A method for quantitating protein expression using LC/MS of whole proteins is described. This method is based on the fact that some proteins present in cells are abundant universal proteins whose expression levels exhibit little variation. This method demonstrates that these coextracted proteins can be used as internal standards to which the other proteins in the sample can be compared. By comparing the intensities of a selected protein to marker proteins, or internal standards, a relative ratio is obtained. This ratio can then be used to determine the relative amount of protein expression between cellular extracts. The validity of this approach is described for a standard protein mixture, as well as, E. coli cells that were known to differentially express green fluorescent protein.  相似文献

Although iron-based permeable reactive barriers are gaining importance for treating groundwater contaminants, little is currently known about the effect of cosolutes on barrier longevity. Because of their corrosion inhibiting properties, dissolved silica species are of particular concern. This research investigates the effect of silica on the reduction of 1,1,1-trichloroethane by granular iron as a function of added silica concentration, pH, and duration of iron exposure to dissolved silica. Batch studies reveal that, at pH 8.5 and above, added silica concentrations as low as 0.17 mM cause a 30% reduction in the reactivity of iron. At higher silica concentrations, reactivity decreases by 65-75%. The inhibitory effect is greater at higher pH: 0.83 mM silica has no apparent adverse effect at pH 7.5, but leads to a 46% decrease in reaction rate at pH 8 and 90% at pH 9. This corresponds to observed trends in silica adsorption onto iron, which is low at pH 7.3 but increases at higher pH. Extending the duration of iron exposure to silica solutions also leads to a more pronounced inhibitory effect. This is in good agreement with the increase in silica coverage on the iron surface as revealed by X-ray photoelectron spectroscopy.  相似文献

The efficacy levels of different physical and chemical washing treatments in the reduction of viral and bacterial pathogens from inoculated strawberries were evaluated. Escherichia coli O157:H7, Salmonella Montevideo, poliovirus 1, and the bacteriophages PRD1, phiX174, and MS2 were used as model and surrogate organisms. Chemicals readily available to producers and/or consumers were evaluated as antimicrobial additives for the production of washes. The gentle agitation of contaminated strawberries in water for 2 min led to reductions in microbial populations ranging from 41 to 79% and from 62 to 90% at water temperatures of 22 and 43 degrees C, respectively. Significant reductions (> 98%) in numbers of bacteria and viruses were obtained with sodium hypochlorite (50 to 300 ppm of free chlorine), Oxine or Carnebon (200 ppm of product generating "stabilized chlorine dioxide"), Tsunami (100 ppm of peroxyacetic acid), and Alcide (100 or 200 ppm of acidified sodium chlorite) washes. Overall, 200 ppm of acidified sodium chlorite produced the greatest reductions of microorganisms. Hydrogen peroxide (0.5%) was slightly less effective than free chlorine in a strawberry wash and caused slight fruit discoloration. Cetylpyridinium chloride (0.1%) was effective in the reduction of bacterial species, while trisodium phosphate (1%) was effective against viruses. The consumer-oriented produce wash Fit was very effective (> 99%) in reducing the numbers of bacteria but not in reducing the numbers of viruses. Another wash, Healthy Harvest, was significantly less effective than Fit in reducing bacterial pathogens but more effective for viruses. The performance of automatic dishwashing detergent was similar to that of Healthy Harvest and significantly better than that of liquid dishwashing detergent. Solutions containing table salt (2% NaCl) or vinegar (10%) reduced the numbers of bacteria by about 90%, whereas only the vinegar wash reduced the numbers of viruses significantly (ca. 95%).  相似文献

A respirometry technique can be applied as an effective method to determine the net maximum specific growth rate of autotrophic biomass under both normal conditions and when inhibition occurs. The net maximum specific growth rate of uninhibited autotrophic biomass, expressed as (mu(A)-b(A)), is approximately 0.8 per day [Proceeding of the International Congress on CHISA, Prague, 2002, p. 1]. Several heavy metals and organic compounds have inhibitory effects. Copper (Cu(2+)) has stronger inhibitory effects than zinc (Zn(2+)), and inhibits the nitrification process by 50% at 0.08 mg/l [(mu(A)-b(A)) = 0.4 per day], while the same concentration of Zn(2+) establishes 12% inhibition only [(mu(A)-b(A)) = 0.75 per day]. Inhibition with Cu(2+) starts at concentrations above 0.05 mg/l, while this is above 0.3mg/l for Zn(2+). The inhibition of the nitrification process is complete at 1.2mg/l for both Cu(2+) and Zn(2+). Among the selected organic compounds tested n the experiments, the degree of inhibition decreases as follow: chlorobenzene>trichloroethylene (TCE)>phenol>ethylbenzene. Chlorobenzene already inhibits the autotrophic biomass at 0.25 mg/l. The nitrification process is totally inhibited by adding 0.75 mg/l of chlorobenzene. TCE has a less inhibitory effect on the nitrification process and 50% inhibition is noticed at 0.75 mg/l TCE. The nitrification process is totally inhibited at 1mg/l TCE. Phenol inhibits the nitrification for 50% at 3 mg/l. The inhibitory effect of phenol is almost constant in the range 4-10 mg/l and complete inhibition is reached at 50 mg/l. The inhibitory effect of ethylbenzene is 50% at 8 mg/l and the autotrophic biomass is totally inhibited at 50 mg/l. Experimental findings are compared with literature data, which generally and significantly overestimate the inhibition threshold concentrations.  相似文献

