Hard-X-ray dark-field imaging using a grating interferometer   总被引:1,自引:0,他引:1  
Imaging with visible light today uses numerous contrast mechanisms, including bright- and dark-field contrast, phase-contrast schemes and confocal and fluorescence-based methods. X-ray imaging, on the other hand, has only recently seen the development of an analogous variety of contrast modalities. Although X-ray phase-contrast imaging could successfully be implemented at a relatively early stage with several techniques, dark-field imaging, or more generally scattering-based imaging, with hard X-rays and good signal-to-noise ratio, in practice still remains a challenging task even at highly brilliant synchrotron sources. In this letter, we report a new approach on the basis of a grating interferometer that can efficiently yield dark-field scatter images of high quality, even with conventional X-ray tube sources. Because the image contrast is formed through the mechanism of small-angle scattering, it provides complementary and otherwise inaccessible structural information about the specimen at the micrometre and submicrometre length scale. Our approach is fully compatible with conventional transmission radiography and a recently developed hard-X-ray phase-contrast imaging scheme. Applications to X-ray medical imaging, industrial non-destructive testing and security screening are discussed.  相似文献

The complexity and variety of bibliographic data is growing, and efforts to define new methodologies and techniques for bibliometric analysis are intensifying. In this complex scenario, one of the most crucial issues is the quality of data and the capability of bibliometric analysis to cope with multiple data dimensions. Although the problem of enforcing a multidimensional approach to the analysis and management of bibliographic data is not new, a reference design pattern and a specific conceptual model for multidimensional analysis of bibliographic data are still missing. In this paper, we discuss ten of the most relevant challenges for bibliometric analysis when dealing with multidimensional data, and we propose a reference data model that, according to different goals, can help analysis designers and bibliographic experts in working with large collections of bibliographic data.  相似文献

We modified and optimized a first generation quadrupole time-of-flight (Q-TOF) 1 to perform tandem mass spectrometry on macromolecular protein complexes. The modified instrument allows isolation and subsequent dissociation of high-mass protein complexes through collisions with argon molecules. The modifications of the Q-TOF 1 include the introduction of (1) a flow-restricting sleeve around the first hexapole ion bridge, (2) a low-frequency ion-selecting quadrupole, (3) a high-pressure hexapole collision cell, (4) high-transmission grids in the multicomponent ion lenses, and (5) a low repetition rate pusher. Using these modifications, we demonstrate the experimental isolation of ions up to 12 800 mass-to-charge units and detection of product ions up to 38 150 Da, enabling the investigation of the gas-phase stability, protein complex topology, and quaternary structure of protein complexes. Some of the data reveal a so-far unprecedented new mechanism in gas-phase dissociation of protein oligomers whereby a tetramer complex dissociates into two dimers. These data add to the current debate whether gas-phase structures of protein complexes do retain some of the structural features of the corresponding species in solution. The presented low-cost modifications on a Q-TOF 1 instrument are of interest to everyone working in the fields of macromolecular mass spectrometry and more generic structural biology.  相似文献

Red blood cells (RBCs) scatter light mainly in the forward direction, where the scattering phase function has a narrow peak. We performed an experimental investigation into the angular distribution of light scattered by blood in the small-angle domain. A highly diluted suspension of RBCs (hematocrits in the range 5 x 10(-5)-10(-2)) was illuminated with a He-Ne laser with 633 nm wavelength. We focused our research on two main topics: the scattering efficiency of the RBCs given by the mean scattering cross section and the scattering anisotropy obtained from the angular distribution of the scattered photons. The collimated beam transmission and the angular distribution of scattered light were measured and compared with the predictions of the effective phase function model. The RBCs' mean scattering cross section and scattering anisotropy were obtained by fitting of the experimental data.  相似文献

Cardiovascular system functions are impaired in altered gravity conditions. In particular endothelial cells play a major role being responsible for the integrity of the vascular wall. Due to obvious difficulties in performing continuous and exhaustive experiments in space, most of the available data have been obtained so far using various simulators of hypergravity and microgravity (µg) conditions. The consistency of the data resides on the reliability of the simulator, being a critical point in the development of the research. We exposed the cell cultures to 1) hypergravity (launch condition) using MidiCAR at Dutch Experiment Support Center (DESC, NL); 2) simulated µg using the Rotating Wall Vessel (RWV) and the Random Positioning Machine (RPM). We used two different cellular models: human umbilical vein endothelial cells (HUVEC) and human leukocytes (U937). Only few experiments on cells using RPM have been reported. To assess the RPM best operative parameters we considered data from experiments in space on U937 as reference standard. Differently, cultures in modelled µg using RWV have been extensively reported. Our data on HUVEC indicate that the two µg simulators provide analogous results in terms of proliferation and cytoskeletal organization. Finally, to investigate the effects of spaceflight on different human cells, we developed a spaceflight-like protocol consisting of an initial hypergravity phase (launch), followed by a µg simulation (orbital flight). Using this protocol, results show that hypergravity limits in our models the effects on proliferation induced by modelled µg.  相似文献

A crucial point in developing walking machines is the load to weight ratio. Leg design and in it the drives essentially determine the leg's weight. Therefore it makes sense to optimize the design as well as the best motor-gear-combination with respect to power density. Some typical principles of legs have been derived from biological findings, especially for insects. The technical solution was adapted to these principles in an optimal way. Explored whether disgusting or disrespectful actions are judged to be moral violations, even when these actions are harmless. Stories about victimless yet offensive actions (such as cleaning one's toilet with a flag) were presented to Brazilian and US adults and children of high and low SES (N?=?360). Results show that college students at elite universities judged these stories to be matters of social convention or of personal preference. Most other Ss, especially in Brazil, took a moralizing stance toward these actions. For these latter Ss, moral judgments were better predicted by affective reactions than by appraisals of harmfulness. Results support the claims of cultural psychology (R. A. Shweder, 1991) and suggest that cultural norms and culturally shaped emotions have a substantial impact on the domain of morality and the process of moral judgment.
