High-performance liquid chromatography (HPLC) methods were developed for the optimised determination of five herbicide residues (dichlorprop, isoproturon, mecoprop, metsulfuron-methyl and 2,4,5-T) and major metabolites. These compounds represent important groups of herbicides and several residues have been found as contaminants in groundwater. The methods make it possible to study these herbicides and several transformation products through simultaneous detection and quantification. Culture media as well as cleaned up extracts from sediment and groundwater can be analysed. Using HPLC with UV detection the general limit of quantification was 1.8 ng injected corresponding to a detection limit of 1-2 micrograms/l when analysing a cleaned up extract from a 20 ml water sample. The method was verified by analysing herbicide residues in groundwater collected from a wetland area. Cleaning up 20 ml groundwater with a residue level of 25 micrograms/l the general recovery was within 58-82%.  相似文献

A cell surface lectin was isolated and purified to homogeneity from the cell walls of a highly flocculent strain of Saccharomyces cerevisiae (NCIM 3528) by chromatography on DEAE-cellulose, phenyl Sepharose and Sephacryl S-300. It showed a molecular mass of 40 kDa on SDS-PAGE. It is an acidic protein with a pI of 4.0 and contains 44% hydrophobic amino acids. The N-terminal sequence up to 10 amino acid residues showed at least 70% homology with the predicted N-terminal sequence of the putative FLO1 as well as FLO5 gene products. The mannose-binding nature of the lectin was indicated by its high affinity and specificity towards the branched trisaccharide of mannose, a ligand which also inhibits the flocculation of yeast cells. Immunofluorescence studies confirmed the presence of lectin on the yeast cell surface and lectin-specific IgGs prevented flocculation of the cells. This cell surface mannose-specific lectin probably plays an important role in flocculation, with the branched trimannoside on the cell wall being the apparent carbohydrate receptor. The N-terminal sequence data gives a primary indication that the lectin could be a product of one of the FLO genes.  相似文献

In video-based particle-image velocimetry (PIV) systems for fluid mechanics research, it is sometimes desirable to image seed particles to be smaller than a camera pixel. However, imaging to this size can lead to marginal image contrast such that significant numbers of erroneous velocity vectors can be computed, even for simple flow fields. A variety of image-enhancement techniques suitable for a low-cost PIV system that uses video cameras are examined and tested on three representative flows. Techniques such as linear contrast enhancement and histogram hyperbolization are shown to have good potential for improving the image contrast and hence the accuracy of the data-reduction process with only a 15% increase in the computational time. Some other schemes that were examined appear to be of little practical value in PIV applications. An automated shifting algorithm based on mass conservation is shown to be useful for displacing the second interrogation region in the direction of flow, which minimizes the number of uncorrelated particle images that contribute noise to the data-reduction process.  相似文献

In this paper we report on the fabrication and characterization of SrHfO(3):Ce ceramics. Powders were prepared by solid-state synthesis using metal oxides and carbonates. X-ray diffraction measurements showed that phase-pure SrHfO(3) is formed at 1200°C. Inductively coupled plasma spectroscopy confirmed the purity and composition of each batch. SrHfO(3) exhibits several phase changes in the solid, but this does not appear to be detrimental to the ceramics. Microprobe experiments showed uniform elemental grain composition, whereas aluminum added as charge compensation for trivalent cerium congregated at grain boundaries and triple points. Radioluminescence spectra revealed that the light yield decreases when the concentration of excess Sr increases. The decrease in the light yield may be related to the change of Ce(3+) into Ce(4+) ions. For stoichiometric SrHfO(3):Ce, the light yield is about four times that of bismuth germanate (BGO), the conventional benchmark, indicating great potential for many scintillator applications.  相似文献

For many important classes of pesticides including phenylurea herbicides (PUHs) and triazines, photosensitized transformation may be the only relevant elimination process in surface waters. In this study, the dissolved organic matter (DOM) mediated phototransformation of PUHs has been investigated in laboratory and field experiments. The results indicate that, in surface waters, the photosensitized transformation of PUHs may be significant and occurs primarily by an initial one-electron oxidation most likely involving excited triplet states of DOM (3DOM*) constituents. Using isoproturon and diuron as model compounds, it is shown that for a given DOM, quantum yield factors determined in the laboratory at a few selected wavelengths can be used to quantify the overall DOM- mediated phototransformation of a given PUH under sunlight irradiation. Furthermore, it is demonstrated that this process can be modeled for a given surface water, by applying the program GCSOLAR and a simple algorithm for cloud cover for quantification of average daily light intensities. Finally, the model has been successfully applied to predict vertical concentration profiles of isoproturon and diuron in a small lake in Switzerland. To our knowledge, this is the first study in which DOM-mediated phototransformation of organic pollutants has been quantitatively validated in the field.  相似文献

