Activation of thymic B cells by signals of CD40 molecules plus interleukin-10

We have previously found that thymic B cells, particularly thymic CD5+ B cells, show low responsiveness to the usual B cell stimulants such as lipopolysaccharide or anti-IgM plus interleukin (IL)-4, although they proliferate and produce antibodies after direct interaction with major histocompatibility complex class II-restricted T blasts. These findings raise the possibility that a CD40-CD40 ligand (L) interaction is involved in the activation of thymic B cells. In the present study, we therefore examine this possibility using CD40L-transfected Chinese hamster ovary (CHO) cells or anti-CD40 monoclonal antibody (mAb). When B cells in the spleen and peritoneal cavity were stimulated, they proliferated and produced immunoglobulin (Ig) in the presence of CD40L-CHO cells or anti-CD40 mAb alone. However, another signal delivered by IL-10 in addition to CD40L-CHO cells or anti-CD40 mAb was found to be necessary for thymic B cells to proliferate and secrete Ig. Other interleukins acting on B cells, such as IL-4, IL-5, and IL-6, had no effect on the activation of thymic B cells, which thus have unique characteristics not found in peripheral B cells. This report discusses the physiological significance of IL-10- and CD40-driven signals in the activation of thymic B cells.     

Colony promoting activity (CPA) is a novel factor distinct from IL-6

The supernatant (CM) of long-term bone marrow culture (LTBMC) contains colony promoting activity (CPA) which does not have granulocyte-macrophage (GM) colony-stimulating activity but which enhances GM-colony formation in the presence of CSF. CPA is different from IL-1, IL-3 and GM, G-, and M-CSF. Since CPA-containing LTBMC-CM always contains a substantial level of IL-6, CPA was thought to be similar to IL-6. In the present study, we found that LTBMC with a particular batch of horse serum produced IL-6 without a corresponding production of CPA. Addition of IL-6 to GM-colony assay system in the presence of GM-CSF did not enhance the colony formation. LTBMC-CM did not stimulate proliferation nor differentiation of mast cell progenitors. Anti-IL-6 antibodies suppressed IL-6 activity, but not CPA. These results indicate that CPA is a novel factor distinct from IL-1, IL-3, G-, M-, GM-CSF, IL-6 and SCF (c-kit ligand).     

Requirement for transglutaminase in progesterone-induced decidualization of human endometrial stromal cells

Differentiation of endometrial stromal cells (decidualization) is essential for embryo implantation and maintenance of pregnancy. By sequential complementary DNA subtractive hybridization, one of the messenger RNAs (mRNA) induced by progesterone in human endometrial stromal cells decidualized in vitro was identified as that of a tissue transglutaminase type II (TGase). TGase mRNA was induced within 6 h after the addition of progesterone to the culture, and the effect was dose dependent. Both the TGase inhibitor monodansylcadaverine and oligodeoxynucleotide complementary to the TGase mRNA inhibited the decidualization, as assessed by PRL production and morphological transformation. Expression of TGase mRNA in human decidua and endometria exposed to high levels of progesterone in vivo was demonstrated by Northern blotting and in situ hybridization. These data suggest that TGase is necessary for the decidualization of human endometrial stromal cells and that clarification of the mechanism of action of TGase will facilitate further insight into the diagnosis and treatment of infertility.     

Interferon therapy after ablation of Kent bundle for a patient with chronic hepatitis type B complicated with WPW syndrome

Cardiotoxicity of interferon-alpha or gamma, such as fatal arrhythmia and myocardial infarction, has been reported. Therefore cardiotoxicity of interferon should be seriously considered before administration for patients with a pre-existing heart disease. We treated a patient with chronic active hepatitis type B, coexisted with Wolff-Parkinson-White syndrome, who has had frequent attacks of paroxysmal atrial fibrillation. To prevent the occurrence of fatal arrhythmia with an interferon therapy in this patient, we performed radiofrequency catheter ablation of the Kent bundle. After the successful ablation, we could safely administered recombinant interferon alpha-2b for chronic hepatitis type B.     

