There are large differences in sensitivity to the toxic and biochemical effects of dioxins and dioxin-like compounds (DLCs) among vertebrates. Previously, we demonstrated that the difference in sensitivity between domestic chicken (Gallus gallus domesticus) and common tern (Sterna hirundo) to aryl hydrocarbon receptor 1 (AHR1)-dependent changes in gene expression following exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) is based upon the identities of the amino acids at two sites within the ligand binding domain of AHR1 (chicken--highly sensitive; Ile324_Ser380 vs common tern--250-fold less sensitive than chicken; Val325_Ala381). Here, we tested the hypotheses that (i) the sensitivity of other avian species to TCDD, 2,3,4,7,8-pentachlorodibenzofuran (PeCDF), and 2,3,7,8-tetrachlorodibenzofuran (TCDF) is also determined by the amino acids at sites that are equivalent to sites 324 and 380 in chicken, and (ii) Ile324_Ala380 and Val324_Ser380 genotypes confer intermediate sensitivity to DLCs in birds. We compared ligand-induced transactivation function of full-length AHR1s from chicken, common tern, ring-necked pheasant (Phasianus colchicus; Ile324_Ala380) and Japanese quail (Coturnix japonica; Val324_Ala380), and three Japanese quail AHR1 mutants. The results support our hypothesis that avian species can be grouped into three general classes of sensitivity to DLCs. Both AHR1 genotype and in vitro transactivation assays predict in vivo sensitivity. Contrary to the assumption that TCDD is the most potent DLC, PeCDF was more potent than TCDD at activating Japanese quail (13- to 26-fold) and common tern (23- to 30-fold) AHR1. Our results support and expand previous in vitro and in vivo work that demonstrated ligand-dependent species differences in AHR1 affinity. The findings and methods will be of use for DLC risk assessments.  相似文献

Invertase was immobilized into polyacrylamide–gelatin carrier system by chemical cross-linking with chromium (III) acetate, chromium (III) sulphate, and potassium chromium (III) sulphate. The optimum conditions, namely substrate concentration, temperature, and pH were determined. The effect of polyacrylamide–gelatin ratio and cross-linker concentration on immobilized enzyme activity were analysed. Maximum immobilized enzyme activities were obtained with chromium (III) acetate (0.01 mol dm−3), chromium (III) sulphate (0.004 mol dm−3) and potassium chromium (III) sulphate (0.001 mol dm−3) for 0.177 (w/w) polyacrylamide–gelatin carrier ratio as 79%, 72% and 79%, respectively. The Km values were 86 and 166 mM for free and immobilized enzyme, respectively. All immobilized samples were used 20 times over a period of 2 months without a considerable loss of activity.  相似文献

Microstructural and phase transformations during sintering of a Phillipsite rich zeolitic tuff, from Tenerife, Canary Islands, was investigated in order to their utilization in ceramic manufacturing industries. Green samples were obtained from powders and pressed at 150 MPa and heat-treated for 1 hour in the temperature range between 900-1080 °C. The zeolitic tuffs show endothermic peaks at about 100 °C and 200 °C, corresponding to dehydration of zeolitic water observed in the thermo- gravimetric curves and at ∼700 °C a small endothermic peak was identified corresponding to the structural breakdown of Phillipsite. According to the dilatometric traces, at about 870 °C, sintering of solid particles starts and a linear shrinkage of about 6% is reached at about 1050 °C. The maximum absolute and apparent densities were obtained after sintering at 1040 °C (absolute density = 2.59 g/cm3; apparent density = 2.33 g/cm3). Over this temperature, the sample heat-treated at 1060 °C, density results show a decreasing trend. The chemical composition of studied zeolitic tuffs make possible the liquid phase formation during heat-treatment, through the supplement of alkaline oxides. SEM image of the sample obtained at 1040 °C shows a zone with micro-crystallinity around the boundary of Sanidine grain highlighting the beginning of the phase transition from Sanidine, to Microcline. Heat-treatment effect of zeolitic tuff leads to the decomposition of Phillipsite and Sanidine and the formation of a new crystal phase of Hematite.  相似文献

Characterization of the fatty acid and triacylglycerol composition of terebinth fruit oil and the synthesis of structured lipids (SL) were performed in this study. Interesterification reaction of terebinth fruits oil (Pistacia terebinthus L.) with caprylic acid (CA) and stearic acid (SA) to produce a SL was performed in n-hexane using immobilized sn-1,3 specific lipase from Mucor miehei. The effect of reaction conditions and relationship among them were analyzed by response surface methodology (RSM) with a four-factors five-level central composite rotatable experimental design. The four major factors chosen were enzyme load (10–30 wt% based on substrates), reaction time (7–18 h), reaction temperature (40–60 °C) and substrate mole ratio (terebinth oil:SA:CA 1:1:1–1:1:3). The best fitting quadratic model was determined by regression and backward elimination. Based on the fitted model, the optimal reaction conditions for the incorporation of CA and SA were found to be temperature 50 °C; time 18 h; enzyme load 30 wt%; substrate ratio 1:1:3. Under these optimum conditions, the incorporation of SA and CA could be obtained as 19 and 14%, respectively.  相似文献

