A lap-shear test was employed to investigate the failure mechanism of a chlorinated polyolefin (CPO) coating on a high-modulus thermoplastic olefin (TPO) substrate fabricated as a blend of a highly crystalline Ziegler-Natta isotactic polypropylene (iPP) and a crystalline metallocene poly(ethylene-butene) (9 wt% butene, EB9) impact modifier. The CPO was a chlorinated polypropylene containing 20 wt% Cl. The results showed that the fracture strength increased with increasing EB9 content in TPO blends. They also showed that the presence of xylene vapor during the bake step improved the adhesion between CPO and iPP itself (by 40%), but had a much smaller effect for the TPOs. Optical and transmission electronic microscopy images revealed a well-defined skin layer approximately 230 μm thick at the mold surface of the injection molded substrates. For the 25 wt% EB9 blend (TPO25), this skin layer consists of thin fibers of EB trapped in a transcrystalline iPP matrix, with crystalline lamellae propagating from the matrix across the EB9 domains. Laser scanning confocal fluorescence microscopy (LCFM) and scanning electron microscopy images of iPP/CPO/iPP samples indicate that failure occurred close to the interface between the CPO and the iPP substrate, and, during fracture, the CPO layer maintained its original thickness. For the TPO/CPO/TPO sandwich samples, the fracture surfaces themselves were much rougher than that between CPO and iPP. Substantial deformation of the CPO layer was seen in the fractured samples, and failure was due primarily to cohesive fracture of the CPO in the region adjacent to the TPO substrate. From the perspective of newly introduced environmental regulations restricting aromatic hydrocarbons in automotive coatings, the most important result was the strong adhesion between CPO and TPO25, with little difference between the samples exposed to xylene vapor and those not exposed to xylene. 相似文献
The glucooligosaccharides (GOS), produced by Leuconostoc mesenteroides NRRL B-1299 dextransucrase through an acceptor reaction with maltose and sucrose, were purified by reverse phase chromatography. Logarithmic plots of retention time vs. dp of the GOS gave three parallel lines suggesting the existence of at least three families of homologous molecules. The structure (13C and 1H NMR spectroscopy) and reactivity of the purified molecules of the three families were investigated. All the products bear a maltose residue at the reducing end. The GOS in the first family (named OD) contained additional glucosyl residues all alpha-(1-->6) linked. The smallest molecule in this first series was panose or alpha-D-glucopyranosyl-(1-->6)-D-maltose (dp 3). All the OD molecules were shown to be good acceptors for dextransucrase in the presence of sucrose. The second family, named R, was composed of linear GOS containing alpha-(1-->6)-linked glucosyl residues and a terminal alpha-(1-->2)-linked residue at the non-reducing end of the molecule; the smallest molecule in this family was alpha-D-glucopyranosyl-(1-->2)-D-panose (dp 4). The third family, R', was formed of GOS containing additional residues linked through alpha-(1-->6) linkages that constitute the linear chain, and an alpha-(1-->2)-branched residue located on the penultimate element of the chain, near the non-reducing end. The smallest molecule in this series is alpha-D-glucopyranosyl-(1-->6)-[alpha-D-glucopyranosyl-(1-->2)]-alpha-D- glucopyranosyl-(1-->6)-D-panose, dp 6. R and R' GOS are very poor acceptors for L. mesenteroides NRRL B-1299 dextransucrase. This study makes it possible to suggest a rather simple reaction scheme, where molecules Ri, R'i and ODi of the same dp all result from the glucosylation of the same GOS: ODi-l. 相似文献
Sedimentary basins in the NE Caribbean Plate Boundary Zone are associated with the evolution of a Cretaceous to Early Tertiary island arc, and with carbonate banks which formed following cessation of volcanism in the Eocene to Oligocene. Neogene sedimentary basins are typically present where crustal blocks in the plate boundary zone have undergone divergence, with or without rotation. Cretaceous through Oligocene basins may also have formed during divergence between crustal blocks, although this is difficult to prove owing to limited exposure and subsequent tectonic deformation. Other basins are associated with localized overthrusting along the plate boundaries. Absence of source rocks and complicated and variable geothermal gradients make the area one of high risk for petroleum exploration. However. the paucity of data collected in the region allows for significant future discoveries, and the presence of minor seeps and traces of petroleum from two wells gives some hope. Finally, there is some evidence that seismic reflection amplitude anomalies may be associated with gas occurrences in the NE Caribbean, such that imaging of "bright spots" may prove to be a successful exploration tool. 相似文献
BACKGROUND: Immunization to platelet alloantigens can occur during pregnancy or after the transfusion of blood components. Platelet alloantibodies can cause neonatal alloimmune thrombocytopenia and posttransfusion purpura. Transfusion-induced alloimmunization to a novel platelet alloantigen system, Gov, expressed on the 175-kDa glycosyl phosphatidylinositol-anchored platelet glycoprotein, CD109, was previously described. This report describes three unrelated patients who were alloimmunized to Gov(a) or Gov(b) during pregnancy. STUDY DESIGN AND METHODS: Platelets were typed by using radioimmunoprecipitation for HPA-1a, -3a, -5a, -5b, Gov(a), and Gov(b) and by polymerase chain reaction-restriction fragment length polymorphism for HPA-1a, -1b, -3a, and -3b. Maternal sera were screened for platelet antibodies by using radioimmunoprecipitation and the antigen capture assay. RESULTS: Patients 1 and 2 were investigated after the diagnosis of neonatal alloimmune thrombocytopenia in their children, and alloantibodies specific for Gov(b) and Gov(a), respectively, were detected in maternal serum. Serum from patient 3, who had mild idiopathic thrombocytopenia purpura with no detectable autoantibody, was found to contain alloantibodies to Gov(b) and to HPA-5b, presumably as a result of immunization during pregnancy. Platelet typings confirmed that the patients were at risk for alloimmunization to the respective antigen. CONCLUSION: This report of three cases of maternal alloimmunization to antigens in the Gov system indicates that immunization can occur via placental transfer of antigen and that Gov system alloantibodies may be associated with neonatal alloimmune thrombocytopenia. 相似文献
