, the representational momentum, and the Fr?lich effect are discussed.