The effect of fortification of yoghurt with sodium–calcium caseinate (SCC) and whey protein concentrate (WPC) on some properties of set‐type yoghurt were investigated. The addition of WPC enhanced the viability of Lactobacillus delbrueckii subsp. bulgaricus more than SCC. The highest firmness values were obtained from SCC‐fortified yoghurts, whereas yoghurts fortified with WPC had the highest water‐holding capacity during storage. The yoghurts fortified with 4% w/w SCC or 4% w/w WPC had the highest viscosity. Yoghurts fortified with 2% w/w SMP, SCC or WPC showed similar taste and overall acceptability scores; however, samples containing 4% w/w SCC or 4% w/w WPC had the lowest scores.  相似文献

In this study, phenolic compounds of Rhododendron honey (also known as mad honey) samples collected from the Black Sea Region were identified using high performance liquid chromatography-diode array detector system. The major phenolic substances in Rhododendron honeys were found to be chlorogenic and coumaric acids with the amounts of 0.11–191.54 mg/kg and 0–82.83 mg/kg, respectively. Gallic and ferulic acids were detected in the most honey samples. Additionally, significant correlations were determined between the phenolic substances. The present study showed that Rhododendron honeys contained higher quantities of phenolic acids than flavonoids. Chlorogenic and coumaric acids were the dominant phenolic substances detected in honey samples.  相似文献

We present the effects of surface treatments on the structural and electronic properties of chemomechanically polished Cd0.9Zn0.1Te before contact deposition. Specifically, polished CdZnTe (CZT) samples were treated with four distinct chemical etchants: (1) bromine methanol (BM), (2) bromine in lactic acid, (3) bromine in methanol followed by bromine–20% lactic acid in ethylene glycol, and (4) hydrochloric acid (HCl). The surface structure and surface electronic properties were studied with atomic force microscopy (AFM) and x-ray photoelectron spectroscopy (XPS). AFM images showed that three of the four etchants significantly altered the surface morphology and structure of CZT. All etchants created smoother surfaces; however, all except HCl also introduced high densities of defects. HCl was found to not affect the surface structure. XPS measurements indicated that a thick, ~3 nm to 4 nm, TeO2 layer formed about 1 h after etching; hence, it is very important to process devices immediately after etching to prevent oxide formation.  相似文献

In this study, N-pyrrole (Py), N-phenylpyrrole (PhPy), and 1[4-methoxyphenyl]-1H-pyrrole (MPhPy) homopolymers were synthesized electrochemically onto carbon fiber microelectrodes (CFMEs). The influences of the substituent effect on electrochemical impedance spectroscopy (EIS) were studied comparatively. All the monomers were electrodeposited in 0.05 M tetraethyl ammonium perchlorate (TEAP)/dichloromethane (CH2Cl2) solution and characterized by cyclic voltammetry (CV), Fourier transform infrared reflectance spectrophotometry (FTIR-ATR), scanning electron microscopy (SEM), and atomic force microscopy (AFM). The morphological study reveals that the polymers were deposited as a continuous and well adhered film to surface of the CFME. An equivalent electrical circuit for three different monomers on CFMEs was proposed and experimental data were simulated to obtain the numerical values of circuit components. All results support the high quality film deposition that resulted in desired electronic properties due to the electron donating behaviors of substituent group of phenyl and methoxy.  相似文献

The roles of Bcl-2 protein and the protein ratio of Bcl-2/Bax in regulating cell growth in various lymphoma cell lines were examined. A dose-dependent decrease in Bcl-2 protein expression was observed in the different lymphomas incubated with lipid-incorporated bcl-2 antisense oligonucleotides (L-bcl-2). Growth inhibition was observed in a transformed follicular lymphoma (FL) cell line, which has the t(14;18) translocation and Bcl-2 protein overexpression. One of the mechanisms by which L-bcl-2 growth inhibition is mediated in these transformed FL cells might be through apoptotic induction, because the treated cells had an increased apoptotic index and showed the typical DNA fragmentation. These studies indicate that Bcl-2 protein is critical in the growth regulation of transformed FL cells. L-bcl-2 did not induce growth inhibition in lymphoma cells not expressing Bcl-2 or Bax protein. Thus, the protein ratio of Bcl-2/Bax may also be important in regulating the growth of these lymphomas.  相似文献

(1) Background: Placental immune cells are playing a very important role in a successful placentation and the prevention of pregnancy complications. Macrophages dominate in number and relevance in the maternal and the fetal part of the placenta. The evidence on the polarization state of fetal and maternal macrophages involved in both, healthy and pregnancy-associated diseases, is limited. There is no representative isolation method for the direct comparison of maternal and fetal macrophages so far. (2) Material and Methods: For the isolation of decidual macrophages and Hofbauer cells from term placenta, fresh tissue was mechanically dissected and digested with trypsin and collagenase A. Afterwards cell enrichment was increased by a Percoll gradient. CD68 is represented as pan-macrophage marker, the surface markers CD80 and CD163 were further investigated. (3) Results: The established method revealed a high cell yield and purity of the isolated macrophages and enabled the comparison between decidual macrophages and Hofbauer cells. No significant difference was observed in the percentage of single CD163+ cells in the distinct macrophage populations, by using FACS and immunofluorescence staining. A slight increase of CD80+ cells could be found in the decidual macrophages. Considering the percentage of CD80+CD163 and CD80CD163+ cells we could not find differences. Interestingly we found an increased number of double positive cells (CD80+CD163+) in the decidual macrophage population in comparison to Hofbauer cells. (4) Conclusion: In this study we demonstrate that our established isolation method enables the investigation of decidual macrophages and Hofbauer cells in the placenta. It represents a promising method for direct cell comparison, enzyme independently, and unaffected by magnetic beads, to understand the functional subsets of placental macrophages and to identify therapeutic targets of pregnancy associated diseases.  相似文献
