Mesoporous silica nanoparticles (MSNs) have been proposed as DNA and drug delivery carriers, as well as efficient tools for fluorescent cell tracking. The major limitation is that MSNs enter cells regardless of a target-specific functionalization. Here we show that non functionalized MSNs, synthesized using a PEG surfactant-based interfacial synthesis procedure, do not enter cells, while a highly specific, receptor mediated, cellular internalization of folic acid (FOL) grafted MSNs (MSN-FOL), occurs exclusively in folate receptor (FR) expressing cells. Neither the classical clathrin pathway nor macropinocytosis is involved in the MSN endocytic process, while fluorescent MSNs (MSN-FITC) enter cells through aspecific, caveolae-mediated, endocytosis. Moreover, internalized particles seem to be mostly exocytosed from cells within 96 h. Finally, cisplatin (Cp) loaded MSN-FOL were tested on cancerous FR-positive (HeLa) or normal FR-negative (HEK293) cells. A strong growth arrest was observed only in HeLa cells treated with MSN-FOL-Cp. The results presented here show that our mesoporous nanoparticles do not enter cells unless opportunely functionalized, suggesting that they could represent a promising vehicle for drug targeting applications. 相似文献

In this work the effect of the ratio of starting reactants on the hydrogen absorption reaction of the system xNaH + MgB2 is investigated. At a constant hydrogen pressure of 50 bar, depending on the amount of NaH present in the system NaH + MgB2, different hydrogen absorption behaviors are observed. For two system compositions: NaH + MgB2 and 0.5NaH + MgB2, the formation of NaBH4 and MgH2 as only crystalline hydrogenation products is achieved. In vitro culture (IVC) of preimplantation mouse embryos is associated with changes in gene expression. It is however, not known if the method of fertilization affects the global pattern of gene expression. We compared gene expression and development of mouse blastocysts produced by in vitro fertilization (IVF) versus blastocysts fertilized in vivo and cultured in vitro from the zygote stage (IVC) versus control blastocysts flushed out of the uterus on post coital day 3.5. The global pattern of gene expression was assessed using the Affymetrix 430 2.0 chip. It appears that each method of fertilization has a unique pattern of gene expression and development. Embryos cultured in vitro had a reduction in the number of trophoblastic cells (IVF 33.5 cells, IVC 39.9 cells, and 49.6 cells in the in vivo group) and, to a lesser degree, of inner cell mass cells (12.8, 11.7, and 13.8 respectively). The inner cell mass nuclei were larger after culture in vitro (140 microm(2), 113 microm(2), and 86 microm(2) respectively). Although a high number of genes (1912) was statistically different in the IVF cohort when compared with the in vivo control embryos, the magnitude of the changes in gene expression were low and only a minority of genes (29 genes) was changed more than fourfold. Surprisingly, IVF embryos were different from IVC embryos (3058 genes were statistically different, but only three changed more than fourfold). Proliferation, apoptosis, and morphogenetic pathways are the most common pathways altered after IVC. Overall, IVF and embryo culture have a profound effect on gene expression pattern and phenotype of mouse preimplantation embryos. 相似文献

The work focuses on the hydrogenation of carbon monoxide over solid catalysts undergoing mechanical activation by ball milling. The rate of carbon monoxide conversion at individual ball impacts was estimated by measuring the impact frequency, the mass of powders involved in individual impacts and their duration. The rate of the mechanochemical hydrogenation process was compared with the one of the corresponding thermal process. In the context of remotely sensed data analysis, an important problem is the development of accurate models for the statistics of the pixel intensities. Focusing on synthetic aperture radar (SAR) data, this modeling process turns out to be a crucial task, for instance, for classification or for denoising purposes. In this paper, an innovative parametric estimation methodology for SAR amplitude data is proposed that adopts a generalized Gaussian (GG) model for the complex SAR backscattered signal. A closed-form expression for the corresponding amplitude probability density function (PDF) is derived and a specific parameter estimation algorithm is developed in order to deal with the proposed model. Specifically, the recently proposed "method-of-log-cumulants" (MoLC) is applied, which stems from the adoption of the Mellin transform (instead of the usual Fourier transform) in the computation of characteristic functions and from the corresponding generalization of the concepts of moment and cumulant. For the developed GG-based amplitude model, the resulting MoLC estimates turn out to be numerically feasible and are also analytically proved to be consistent. The proposed parametric approach was validated by using several real ERS-1, XSAR, E-SAR, and NASA/JPL airborne SAR images, and the experimental results prove that the method models the amplitude PDF better than several previously proposed parametric models for backscattering phenomena. 相似文献