Abstract: Knowledge of the relative importance of food quality attributes in determining consumer purchase intention is critical for robust assessment of economic opportunities for industry growth. The aim of this study is to demonstrate how conjoint analysis methodology that incorporates tasting of fruit can be used to collect such information. Three hundred Japanese consumers took part in research designed to measure the importance of dry matter (DM), size, and price of kiwifruit (Actinidia deliciosa “Hayward” and Actinidia chinensis “Hort16A”) for purchase intention. Measurement of consumer liking for kiwifruit of different DM content was a key first step. Liking increased as DM increased and was accompanied by increased purchase likelihood/choice probability for kiwifruit. The size of kiwifruit presented to consumers varied from “small” to “extra large.” Consumers liked “mid‐sized” kiwifruit over “small” or “extra‐large” kiwifruit. Despite these differences in liking, size was of little importance in determining purchase likelihood/choice probability for kiwifruit. Price was a very important factor in determining purchase likelihood/choice probability but was less important than DM content. As price increased, purchase likelihood/choice probability decreased. Beneath these general findings, heterogeneity existed. Some consumers placed more/less importance on the focal purchase drivers than suggested by the aggregate model. Overall, the results suggest that incentive schemes already implemented by industry should consider rewarding high‐DM fruit more than fruit size. Practical Application: This research has contributed to the New Zealand kiwifruit industry gaining a better understanding of the relative importance consumers place on DM, size, and price of kiwifruit and has resulted in changes to grower incentive schemes. The research approach presented forces consumer to tradeoff attributes of kiwifruit against each other and decide on how important two key quality attributes—DM and size—are relative to each other and relative to price. The application of conjoint analysis in this article can be transferred to other fruits, food, and beverages and help guide consumer‐led innovation.  相似文献

Uranium contamination of soils and sediments often originates from acidic or alkaline waste sources, with diffusion being a major transport mechanism. Measurements of U(VI) diffusion from initially pH 2 and pH 11 solutions into a slightly alkaline Altamont soil and a neutral Oak Ridge soil were obtained through monitoring uptake from boundary reservoirs and from U concentration profiles within soil columns. The soils provided pH buffering, resulting in diffusion at nearly constant pH. Micro X-ray absorption near edge structure spectra confirmed that U remained in U(VI) forms in all soils. Time trends of U(VI) depletion from reservoirs and U(VI) concentration profiles within soil columns yielded Kdvalues consistent with those determined in batch tests at similar concentrations (approximately 1 mM) and much lower than values for sorption at much lower concentrations (nM to microM). These results show that U(VI) transport at high concentrations can be relatively fast at non-neutral pH, with negligible surface diffusion, because of weak sorption.  相似文献

Incidence of Salmonella in fish and seafood   总被引:4,自引:0,他引:4  
Field laboratories of the U.S. Food and Drug Administration collected and tested 11,312 import and 768 domestic seafood samples over a 9-year period (1990 to 1998) for the presence of Salmonella. The overall incidence of Salmonella was 7.2% for import and 1.3% for domestic seafood. Nearly 10% of import and 2.8% of domestic raw seafood were positive for Salmonella. The overall incidence of Salmonella in ready-to-eat seafood and shellfish eaten raw was 0.47% for domestic--one shucked oyster and one shark cartilage powder. The incidence in the 2,734 ready-to-eat import seafood was 2.6%--cooked shrimp, shellfish or fish paste, smoked fish, salted/dried fish, and caviar. The incidence in import shellfish consumed raw was 1% in oyster, 3.4% in clams, and 0% in mussels. The incidence in raw, import fish was 12.2%. Distribution of Salmonella in seafood on a regional basis indicated the incidence to be highest in central Pacific and Africa and lowest in Europe/Russia and North America (12% versus 1.6%). Data on a country basis indicated Vietnam to have the highest (30%) and Republic of Korea the lowest (0.7%). While the most frequent serotypes in import seafood were Salmonella Weltevreden (1st), Salmonella Senftenberg (2nd), Salmonella Lexington, and Salmonella Paratyphi-B (3rd, equal numbers for each serotype), the top 20 list included Salmonella enteritidis (5th), Salmonella Newport (6th), Salmonella Thompson (7th), Salmonella typhimurium (12th), and Salmonella anatum (13th), commonly involved in foodborne illness in the United States. Because the incidence in the present study is based on only a small fraction of the seafood imported into the United States, efforts should be directed toward implementation of hazard analysis and critical control points to reduce the incidence of Salmonella in seafood without relying on testing for Salmonella.  相似文献
