A full-length feline immunodeficiency virus NCSU1 (FIV-NCSU1) genome (JSY3) was cloned directly from FIV-NCSU1-infected feline CD4+ lymphocyte (FCD4E) genomic DNA and identified by PCR amplification with 5' long terminal repeat, gag, env, and 3' long terminal repeat primer sets. Supernatant from FCD4E cells cocultured with JSY3-transfected Crandell feline kidney (CrFK) cells was used as an inoculum. Cell-free JSY3 virus was cytopathogenic for FCD4E lymphocytes but did not infect CrFK cells in vitro. To determine in vivo infectivity and pathogenesis, six young adult specific-pathogen-free cats were inoculated with cell-free JSY3 virus. Provirus was detected at 2 weeks postinfection (p.i.) and was still detectable at 25 weeks p.i. as determined by gag region PCR-Southern blot analysis of peripheral blood mononuclear cell lysates. Infectious virus was recovered from peripheral blood mononuclear cells at 6 and 25 weeks p.i., and an antibody response to FIV was detected by 4 weeks. In the acute phase of infection, JSY3 provirus was found only in the CD4+ lymphocyte subset; however, by 14 weeks p.i., the greatest provirus burden was detected in B lymphocytes. All six cats were panlymphopenic at 2 weeks p.i., CD4+/CD8+ ratios were inverted by 6 weeks p.i., and five of the six cats developed lymphadenopathy by 10 weeks p.i. To determine if the JSY3 molecular clone caused immunodeficiency similar to that of the parental wild-type FIV-NCSU1, the cats were challenged with the low-virulence ME49 strain of Toxoplasma gondii at 29 weeks p.i. Five of six cats developed clinical signs consistent with generalized toxoplasmosis, and three of six cats developed acute respiratory distress and required euthanasia. Histopathologic examination of the severely affected cats revealed generalized inflammatory reactions and the presence of T. gondii tachyzoites in multiple tissues. None of the six age- and sex-matched specific-pathogen-free cats inoculated with only T. gondii developed clinical disease. Our results suggest that the pathogenesis of the molecularly cloned NCSU1 JSY3 is similar to that of wild-type FIV-NCSU1.  相似文献

The effective linewidth enhancement factor αeff of DFB lasers which have both gain and index coupling coefficients is theoretically analyzed. The unique reduction mechanism of αeff due to the optical negative feedback of modal phase and modal gain in gain coupled DFB lasers was found for the first time. The numerical result showed that αeff can be reduced to almost one half of the material defined linewidth enhancement factor α when there exists an index coupling coefficient comparable to the gain coupling coefficient  相似文献

We report 8 newly established gastric-carcinoma cell lines (SNU-216, 484, 520, 601, 620, 638, 668, 719) from Korean patients. Morphologic study was carried out using light and electron microscopes. CEA, alpha FP, and CA 19-9 and TPA in supernatant and in cell lysate were measured by radioimmunoassay. p53 and c-Ki-ras gene mutations were screened and confirmed by sequencing. The cell lines, derived from tumors with moderate differentiation, grew as a diffuse monolayer, and those from tumors with poor differentiation and minimal desmoplasia grew exclusively as non-adherent. Out of the 8 gastric-cancer cell lines, 5 had detectable levels of CEA both in supernatant and in cell lysate; there was no expression or secretion of alpha FP in these cells; 4 cell lines showed high levels of CA 19-9 in cell pellets. All cell lines except SNU-484 had high concentrations of TPA both in cell lysate and in supernatants. p53 mutation was found in 6 cell lines (75%): 2 (SNU-216 and SNU-668) had mutations in exon 6, and other 3 in exon 8. The c-Ki-ras mutation was found in 2 cell lines (25%), SNU-601 and SNU-668. The former showed GGT-to-GAT transition mutation at codon 12, while the latter showed CAA-to-AAA transversion mutation at codon 61. DNA profiles using restriction endonuclease HinfI and polymorphic DNA probes ChdTC-15 and ChdTC-114 showed different unique patterns; which suggests that these cell lines are unique and not cross-contaminated. We believe that the newly characterized gastric-cancer cell lines presented in this paper will provide a useful in vitro model for studies related to human gastric cancer.  相似文献

