There are presently two competitive theories that attempt to explain the etiology of multiple sclerosis (MS). Briefly summarized, they are: 1. An infection, probably of viral type, may attack the oligodendroglia of the central nervous system; or, 2. An autoimmune process may begin with an infection of the peripheral lymphatic immune system, producing antibodies that cross the blood-brain barrier, leading to myelinoclasia. Since 1935, research has been directed toward myelin of the central nervous system and the myelin sheaths of peripheral nerve; however, dorsal root and cranial sensory ganglia (DRG) have apparently not been studied. The present hypothesis states that an infectious agent (probably viral) finds privileged sanctuary in the dorsal root and cranial sensory ganglia (DRG): thereafter periodically invading the spinal cord, brain, or peripheral nerve. Previously reported erratic spinal fluid viral titers and cultures can be explained by differences in the anatomy of the DRG in which there is a variable and limited contact of spinal fluid with sensory ganglia. Clues to this hypothesis were noted by the author during routine neurological examinations of patients with MS, in which sensory signs and symptoms were frequently encountered. This clinical observation has also been reported by others who found such symptoms in 75% of MS patients, ranking second only to incoordination. 相似文献

AXL, a member of the TAM family, is a promising therapeutic target due to its elevated expression in advanced hepatocellular carcinoma (HCC), particularly in association with acquired drug resistance. Previously, RNA interference was used to study its role in cancer, and several phenotypic changes, including attenuated cell proliferation and decreased migration and invasion, have been reported. The mechanism of action of AXL in HCC is elusive. We first studied the AXL expression in HCC cell lines by real-time PCR and western blot and showed its stringent association with a mesenchymal phenotype. We then explored the role of AXL in mesenchymal SNU475 cells by CRISPR-Cas9 mediated gene knock-out. AXL-depleted HCC cells displayed drastic phenotypic changes, including increased DNA damage response, prolongation of doubling time, G2 arrest, and polyploidization in vitro and loss of tumorigenicity in vivo. Pharmacological inhibition of AXL by R428 recapitulated G2 arrest and polyploidy phenotype. These observations strongly suggest that acute loss of AXL in some mesenchymal HCC cells is lethal and points out that its inhibition may represent a druggable vulnerability in AXL-high HCC patients. 相似文献

The out-of-box experience (OOBE) has been identified as a significant factor contributing to user perception and acceptance of products and technologies. Whilst there has been considerable emphasis placed on formalising methodological procedures for evaluating the OOBE and on the creation of positive user experiences through appropriate interfaces and applications, relatively little work has been undertaken examining how the OOBE is impacted when the experience itself covers a range of (possibly interconnected) devices. In this paper we report the results of an empirical study which examined the OOBE when a Personal Digital Assistant (PDA) and Head Mounted Device (HMD) were configured and then connected for inter-operability purposes. Our findings show that type of device has a considerable impact on the OOBE, with the ask of interconnecting devices having a detrimental effect on the OOBE. The OOBE, however, is in main unaffected by user type and gender. 相似文献

The aim of this study was to evaluate the fatty acid composition, enzyme inhibitory, and antioxidant activities of the ethanol extracts of selected wild edible plants consumed as vegetable in the Aegean region of Turkey. In general, Mentha piperita L., Rumex patientia L., and R. acetosella L. exhibited quite strong antioxidant activities in the majority of test systems, whereas Urtica dioica L. and Eruca sativa Mill. show quite weak antioxidant activities. Enzyme inhibitory activities of the plants were found quite different than those of their antioxidant activities. Cardaria draba (L.) Desv., E. sativa, R. patientia, and E. cicutarium (L.) L’Hér. showed the highest inhibitory activities on acetylcholinesterase, butyrylcholinesterase, α-amylase, and α-glucosidase, respectively. U. dioica also showed a promising inhibitory activity on these enzymes. In parallel to the experiments, total phenolic, flavonoid, flavonol, and saponin contents of the extracts were also determined. According to the results of these assays, M. piperita had the highest amounts of phenolics, flavonols, and saponins (162.36 mg gallic acid equivalents/g extract, 3.52 mg CEs/g extract and 761.54 mg QAEs/g extract, respectively). Opopanax hispidus (Friv.) Griseb. and Lepidium sativum L. were found to be rich in flavonoid compounds (121.18 and 104.21 mg Res/g extract, respectively). In general, a strong correlation was determined between the phytochemical profile and antioxidant activity of the plant species. 相似文献
