Biotinylation of a protein generally involves chemical modification of a translated protein. Using this methodology, however, biotinylation at a specific position remains difficult. We investigated whether it would be possible to use an Escherichia coli initiator tRNA(fmet) aminoacylated with methionine biotinylated at the alpha-amino group to introduce a biotin tag specifically at the N terminus. We report here that a biotin tag could be incorporated into the green fluorescent protein (GFP) at the N-terminal site, in the presence of an E. coli initiator tRNA(fmet) aminoacylated with methionine biotinylated at the alpha-amino group. The biotinylated GFP was purified by simple monomeric streptavidin-agarose affinity column chromatography. Based on the total amount of GFP molecules, the purification yield and the biotin labelling efficiency of this system were approximately 7% and 10-20%, respectively, according to the densitometric analysis of Western blots. Judging from the results of a fluorescence imaging experiment, almost all the purified GFP molecules retained the native fluorescence activity. Importantly, the present results support the hypothesis that the E. coli initiator tRNA(fmet) aminoacylated with a relatively large substituent can be recognized by an E. coli ribosome and adequately placed at the P site to initiate translation.  相似文献

We present a simple design rule for diode-laser pumped quasi-three-level lasers by using the M2 factor. The validity of this model was demonstrated by diode-pumped Yb:YAG laser experiments. The maximum output power of 1.33 W and optical slope efficiency of 63% were obtained in a 400-μm Yb:YAG chip miniature laser. Using a 200-μm Yb:YAG chip, a 70% optical slope efficiency was reached. In a coupled-cavity configuration, with a quartz birefringent tuning filter, 8.2 THz (29 nm) of tuning was obtained at room temperature. By changing to a calcite birefringent filter, single-axial-mode oscillation with an output power of 500 mW was observed  相似文献

This paper deals with a motion control system for a space robot with a manipulator. Many motion controllers require the positions of the robot body and the manipulator hand with respect to an inertial coordinate system. In order to measure them, a visual sensor using a camera is frequently used. However, there are two difficulties in measuring them by means of a camera. The first one is that a camera is mounted on the robot body, and hence it is difficult to directly measure the position of the robot body by means of it. The second one is that the sampling period of a vision system with a general-purpose camera is much longer than that of a general servo system. In this paper, we develop an adaptive state observer that overcomes the two difficulties. In order to investigate its performance, we design a motion control system that is constructed by combining the observer with a PD control input, and then conduct numerical simulations for the control system. Simulation results demonstrate the effectiveness of the proposed observer.  相似文献

A knowledge-based approach to retrieve medical images by feature and content with spatial and temporal constructs is developed. Selected objects of interest in an image are segmented and contours are generated. Features and content are extracted and stored in a database. Knowledge about image features can be expressed as a type abstraction hierarchy (TAH), the high-level nodes of which represent the most general concepts. Traversing TAH nodes allows approximate matching by feature and content if an exact match is not available. TAHs can be generated automatically by clustering algorithms based on feature values in the databases and hence are scalable to large collections of image features. Since TAHs are generated based on user classes and applications, they are context- and user-sensitive. A knowledge-based semantic image model is proposed to represent the various aspects of an image object's characteristics. The model provides a mechanism for accessing and processing spatial, evolutionary and temporal queries. A knowledge-based spatial temporal query language (KSTL) has been developed that extends ODMG's OQL and supports approximate matching of features and content, conceptual terms and temporal logic predicates. Further, a visual query language has been developed that accepts point-click-and-drag visual iconic input on the screen that is then translated into KSTL. User models are introduced to provide default parameter values for specifying query conditions. We have implemented the KMeD (Knowledge-based Medical Database) system using these concepts  相似文献

We have studied the cooperative manipulation of floating object by several space robots. For these cooperative motions, we have reported that a tracking control method using the transpose of the generalized Jacobian matrix (GJM) can be utilized for robots with joint torque controllers. For cooperative motions by some robots with joint velocity controllers, we proposed a tracking control method using the transpose of the GJM. Simulation results show the effectiveness of the proposed control method.  相似文献

We introduce a semantic data model to capture the hierarchical, spatial, temporal, and evolutionary semantics of images in pictorial databases. This model mimics the user's conceptual view of the image content, providing the framework and guidelines for preprocessing to extract image features. Based on the model constructs, a spatial evolutionary query language (SEQL), which provides direct image object manipulation capabilities, is presented. With semantic information captured in the model, spatial evolutionary queries are answered efficiently. Using an object-oriented platform, a prototype medical-image management system was implemented at UCLA to demonstrate the feasibility of the proposed approach.  相似文献

During iron-carbon eutectic solidification, the coefficients for partition of a third element between the eutectic liquid and its solid were evaluated thermodynamically. The coefficientk M A/L for the equilibrium partition of the third element (M) between austenite and liquid iron largely depended on the interaction between carbon and the third element and a simplified method for the evaluation ofk M A/L was introduced. The coefficients,K S andK M, for the partition of the element between the eutectic liquid and its solid in the stable and metastable eutectic solidification, respectively, were also calculated fromk M A/L and the coefficientk M C/A for the equilibrium partition of the element between cementite and austenite. It was indicated by the thermodynamics of the free energy for the co-existing phases that the effect of a third element on graphitization occurring during eutectic solidification was related quantitatively to the value of K which was represented byK S-K M. The effect of a third element on the difference between the stable and metastable eutectic temperatures and on the carbon activity of liquid iron was closely related to K or the equilibrium partition coefficient,k M C/A .  相似文献

A knowledge based approach is introduced for retrieving images by content. It supports the answering of conceptual image queries involving similar-to predicates, spatial semantic operators, and references to conceptual terms. Interested objects in the images are represented by contours segmented from images. Image content such as shapes and spatial relationships are derived from object contours according to domain specific image knowledge. A three layered model is proposed for integrating image representations, extracted image features, and image semantics. With such a model, images can be retrieved based on the features and content specified in the queries. The knowledge based query processing is based on a query relaxation technique. The image features are classified by an automatic clustering algorithm and represented by Type Abstraction Hierarchies (TAHs) for knowledge based query processing. Since the features selected for TAH generation are based on context and user profile, and the TAHs can be generated automatically by a clustering algorithm from the feature database, our proposed image retrieval approach is scalable and context sensitive. Developers used in photolithography contain toxic tetramethylammoniumhydroxide (TMAH) and this creates a problem of how to properly treat developer wastewater. We have developed a TMAH wastewater treatment technique that consists of a combination of two novel decomposition processes: pyrolyzing TMAH to TMA and decomposing TMA to N 2, H2O, and CO2 by means of a selective oxidation catalyst for nitrogenous compounds. We have tested a system using this technique in long-term treatment of the actual wastewater and have found it to be sufficiently practical. The running cost of a treatment system using our technique would be about one-ninth that of disposing of the wastewater as industrial waste but about 2.3 times that of biological treatment. Compared with biological treatment, however, our system is tolerant to many treatment conditions and operation management is much easier.