The specific effects of vehicular type on the likelihood of an injury occurring are relatively unexplored. This study sought to assess the relative risk of injury to occupants of four-wheel drive vehicles and their counterparts in passenger cars.Data for 1143 occupants from all of the 454 crashes in Oklahoma, in 1995 that involved a four-wheel drive vehicle were used. Multiple logistic regression analysis determined the association between potential predictive factors and vehicular injury. Odds ratios revealed occupancy in a passenger car to be a major predictor of the likelihood of injury. Other factors include the driver being female, driving too fast, travel on curved or level roadways, and being hit laterally or from the rear.  相似文献

A novel sample pretreatment device is described, and its application to the concentration and purification of crude DNA samples in a flowing stream for subsequent capillary electrophoresis is demonstrated. The device consists of two gap junctions, each covered with a conductive membrane material and built upon a flow channel made of PEEK tubing. Upon the application of an electric field between the junctions, the negatively charged DNA fragments can resist the hydrodynamic flow stream and are trapped between the junctions. DNA fragments dissolved in microliter volumes are captured in a nanoliter-sized band by simply pushing the sample solution through the device. Depending on their electrophoretic mobility, other interfering materials in a crude sample can be removed from the trapped DNA fragments by washing. The selective permeability of the membrane to small ions allows efficient desalting. The concentrated and purified DNA fragments are released by simply turning off or reversing the electric field. Recovery is up to 95%. Performance of the device was evaluated using crude products of fluorescent dye-primer cycle-sequencing reactions. Compared to these crude reaction products, samples purified in the capture device and subsequently collected showed dramatically enhanced signal and resolution when run on a conventional capillary-electrophoresis instrument. Furthermore, the device could be connected in-line to a capillary system for direct injection. The device has great potential for enabling lab-on-a-chip systems to be used with real-world samples.  相似文献

We compare the radiative transfer equation for media with constant refractive index with the radiative transfer equation for media with spatially varying refractive indices [J. Opt. A Pure App. Opt. 1, L1 (1999)] and obtain approximate conditions under which the former equation is accurate for modeling light propagation in scattering media with spatially varying refractive indices. These conditions impose restrictions on the variations of the refractive index, the gradient of the refractive index, the divergence of the rays, the frequency of modulation, and the widths of light pulses, which are related to the mean refractive index, the absorption coefficient, and the reduced scattering coefficient of the medium. Each condition is geometrically interpreted. Some implications of the results are discussed.  相似文献

Ultrasound is used extensively in the medical field for the detection and characterization of a variety of features in the human body. Finite element models used to understand ultrasonic wave propagation in teeth have been developed so that ultrasound techniques could be realized in dentistry. This paper presents a hypothesis that underlies one possible design of an ultrasonic tool that can be used in a clinical environment, as well as several models that describe acoustic field simulation, propagation, and interaction with the layers of several tooth structures. A complete PSpice model of a single-element transducer based on Redwood's version of Mason's equivalent circuit, a focusing lens, and a multi-layer tooth structure is used to illustrate the validity of this hypothesis. Transmission line theory is employed as a basis for the models of the piezoceramic, the lens, and the different tooth layers. Results clearly depict the transmission and reflection of the ultrasonic waves as they travel through the layers within the tooth structure and point out the noticeable similarity to longitudinal L-wave signatures produced by axisymmetric finite element models presented in earlier studies.  相似文献

The goal of this study was to evaluate methods of multidimensional wavelet denoising on restoring the fidelity of biological signals hidden within dynamic positron emission tomography (PET) images. A reduction of noise within pixels, between adjacent regions, and time-serial frames was achieved via redundant multiscale representations. In analyzing dynamic PET data of healthy volunteers, a multiscale method improved the estimate-to-error ratio of flows fivefold without loss of detail. This technique also maintained accuracy of flow estimates in comparison with the "gold standard," using dynamic PET with O15-water. In addition, in studies of coronary disease patients, flow patterns were preserved and infarcted regions were well differentiated from normal regions. The results show that a wavelet-based noise-suppression method produced reliable approximations of salient underlying signals and led to an accurate quantification of myocardial perfusion. The described protocol can be generalized to other temporal biomedical imaging modalities including functional magnetic resonance imaging and ultrasound.  相似文献