Oligosaccharide structures formed during the hydrolysis of lactose by Aspergillus oryzae β-galactosidase

Di-, tri-, tetra-, penta- and hexasaccharides were formed during the hydrolysis of lactose by transgalactosylation reaction of Aspergillus oryzae β-galactosidase. In this study the isolation and characterization of the major constituents of tri-, tetra- and pentasaccharides are described. The structure elucidation of 3 tri-, 2 tetra- and 1 pentasaccharides was carried out by methylation analysis, mass spectrometry and ¹³C-nmr spectrometry. The trisaccharides are $\text{O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 3)-O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 4)-}\text{d}\text{-}\text{glucose}\text{(3′-}\text{galactosyl-lactose}\text{)}$, $\text{O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 6)-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 4)-}\text{d}\text{-}\text{glucose}\text{(6′-}\text{galactosyl-lactose}\text{)}$ and $\text{O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 4)-O-[β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 6)]-}\text{d}\text{-}\text{glucose}\text{(4,6-}\text{digalactosyl-glucose}\text{)}$. Tetrasaccharides are $\text{O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 6)-O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 6)-O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 4)-}\text{d}\text{-}\text{glucose}$ and $\text{O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 6)-O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 3)}$ [or $\text{O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 3)-O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 6)]-O-β-}\text{d}\text{-}\text{galactopyranosyl}\text{-(1 → 4)-}\text{d}\text{-}\text{glucose}$. Breast carcinoma with myeloid metaplasia--a case report

A 49 year-old female had been suffering from primary myelofibrosis since February 1987 without receiving any treatment. In 1994, a breast mass was detected. Breast tumor biopsy revealed tubular carcinoma with intraductal components and multinucleated giant cells in the loose and myxoid stroma. The giant cells were thought to be megakaryocytes because both Factor VIII and platelet glycoprotein GP IIIa were detected in their cytoplasm. While additional mastectomy specimens and the axillary lymph nodes also revealed prominent myeloid metaplasia, there was no proliferation of the cancer cells. Granulocytic series stained for chloroacetate esterase and very few erythrocytic series were observed. This is the first case in which breast carcinoma and myeloid metaplasia coexisted in the same breast tumor.     

High performance 35 nm gate length CMOS with NO oxynitride gate dielectric and Ni salicide

The 35 nm gate length CMOS devices with oxynitride gate dielectric and Ni salicide have been fabricated to study the feasibility of higher performance operation. Nitrogen concentration in gate oxynitride was optimized to reduce gate current I/sub g/ and to prevent boron penetration in the pFET. The thermal budget in the middle of the line (MOL) process was reduced enough to realize shallower junction depth in the S/D extension regions and to suppress gate poly-Si depletion. Finally, the current drives of 676 /spl mu/A//spl mu/m in nFET and 272 /spl mu/A//spl mu/m in pFET at V/sub dd/=0.85 V (at I/sub off/=100 nA//spl mu/m) were achieved and they are the best values for 35 nm gate length CMOS reported to date.     

Robust H∞ performance for lineardelay-differential systems with time-varying uncertainties

The present paper concerns robust H∞ performance for linear delay-differential systems which involve an uncertain time delay and time-varying norm-bounded parameter uncertainties. Based on the Lyapunov functional, a simple criterion is proposed which assures the pseudo-quadratic stability as a well as an H_∞-norm bound. The criterion is given in the form of a linear matrix inequality which is affine or convex in additional scalar parameters. A simple criterion is presented to evaluate the extent of the performance robustness     

Analysis of ink flow in a mimeograph printer based on the mechanics of fluid-filled poroelastic solids

Summary To understand some main ink flow mechanisms in a mimeograph printer, the continuum mechanics theory of fluid-saturated porous solids is applied to analyze the ink flow through screens and a printing master, which are modeled by a fluid-filled poroelastic single layer. The layer is assumed to be pressed by a press roller from below and to be subjected from above to the hydrodynamic lubrication pressure induced between a printing drum and an ink roller. Nonlinear elasticity of the layer and dependence of the permeability on its deformation are taken into account. The numerical results show some important results: e.g., squeezing-out of the ink from the layer by the layer compaction by the press roller plays an important role, especially for the case where the compression duration is small in comparison with the characteristic time of the ink-filled poroelastic layer.
