Entry inhibitors against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) are urgently needed to control the outbreak of coronavirus disease 2019 (COVID-19). This study developed a robust and straightforward assay that detected the molecular interaction between the receptor-binding domain (RBD) of viral spike protein and the angiotensin-converting enzyme 2 (ACE2) receptor in just 10 min. A drug library of 1068 approved compounds was used to screen for SARS-CoV2 entry inhibition, and 9 active drugs were identified as specific pseudovirus entry inhibitors. A plaque reduction neutralization test using authentic SARS-CoV-2 virus in Vero E6 cells confirmed that 2 of these drugs (Etravirine and Dolutegravir) significantly inhibited the infection of SARS-CoV-2. With molecular docking, we showed that both Etravirine and Dolutegravir are preferentially bound to primary ACE2-interacting residues on the RBD domain, implying that these two drug blocks may prohibit the viral attachment of SARS-CoV-2. We compared the neutralizing activities of these entry inhibitors against different pseudoviruses carrying spike proteins from alpha, beta, gamma, and delta variants. Both Etravirine and Dolutegravir showed similar neutralizing activities against different variants, with EC50 values between 4.5 to 5.8 nM for Etravirine and 10.2 to 22.9 nM for Dolutegravir. These data implied that Etravirine and Dolutegravir may serve as general spike inhibitors against dominant viral variants of SARS-CoV-2. 相似文献

A radio frequency (RF) plasma system was used to decompose the ethoxyethane (EOE) contained gas. The reactants and final products were analyzed by using an FTIR (Fourier Transform Infrared) spectrometer. The effects of plasma operational parameters, including input power wattage (W), equivalence ratios (Φ), feeding concentration (C) of EOE and total gas flow rate (Q) for EOE decomposition were evaluated. In addition, the possible reaction pathways for EOE decomposition and the formation of final products were built up and are discussed in this paper. The mole fraction profiles of C2H5OC2H5, CH3CHO, CH4, C2H6, C2H4, C2H2, CO2 and CO were detected and are also presented in this paper. At lower input power wattages, the creation of glow discharge is strongly dependent on the plasma production index ( PPI ). When input power wattages are smaller than 30 W, the minimum values of PPI to create glow discharge ranged between 18.2 and 19.0. The results of this study revealed that, in the RF plasma reactor, the decomposition fraction of EOE could reach 100% under most operational conditions. © 2000 Society of Chemical Industry 相似文献

Fractionation of an EtOAc-soluble fraction of the solid fermentate of an endophytic fungus, Lachnum abnorme Mont. BCRC 09F0006, derived from the endemic plant, Ardisia cornudentata Mez. (Myrsinaceae), resulted in the isolation of three new chromones, lachnochromonins D–F (1–3), one novel compound, lachabnormic acid (4), along with nine known compounds (5–13). Their structures were elucidated by spectroscopic analyses. Alternariol-3,9-dimethyl ether (6) was given the correct data as well as 2D spectral analyses for the first time. This is the first report of the isolation of one unprecedented compound 4 from Lachnum genus, while all known compounds were also found for the first time from Lachnum. The effects of some isolates (3, 4, 7–9, 10, and 13) on the inhibition of nitric oxide (NO) production in lipopolysaccharide (LPS)-activated RAW 264.7 murine macrophages were also evaluated. The UV‐curable urethane‐acrylates based on 2‐hydroxyethyl methacrylate (HEMA)‐terminated polyurethane (PU) for lithographic and coating applications are investigated in this study. Series of PU prepolymers were made from 4,4‐diphenyl methane diisocyanate (MDI), poly(propylene oxide) glycol (PPG 400), poly(butylene adipate)glycol (PBA 500), or poly(tetramethylene oxide) glycol (PTMO 1000) and are terminated with HEMA. The 2,2‐azobisisobutyronitrile (AIBN) was used as a UV‐initiator under air atmosphere. The curing kinetics of HEMA‐terminated PU film were studied. The curing analysis, using FTIR and reaction kinetics, indicate the reaction rate equation correlates well with the film thickness [T], initiator concentration [I], unreacted double bond concentration [C?C], and exposed energy [E] of the reaction system. Optimal process conditions of thin‐wall injection molding of a cellular phone cover were investigated with the consideration of interaction effects between process parameters. L27 experimental tests based on Taguchi's method were performed, and then Cyclone Scanner, PolyCAD and PolyWorks were used to measure the shrinkage and warpage of the thin‐wall injected parts to determine the optimal process conditions. Based on the results of the analysis of variables and the F‐test, interaction effects for each observed factor were determined. The results indicated that the packing pressure was the most important process parameter affecting the shrinkage and warpage of the thin‐wall part. The optimal process conditions were different for the shrinkage and the warpage. This was because during the injection process, the mechanisms affecting shrinkage or warpage were different. Compared with the results obtained with simplified thin‐wall parts in the literature, it was found that the geometry of a real commercial part did affect the optimal process conditions and the order of influence of process parameters. One of the major survival challenges of premature birth is production of lung surfactant. The lipid component of surfactant,
dipalmitoyl PC (DPPC), increases in concentration in the period before normal term birth via a net shift in FA composition away from unsaturates. We investigated the influence of dietary DHA and arachidonic acid (AA)
on lung FA composition and DPPC concentration in term and preterm baboons. Pregnant animals/neonates were randomized to one
of four groups: breast-fed (B), term formula-fed (T−), preterm formulafed (P−), and preterm fed formula supplemented with DHA-AA (P+). Breast milk contained 0.68%wt DHA and the P+ group formula contained 0.61%wt DHA. In the preterm groups (P− and P+), pregnant females received a course of antenatal corticosteroids. At the adjusted age of 4 wk, neonate lung tissue was harvested,
and FA composition and DPPC were analyzed. Palmitate was ∼28%wt of lung total FA and no significant differences were found
among the four treatment groups. In contrast, DPPC in the B group lung tissue was significantly greater than DPPC in the unsupplemented
groups, but not compared with the P+ group. The B and P+ groups were not significantly different in DHA and AA, but were different compared with the unsupplemented (T, P−) groups. These results indicate that LCP supplementation increases lung DHA and AA, without compromising overall lung 16∶0
or DPPC. The shift in FA composition toward greater unsaturation in the groups consuming LCP supported improved surfactant

The effects of molecular weight and molecular structure of styrene(St)‐based and vinyl acetate(VAc)‐based low‐profile additive (LPA) on the curing kinetics and compatibility of unsaturated polyester (UP)/LPA system and linear shrinkage, water absorption rate, surface gloss and pigmentability of bulk molding compound (BMC) were investigated. Results show that the curing reaction rate decreases with an increase of the molecular weight of LPA due to the chain entanglement effect. The plasticizing effect of LPA on the (UP) network was reduced with an increase of the molecular weight of LPA. Water absorption of BMC increases as the molecular weight of LPA increases, implying that more microvoids were formed inside the BMC, resulting a lower linear shrinkage rate, and worse pigmentability. However, good shrinkage control LPA does not necessarily lead to a smoother surface and better surface gloss. Furthermore, modified LPAs possess better compatibility with UP, the final curing conversion of UP is elevated, and both better shrinkage control and surface properties are also observed. 相似文献
