Sweet enhancing effect of neohesperidin dihydrochalcone (NHDC) or cyclamate has been reported to be synergistic in human sensory tests. However, little is known about whether these synergisms are caused by the mechanism mediated by the human sweet-taste receptor. Here, we examined the sweetness intensity of sweet tastant mixtures by measuring the responses of cultured cells stably expressing the human sweet-taste receptor. The results showed that the cell response to sucrose was synergistically potentiated by the addition of NHDC or cyclamate. Moreover, a point mutation in the transmembrane domain of hT1R3 almost completely eliminated the enhancing effects of NHDC and cyclamate. These results suggest that ligand–receptor interactions in the transmembrane domain of hT1R3 are necessary for NHDC and cyclamate to elicit the synergistic potentiation of the receptor activation. Our results may provide the foundation of a molecular basis for receptor-based synergisms of sweet tastes in mixtures of diverse sweet substances.  相似文献

We investigated the precipitation processes in Cu-4 mol pct Ti alloy specimens aged at 723 K (450 °C), by means of X-ray diffraction and chemical analyses of the precipitates extracted from the parent alloy specimens. Aging-induced precipitate particles of a spinodally decomposed disorder, α′; those of a metastable order, β′-Cu4Ti; and those of a stable order, β-Cu4Ti, were continuously formed in the aged specimens. The extraction of the precipitate particles from the aged specimens by submergence in a nitric solution allowed for not only the structural analyses of the constituent precipitate phases but also the quantitative evaluation of their chemical compositions and volume fractions. Early during the aging process, the supersaturated Cu solid solution decomposes spinodally in a continuous manner, and an unstable disorder, α′, appears. Then, fine needle-shaped β′-Cu4Ti particles, which have a Ti content of approximately 37.5 mol pct, form in the Cu matrix. During prolonged aging, coarse cellular components composed of the terminal Cu solid solution and stable β-Cu4Ti particles which have a Ti content of 20.5 mol pct nucleate and grow, primarily in the grain boundaries, at the expense of the metastable β′-Cu4Ti particles. The volume fraction of the β′-Cu4Ti particles in the alloy reaches a maximum of approximately 1.7 pct after aging for 24 hours, while that of the β-Cu4Ti particles increases steadily to more than 18 pct after 480 hours. The volume fraction of the fine β′-Cu4Ti particles in the alloy specimens remained constant throughout the age-hardening, indicating that the hardening is primarily owing to the fine dispersion of the β′-Cu4Ti particles and not because of the large volume fraction of coarse β-Cu4Ti particles.  相似文献

In this study, we validated the usefulness of examiners’ forward citations, especially from the viewpoint of the applicants’ self-selection (ASS) decisions during the patent application procedure. We believe that the ASS in an early stage would be decided by a potential-value comparison among patent applications. We focused on six self-selection decision points of the applicants: whether to file patent applications in foreign countries, request for examination, request for accelerated examination, reply to a notification of reasons for refusal, appeal after receiving a decision of refusal, and register after receiving a decision to grant a patent as patent value parameters. We found that application groups that selected “Yes” have a significantly larger number of examiners’ forward citations than groups that selected “No” at all decision points. In addition, we confirmed that applications that were finally granted and those that were renewed for a full term after grant have a significantly large number of examiners’ forward citations. We concluded that the number of examiners’ forward citations would be a useful indicator of the potential value of patent applications in macroscopic analysis.  相似文献

We evaluated the effects of seven mushroom extracts (Grifola frondosa, Pholiota nameko, Panellus serotinus, Hypsizygus marmoreus, Pleurotus cornucopiae, Armillaria mellea, and Flammulina velutipes) on cytotoxic activity and cytokine production of lamina propria leukocytes (LPLs) isolated from rat small (S) and large (L) intestinal mucosa. Boiling water extracts from seven species of mushrooms showed no direct cytotoxicity against the YAC-1 target cells. However, prominent increases of cytotoxicity were observed in S- and L-LPLs co-cultured with P. serotinus extract. Cytokine production (TNFα, IFNγ, IL-12 p70, and IL-4) of S- and L-LPLs was stimulated in response to P. cornucopiae extract. Mushroom extracts contributed to target cell adhesion and/or cytokine production in the effector cells. The promotion of cytotoxic activity in S- and L-LPLs was not necessarily related to β-glucan content of the mushroom.  相似文献

Effects of extrusion ratio (ER) during extruding on mechanical properties of poly(lactic acid) (PLA) screws were investigated in terms of shear and twist strength. Shear strength increased by drawing, while twist strength decreased. This might be due to orientation of molecular chains during drawing. In order to evaluate orientation of molecular chains, orientation function in extruded billets was measured as a function of ER. Since orientation function increased with ER, drawing is effective method to improve shear strength of screw, where fibrous structure which was formed during extruding resisted shear stress in cross-section perpendicular to screw axis. Although orientation functions for billet extruded at ER 1.3–4 were equivalent, both strengths changed for the screw extruded at the range of this ER. Infrared spectra suggested transforming from α to β crystal for the billet extruded at the range of this ER. This result suggested that mechanical properties of screw also depended on the crystal forms.  相似文献
