Two new oligosaccharides were prepared from heparin by its partial depolymerization using heparin lyase I (EC 4.2.2.7) in an attempt to prepare oligosaccharides having intact antithrombin III binding sites. The oligosaccharides were purified by chromatography on the basis of both size and charge and demonstrated a high level of purity by capillary electrophoresis. One- and two-dimensional 1H NMR spectroscopy at 500 MHz revealed the structure of each oligosaccharide. The octasaccharide and decasaccharide are DeltaUAp2S(1-->4)-alpha-DGlcNpS6S(1-->4)-alpha-L-IdoAp (1-->4)-alpha-D -GlcNpAc6S(1-->4)-betaD-GlcAp(1-->4)-alpha-D-GlcNpS 3S6S(1-->4)-alpha- L-IdoAp2S(1-->4)alpha-D-GlcNpS6S (where DeltaUAp is 4-deoxy-alpha-L-threo-hex-enopyranosyluronic acid, GlcNp is 2-amino-2-deoxy-glucopyranose, GlcAp is glucopyranosyluronic acid, S is sulfate and Ac is acetate) and DeltaUAp2S(1-->4)-alpha-D-GlcNpS6S(1-->4)-alpha-L-IdoAp++ +(1-->4)-alpha- D-GlcNpAc6S (1-->4)-beta-D-GlcAp(1-->4)-alpha-D-GlcNpS3S6S(1-->4)-alpha- L-IdoAp2S (1-->4)-alpha-D-GlcNpS6S(1-->4)-alpha-L-IdoAp2S(1-->4)-alpha -D-GlcNpS 6S, respectively. A hexasaccharide containing a similar structural motif to that found in the antithrombin III binding site and having greatly reduced anticoagulant activity was also isolated. The structure of the hexasaccharide is DeltaUAp2S(1-->4)-alpha-D-GlcNpAc6S(1-->4)-beta-D-GlcAp++ +(1-->4)-alpha- D-GlcNpS3S6S(1-->4)-alpha-L-IdoAp(1-->4)-alpha-D-GlcNpS6S . The octasaccharide and decasaccharide correspond to the predominant structural motif found in porcine intestinal mucosal heparin. Sufficient quantities of the decasaccharide were obtained to examine its interaction with antithrombin III using microtitration calorimetry. This decasaccharide bound to antithrombin III with similar avidity as heparin and showed comparable anticoagulant activity, as determined using an antithrombin III dependent anti-factor Xa assay. Interestingly, while both decasaccharide and heparin bound to antithrombin with nanomolar affinity, very little heat of binding was observed.  相似文献

Over a period of 4.5 years, 14 patients with frontoethmoidal meningoencephaloceles were treated. Most patients came from Northern Namibia. Precise delineation of all cranial abnormalities was obtained by modern imaging techniques, and specific patterns of cerebral abnormality were found. The malformation was corrected in a single stage, and significant modifications have been developed to render the procedure simpler and safer. Information from our series favors delayed neural tube closure as the primary pathogenesis of the defect and suggests a common teratogen as the most probable etiological agent. Our experience leads us to advocate early correction of even small defects.  相似文献

We present analytic procedures for calculating the magnetostatic and response functions of magnetoresistive (MR) recording heads. To apply these methods we make several simplifying approximations, which have been found to be valid for typical recording head geometries. In particular, we show how to calculate the bias magnetization distribution, ideal transition pulse shape, and frequency response for the unshielded MR (UMR) head. To do this, we summarize methods based on conformal mapping and reciprocity, and present a simpler direct method which produces identical results. Polarised attenuated total reflection FTIR spectroscopy has been used for detailed orientation mapping of 2l polyethylene terephthalate bottles subjected to different preform heating times. The results for the bottle with the standard preform heating time reveal significant surface orientation variations along the bottle hoop and length directions, with the inner bottle surface also showing consistently greater chain orientation in the hoop direction than found for the outer surface. Decreasing the preform heating time has less effect on the chain orientation than increasing the heating time, with the latter causing reduced chain orientation in the final bottle. The absorbance of the 1340 cm−1 band provides a quick and reliable indication of these chain orientation trends. In contrast, the extent of benzene ring orientation parallel to the bottle surface showed little variation and remained consistently high at the majority of the surface bottle positions probed.  相似文献

The influence of the different types of bonding present in a range of carbon based materials is discussed as a precursor to describing the field emission characteristics of carbon cold cathode materials. Some of the controlling factors which govern electron emission from carbon based cathodes are discussed. It is shown that from disordered carbon films the interplay between the clustered sp2 phase and the insulating sp3 matrix is important. The transition from a ‘back contact’ to ‘front surface’ controlled emission mechanism is described in terms of the sp2 content and field penetration. A possible reason for high field enhancement factors found in disordered films also is provided. It is further shown that changes to the sp2 phase by current stressing can improve the field emission characteristics. Emission from carbon nanotubes is also discussed and the prospects for new types of nanotube – polymer composite based cathodes are also considered.  相似文献

Our previous work has shown that op/op mice hyperabsorb dietary calcium in the vitamin D-deficient state and shunt that calcium into bone. Under these conditions, the op/op mice are hypocalcemic. The purpose of this study was to examine calcium metabolism and bone mineralization in vitamin D-deficient op/op mice. First, the op/op mice and their normal littermates were placed on a vitamin D-deficient, low phosphorus diet to limit bone mineralization. Under these circumstances, op/op mice survived, even when calcium was also removed from the diet. If the diet contained phosphate, op/op mice died from hypocalcemic tetany when calcium was also removed from the diet. Furthermore, serum calcium levels became similar to wild type in the op/op mice administered the vitamin D-deficient, low phosphorus diet, and op/op mice were able to increase serum calcium in response to 1,25-dihydroxyvitamin D3. The op/op mice developed rickets when their serum phosphorus level was too low to support bone mineralization. The op/op mice became hypophosphatemic on regimens in which normal mice were able to maintain normal serum phosphorus levels. It appears that the op/op mouse simply requires a higher dietary calcium and phosphorus level to prevent rickets and hypocalcemic tetany since the bone is not available as a source of these minerals. However, the ability of the op/op mouse to mineralize bone at low serum calcium and phosphorus levels remains unexplained.  相似文献

A clustering algorithm based on the pairwise nearest-neighbor (PNN) algorithm developed by Equitz (1989), is introduced for the design of entropy-constrained residual vector quantizers. The algorithm designs residual vector quantization codebooks by merging the pair of stage clusters that minimizes the increase in overall distortion subject to a given decrease in entropy. Image coding experiments show that the clustering design algorithm typically results in more than a 200:1 reduction in design time relative to the standard iterative entropy-constrained residual vector quantization algorithm while introducing only small additional distortion. Multipath searching over the sequence of merges is also investigated and shown experimentally to slightly improve rate-distortion performance. The proposed algorithm can be used alone or can he followed by the iterative algorithm to improve the reproduction quality at the same bit rate.  相似文献
