Introduction: Resorbable synthetic scaffolds are promising for different indications, especially in the context of bone regeneration. However, they require additional biological components to enhance their osteogenic potential. In addition to different cell types, autologous blood-derived matrices offer many advantages to enhance the regenerative capacity of biomaterials. The present study aimed to analyze whether biologization of a PCL-mesh coated using differently centrifuged Platelet rich fibrin (PRF) matrices will have a positive influence on primary human osteoblasts activity in vitro. A polymeric resorbable scaffold (Osteomesh, OsteoporeTM (OP), Singapore) was combined with differently centrifuged PRF matrices to evaluate the additional influence of this biologization concept on bone regeneration in vitro. Peripheral blood of three healthy donors was used to gain PRF matrices centrifuged either at High (710× g, 8 min) or Low (44× g, 8 min) relative centrifugal force (RCF) according to the low speed centrifugation concept (LSCC). OP-PRF constructs were cultured with pOBs. POBs cultured on the uncoated OP served as a control. After three and seven days of cultivation, cell culture supernatants were collected to analyze the pOBs activity by determining the concentrations of VEGF, TGF-β1, PDGF, OPG, IL-8, and ALP- activity. Immunofluorescence staining was used to evaluate the Osteopontin expression of pOBs. After three days, the group of OP+PRFLow+pOBs showed significantly higher expression of IL-8, TGF-ß1, PDGF, and VEGF compared to the group of OP+PRFHigh+pOBs and OP+pOBs. Similar results were observed on day 7. Moreover, OP+PRFLow+pOBs exhibited significantly higher activity of ALP compared to OP+PRFHigh+pOBs and OP+pOBs. Immunofluorescence staining showed a higher number of pOBs adherent to OP+PRFLow+pOBs compared to the groups OP+PRFHigh+pOBs and OP+pOBs. To the best of our knowledge, this study is the first to investigate the osteoblasts activity when cultured on a PRF-coated PCL-mesh in vitro. The presented results suggest that PRFLow centrifuged according to LSCC exhibits autologous blood cells and growth factors, seem to have a significant effect on osteogenesis. Thereby, the combination of OP with PRFLow showed promising results to support bone regeneration. Further in vivo studies are required to verify the results and carry out potential results for clinical translation.  相似文献

Replacement of milk protein with protein isolates from vegetable resources can significantly influence the characteristics of feta whey less cheese and also decrease the cost of final production. In this study, various blends of milk protein concentrate (MPC) and pea protein isolate (PPI) were mixed at levels of 12% MPC and 0% PPI (MP0), 10% MPC and 2% PPI (MP2), 9% MPC and 3% PPI (MP3), 8% MPC and 4% PPI (MP4), 7% MPC and 5% PPI (MP5), 6% MPC and 6% PPI (MP6) and used in the manufacture of wheyless feta cheese. The chemical, textural, rheological, and sensorial properties, as well as the microstructure of the cheese samples, were evaluated after 1, 15, and 30 days of storage. The general linear model procedure of SAS statistical software was used for statistical analysis. Duncan's multiple range tests was used to compare the means of different treatments. The results showed that all properties of the cheeses were influenced by different levels of PPI due to different total solids content. The use of high concentrations of PPI resulted in a more open protein network, softer structure and decreasing the storage (G′) and loss (G″) moduli in the cheeses. Sensory evaluation of the samples revealed that total score in terms of flavor, texture and overall acceptability was gradually decreased with increasing PPI levels, but still preferable for the panelists. Furthermore, for each sample, with increasing levels of PPI, the whiteness and the greenness were decreased, but the yellowness was increased.  相似文献

This article introduces a comparative study for the simultaneous separation and preconcentration of Fe(III) and Zn(II) traces in various water samples using three well-known ligands as chelating agents and activated carbon (AC) derived from Ramelak bark as a new bio-sorbent prior to the determination by ?ame atomic absorption spectrometry. The chelating agents were 4,4’-[(4-Cyano-phenyl)methylene]bis(3-methyl-1-phenyl-1H-pyrazol-5-ol) (CMBM), diethyldithiocarbamate (DDTC) and ammonium pyrrolidine dithiocarbamate (APDC). CMBM was synthesized by a procedure reported in the literature. The newly prepared AC was characterized by Scanning Electron Microscopy (SEM) and Fourier-Transform Infrared (FTIR) Spectrometry. The analytical parameters affecting the separation efficiency of the analytes including pH, shaking time, chelating agent volume (concentration), sorbent mass, sample ?ow rate and elution conditions were investigated and discussed. Common coexisting ions did not seriously interfere with the separation, showing the good selectivity of the proposed method. The calibration graph was linear in the range of 0.35–70, 0.50–80, 0.9–100, 0.55–75, 0.75–90 and 1.0–120 ng mL?1 for Fe-CMBM, Fe-DDTC, Fe-APDC, Zn-CMBM, Zn-DDTC and Zn-APDC, respectively. Under optimized conditions, the limits of detection were 0.11, 0.13, 0.27, 0.16, 0.22 and 0.30 ng mL?1 for Fe-CMBM, Fe-DDTC, Fe-APDC, Zn-CMBM, Zn-DDTC and Zn-APDC, respectively. The proposed method has been applied to the determination of Fe(III) and Zn(II) in different water samples with satisfactory recovery percentages. The developed method, validated with standard reference materials, was used successfully in determining the concentrations of metal ions in water samples.  相似文献