Replacing wheatflour (WHF) with defatted fluted pumpkin (Telfairia occidentalis Hook) seed flour (FPF) at levels of 0–25% was investigated for its effect on chemical, physical, sensory and nutritional properties of cookies (biscuits). Protein quality was evaluated by using weanling albino rats fed a diet of cookies, which were formulated to supply 10% protein, with a casein diet as a control. Up to 15% substitution of WHF with FPF produced acceptable cookies with spread ratio, hardness, colour and flavour similar to the control (100% WHF) cookies. Use of higher levels (20–25%), however, resulted in reduction in these quality attributes. When WHF was replaced with 15% FPF, there was an increase of 84.6% in crude protein, 62.9% in calcium, 131.0% in potassium and 61.6% in phosphorus contents of composite cookies. There were no significant (P > 0.05) differences between the values obtained for protein efficiency ratio, net protein ratio and true digestibility of cookie diets containing 15–20% FPF, and casein when fed to rats. 相似文献
Unnatural amino acids with bioorthogonal reactive groups have the potential to provide a rapid and specific mechanism for covalently inhibiting a protein of interest. Here, we use mutagenesis to insert an unnatural amino acid containing an azide group (Z) into the target protein at positions such that a “click” reaction with an alkyne modulator (X) will alter the function of the protein. This bioorthogonally reactive pair can engender specificity of X for the Z‐containing protein, even if the target is otherwise identical to another protein, allowing for rapid target validation in living cells. We demonstrate our method using inhibition of the Escherichia coli enzyme aminoacyl transferase by both active‐site occlusion and allosteric mechanisms. We have termed this a “clickable magic bullet” strategy, and it should be generally applicable to studying the effects of protein inhibition, within the limits of unnatural amino acid mutagenesis. 相似文献
Purpose: Nanoparticle size is important in drug delivery. Clearance of nanoparticles by cells of the reticuloendothelial system has been reported to increase with increase in particle size. Further, nanoparticles should be small enough to avoid lung or spleen filtering effects. Endocytosis and accumulation in tumor tissue by the enhanced permeability and retention effect are also processes that are influenced by particle size. We present the results of studies designed to optimize cross-linked biodegradable stealth polymeric nanoparticles fabricated by dispersion polymerization.
Methods: Nanoparticles were fabricated using different amounts of macromonomer, initiators, crosslinking agent and stabilizer in a dioxane/DMSO/water solvent system. Confirmation of nanoparticle formation was by scanning electron microscopy (SEM). Particle size was measured by dynamic light scattering (DLS). D-optimal mixture statistical experimental design was used for the experimental runs, followed by model generation (Scheffe polynomial) and optimization with the aid of a computer software. Model verification was done by comparing particle size data of some suggested solutions to the predicted particle sizes.
Results and conclusion: Data showed that average particle sizes follow the same trend as predicted by the model. Negative terms in the model corresponding to the cross-linking agent and stabilizer indicate the important factors for minimizing particle size. 相似文献
In the present work, we carry out evaluation of gas flow maldistribution in different types of gas distributing devices (GDD) and packings by using experimental data for the output velocity profiles for both the absorption pilot column and industrial column. Two GDD construction types—with circular and with local tube gas feed, together with their modifications—are investigated. Two pilot columns—with random packing RSRM 1.5″ and with structured ceramic Honey-comb packing, and one industrial contact economizer with Honey-comb and inclined rings—have been studied. Six types of quantitative estimations of GDD and packings maldistribution are determined by the help of MATHCAD. Then, their sensitivity toward several factors, such as the type of GDD and packing, the initial gas flow velocity, the number and dimension of measuring cells on the column cross-section, the measurement error, and so on, is investigated. It is found that only two of the quantitative maldistribution estimations take into account the formation of maldistribution clusters, which deteriorates the uniform gas distribution. A novel method for quantitative determination of these clusters and also for identification of the zones at column cross-section, in which they appeared, has been developed. It is established that the measurement error and the dimension of measuring cell deeply influence the number, the area, and spatial distribution of these zones on the cross-section, as well as the values of maldistribution estimates. 相似文献
