Inflammation and oxidative stress are thought to be involved in, or associated with, the development of obesity, dyslipidemia, hepatic steatosis, and insulin resistance. This work was designed to determine the evolution of inflammation and oxidative stress during onset and progression of hepatic steatosis and glucose intolerance. Seventy-five male Wistar rats were divided to control and high-fat high-fructose (HFHFr) groups. A subgroup of each group was sacrificed at 4, 8, 12, 16, and 20 weeks. HFHFr-fed rats exhibited overweight, glucose intolerance, and hepatic steatosis with increased contents of hepatic diacylglycerols and ceramides. The HFHFr diet increased hepatic interleukin 6 (IL-6) protein and adipose tissue CCL5 gene expression and hepatic nicotinamide adenine dinucleotide phosphate (NADPH) oxidase activity but not mitochondrial reactive oxygen species (ROS) production. The HFHFr diet decreased plasma and liver levels of isoprostanoid metabolites as well as plasma thiobarbituric acid-reactive substance (TBARS) levels. Hepatic glutathione content was decreased with a moderate decrease in superoxide dismutase (SOD) and glutathione peroxidase (GPx) with the HFHFr diet. Overall, HFHFr diet led to hepatic lipid accumulation and glucose intolerance, which were accompanied by only moderate inflammation and oxidative stress. Most of these changes occurred at the same time and as early as 8 or 12 weeks of diet treatment. This implies that oxidative stress may be the result, not the cause, of these metabolic alterations, and suggests that marked hepatic oxidative stress should probably occur at the end of the steatotic stage to result in frank insulin resistance and steatohepatitis. These findings need to be further evaluated in other animal species as well as in human studies.  相似文献

The MEGAPIE project aimed to design, build and operate a liquid metal spallation neutron target of about 1 MW beam power in the SINQ facility at the Paul Scherrer Institut (Villigen, Switzerland). This project is an important step in the roadmap towards the demonstration of the accelerator driven system (ADS) concept and high power liquid metal targets in general. Following the design phase, an experimental program was defined to provide a complete characterization of the facility by performing a “mapping” of the neutron flux at different points, from the center of the target to the beam lines. The neutronic performance of the target was studied using different experimental techniques with the goals of validating the Monte Carlo codes used in the design of the target; additionally, the performance was compared with the solid lead targets used before and after the MEGAPIE experiment.  相似文献

Eight calibration transfer methods based on the removal of orthogonal signal were compared for the standardization of whole soybean protein and oil models. Dynamic orthogonal projection (DOP), transfer by orthogonal projection (TOP), error removal by orthogonal subtraction (EROS), orthogonal signal correction (OSC), and orthogonal projections to latent structures (O-PLS) as well as the modification and extension of some of these methods were compared in the transfer of models in intra and inter-brand situations using two Foss Infratecs and two Bruins OmegAnalyzerGs. For each brand, a master was designated and its models transferred onto the second unit of its network and the two units of the second brand. Calibration models were transferable from brand to brand with similar or better precision than when each instrument was calibrated on its own calibration set (for Infratec 1229, the relative predictive determinant (RPD) increased in intra and inter-brand calibration transfer situations from 10.42 to 11.45 and 10.57, with DOP and EROS respectively). Performance of each method was different across parameters, instruments, and validation sets. DOP modifications on the determination of the difference matrix showed promising results while TOP and EROS extensions to include variability specifically present in certain crop years did not bring any beneficial effects.  相似文献

The influence of gas sorption on damage nucleation of a semicrystalline polymer during coupled diffuso‐mechanical monotonic loading in tension was investigated for the first time. Alpha‐poly(vinylidene fluoride) was mechanically strained under the presence of two chemically different gases, hydrogen and carbon dioxide, at a temperature of 30°C and a pressure up to 120 bar. The cavitation damage at the microstructure scale after gas decompression was assessed from a careful SEM and TEM micrographs analysis. Compared with SEM, TEM allowed quantifying the smallest cavities (ten‐nanometer size). If decompression in hydrogen appeared to have only a slight effect on cavitation however, carbon dioxide showed a detrimental effect on damage nucleation with the exhibition of a significant population of small cavities. On the other hand, tension under gas pressure is more sensitive to hydrogen than to carbon dioxide. Once interaction with gaseous carbon dioxide takes place the plasticizing effect is in competition with damage by cavitation. POLYM. ENG. We performed pyrolysis of SiCNAlH and SiCNYOH nanopowder precursors under a reactive atmosphere (Ar/NH3/H2) with various compositions of ammonia (NH3) and dihydrogen (H2) to diminish C content, which is deleterious for thermal stability and sintering of the powders. This paper continues a previous work on the fabrication of an Si3N4/SiC composite without free C by studying the effect of H2 on the C/N atomic ratio of the powder. We studied the influence of the nature of the gaseous mixture (Ar/NH3/H2) on the powder composition. Elemental analysis showed that the introduction of H2 in the pyrolysis atmosphere limited the decomposition of NH3 and allowed for control of the C/N ratio. This behaviour can be explained by the structural evolution observed by 29Si NMR spectrometry but also by Fourier transform infrared and Raman spectroscopy. An Si3N4/SiC composite, with traces of free C, was obtained after post-pyrolysis heat treatment of the powders synthesized with 10 wt.% of H2 and 25 wt.% NH3.  相似文献

Ultrafiltration permeate of whey protein tryptic hydrolyzate was processed by nanofiltration (NF) to obtain retentate (NFR) and permeate (NFP) that were then tested as inhibitors of Listeria, Staphylococcus aureus and Escherichia coli. NFR at 20 mg mL?1 was most effective as an inhibitor (P < 0.001); whereas E. coli was relatively resistant, the effect on Listeria and S. aureus was greater at 20 mg mL?1 than at 10 mg mL?1 (P < 0.01). Peptide analysis revealed that NFR was rich in anionic peptides over eight amino acid residues in length. The antibacterial activity of two anionic peptides (84–91 and 125–135) and a cationic peptide (36–42) derived from β-lactoglobulin was tested. Peptide 125–135 was more inhibitory (P < 0.05) than peptide 84–91 against Listeria monocytogenes and S. aureus; peptide 36–42 was not inhibitory. NFR appears to have potential as a natural bio-preservative.  相似文献

Eleven peptides, selected on the basis of physicochemical characteristics and their theoretical release from β-lactoglobulin (β-Lg) and α-lactalbumin (α-La) by trypsin or chymotrypsin, were chemically synthesised to evaluate their immunomodulating properties. Murine splenocyte proliferation in the absence and presence of mitogen and different peptide concentrations were measured after 72–96 h incubations. β-Lg f78–83 had no effect on proliferation; β-Lg f15–20, f55–60, f84–91, f92–105, f139–148, f142–148 and α-La f10–16 stimulated proliferation to different extents; β-Lg f1–8, f102–105 and α-La f104–108 showed a cytotoxic effect. Regression analysis revealed the relationship of positive charge, hydrophobicity and length to the stimulatory proliferative effect. β-Lg f15–20, f55–60 and f139–148 also induced various inhibiting and/or stimulating effects on cytokine secretion. The results confirm that peptides releasable by digestive enzymes from α-La and β-Lg have the potential to influence the specific immune response through the modulation of splenocyte proliferation and cytokine secretion.  相似文献