While carbon nanotubes are known as efficient adsorbents for removal of a number of contaminants from water, the possibility of their leaching into drinking water has prevented their application in water treatment. In this study, single walled carbon nanotubes (SWCNT) were sandwiched between two electrospun nanofibre membranes (ENM). The relatively small pore size of the ENM prevented the mechanically entangled nanotubes from passing through and contaminating the water. The performance of the SWCNT-ENM was evaluated in a lab-scale setup for the removal of PPCPs. For this purpose, a feed solution spiked with known concentrations of six PPCPs was passed through the membrane system. The target substances included acetaminophen (ACT), bezafibrate (BZF), iopromide (IOP), diclofenac (DCF), carbamazepine (CBZ), and sulphamethoxazole (SMX). The same test was also conducted using a single contaminant (ACT). Results demonstrated a decrease in the overall percent removal of PPCPs as feed flow rate and PPCP concentration increased. For multi-component feeds containing equal amounts of the aforementioned PPCPs, the overall percent removal decreased from 90.8% to 71.0% when increasing the feed concentration from 30 to 600 μg/L. Experiments using sandwiched powdered activated carbon (PAC) showed that the dynamic adsorption capacity of PPCPs by SWCNT-ENM was higher than that of PAC-ENM, and remained unaffected by the feed composition. In addition, the high porosity of this novel membrane allowed for flow of water with low resistance such that the trans-membrane pressure was found to be as low as 4 kPa at a pure water flux of 330 L/m2h.  相似文献

Steam-assisted gravity drainage (SAGD) is a thermal enhanced oil recovery technique through drilling of two horizontal wells. Effects of steam injection temperature, well rates, and their distance on oil recovery were analyzed and optimized. Steam temperature and well distances remarkably affect SAGD performance. Four metaheuristic algorithms (particle swarm optimization (PSO), imperialist competitive algorithm, cultural algorithm, and Bees algorithm) and pattern search optimization algorithm (PSA) are used for optimization. PSO performs better than other metaheuristics and PSA is the fastest one, while it is probable to be trapped in local optimums. Hybrid PSO-PS is proposed that starts with PSO and proceeds with PSA, and tested in an SAGD project and showed excellence over other techniques.  相似文献

In fall 2009, a new speed limit of 40 km/h was introduced on local streets in Montreal (previous speed limit: 50 km/h). This paper proposes a methodology to efficiently estimate the effect of such reduction on speeding behaviors. We employ a full Bayes before–after approach, which overcomes the limitations of the empirical Bayes method. The proposed methodology allows for the analysis of speed data using hourly observations. Therefore, the entire daily profile of speed is considered. Furthermore, it accounts for the entire distribution of speed in contrast to the traditional approach of considering only a point estimate such as 85th percentile speed. Different reference speeds were used to examine variations in the treatment effectiveness in terms of speeding rate and frequency. In addition to comparing rates of vehicles exceeding reference speeds of 40 km/h and 50 km/h (speeding), we verified how the implemented treatment affected “excessive speeding” behaviors (exceeding 80 km/h). To model operating speeds, two Bayesian generalized mixed linear models were utilized. These models have the advantage of addressing the heterogeneity problem in observations and efficiently capturing potential intra-site correlations. A variety of site characteristics, temporal variables, and environmental factors were considered. The analyses indicated that variables such as lane width and night hour had an increasing effect on speeding. Conversely, roadside parking had a decreasing effect on speeding. One-way and lane width had an increasing effect on excessive speeding, whereas evening hour had a decreasing effect. This study concluded that although the treatment was effective with respect to speed references of 40 km/h and 50 km/h, its effectiveness was not significant with respect to excessive speeding-which carries a great risk to pedestrians and cyclists in urban areas. Therefore, caution must be taken in drawing conclusions about the effectiveness of speed limit reduction. This study also points out the importance of using a comparison group to capture underlying trends caused by unknown factors.  相似文献

Operational transconductance amplifiers (OTAs) are widely used in the design of electronically tunable circuits. However, electronic tunability ranges of the OTA based filters are restricted by the limited bandwidth of the transconductance gain of the OTA. Furthermore, stability conditions and the linearity of the OTA which depends on the control current restrict the tunability. In this paper, some trade-offs in the electronically tunable filters are investigated. In addition, the tunability ranges of some first and second order OTA-C and OTA-RC filters are comparatively examined. Moreover, an OTA-C all-pass filter circuit is presented. SPICE simulations are performed and stability analyses are given for both of the OTA-C and OTA-RC filters. Operation of the presented all-pass filter is verified experimentally.  相似文献