In this study cytoskeletal antigens common to brushtail possum and tammar wallaby spermatozoa were characterised using a monoclonal antibody (PSA-10). Using indirect immunofluorescence, the PSA-10 antibody detected antigens predominantly associated with the midpiece and principal piece of mature, permeabilised marsupial spermatozoa. The principal piece determinant, shared by a variety of other species, was found to arise in the marsupial testis. Midpiece localisation of the PSA-10 epitope was detected only in marsupial spermatozoa and shown to arise in the epididymis. Immunogold labelling demonstrated that the PSA-10 antigens were predominantly associated with the fibrous sheath and midpiece fibre network of both possum and wallaby spermatozoa. Western blotting suggested that two major possum and wallaby sperm polypeptides of 158 and 182 kDa were associated with the midpiece fibre network, a cytoskeletal structure unique to marsupial spermatozoa. A 32 kDa polypeptide was associated with the principal piece fibre network and/or fibrous sheath. The finding that these marsupial sperm cytoskeletal proteins share a common linear epitope suggests that they share some sequence similarity. The midpiece fibre network of marsupial sperm, like the fibrous sheath, has been proposed to have a structural role in providing passive stiffening for the flagellum (Harding et al., 1975, 1979; Olsen, 1975). The PSA-10 monoclonal antibody may provide a tool for comparative studies of mammalian sperm cytoskeletal proteins, particularly the marsupial midpiece fibre network. It may also allow the formation of this unique marsupial cytoskeletal structure, and its fate during the fertilisation process, to be followed by immunological means.  相似文献

To assess the longer term outlook for patients who have undergone surgery for acquired (postinfarction) ventricular septal defect, we interviewed and studied 60 survivors from a single regional cardiac center between 3 and 144 months after the operation. Including the patients who died within 1 month of the operation, the 5-, 10-, and 14-year survivals (with standard errors) were 69% (65% to 74%), 50% (44% to 57%), and 37% (27% to 46%). Eighty-two percent of patients were in New York Heart Association class I or II. Ten patients (17%) had a persisting but not hemodynamically significant ventricular septal defect. Mean left ventricular ejection fraction was reduced at 0.39 (standard deviation 0.15), but this did not correlate with either New York Heart Association class or exercise tolerance. Twenty-eight patients (47%) had asymptomatic arrhythmias (17 with ventricular premature beats). Angina and other medical problems were not prevalent.  相似文献

This letter suggests a modified priority scheduling policy for the asynchronous transfer mode (ATM) multiplexer, which is called DQLT. In the dual queue length threshold (DQLT) method, there exist two queues: (1) Q1 is for nonreal-time traffic and (2) Q2 is for real-time traffic and each queue has its own threshold to adaptively control the buffer congestion. If Q1 is congested over the threshold T1 one cell at the head of Q1 moves into Q2 in a slot time. It is shown that the DQLT method gives intermediate performance between those of minimum laxity threshold (MLT) and queue length threshold (QLT) policy, but its control method is quite simpler  相似文献

All animals that are exposed to predators must distinguish dangerous from nondangerous threats and respond correctly. In reptiles, emerging hatchlings are vulnerable to a wide range of predators, particularly if they emerge during daylight. In these experiments I tested the response of pine snake, Pituophis melanoleucus, hatchlings incubated at 22-23, 27-28, or 32-33 degreesC to visual and vibratory stimuli to examine antipredator behaviour. Emerging hatchlings were exposed to one of five conditions: (1) hawk model, (2) white head model with no facial features, (3) white head model with black eyes, (4) a person, or (5) a vibration without a visual stimulus. I tested the null hypotheses of no differences in response as a function of predator type or incubation temperature. Emergence behaviour when undisturbed was affected by incubating temperature, and antipredatory behaviour was affected by both predator type and incubation temperature. Pine snake hatchlings responded more protectively (withdrawal into tunnels) than defensively (striking), responded with less intensity to a vibration compared with visual predator stimuli, and required longer to respond to a head model without eyes than to all other predator types. Given the relatively small size of hatchlings, it is adaptive for them to withdraw into the nest rather than attack a predator. Hatchlings from eggs that were incubated at medium temperatures required less time to emerge from their underground nests when undisturbed, and had stronger protective responses than snakes incubated at other temperatures. These results suggest that hatchlings incubated at medium temperatures are generally less vulnerable to predators than hatchlings incubated at higher or lower temperatures. We investigate the effect of relative wavelength difference between material gain and resonance peaks on lasing performance in vertical-external-cavity surface-emitting lasers (VECSELs). We propose a measurement method of the wavelength-dependent modal gain profiles by inserting a birefringent filter inside a cavity in order to modulate the optical loss according to the wavelength. With knowledge of the detailed gain profiles, we achieve a stable green output power of 3.3 W and a power conversion efficiency (= green output power/pump input power) of 17% in VECSEL with a LiB3O5 crystal as an intracavity frequency doubler  相似文献
