A simple and sensitive method was developed to measure total hydroperoxides. After the reduction of hydroperoxides with triphenylphosphine
(TP) in cyclohexane at 30°C, the amount of triphenylphosphine oxide (TPO) produced is determined by reverse-phase or normal-phase
high-performance liquid chromatography (HPLC) in combination with an ultraviolet (UV)-detector by measuring absorption at
220 or 260 nm. TPO was shown to be gener-ated stoichiometrically by reduction of known amounts of either cumene hydroperoxide
or methyl 13-hydroper-oxyoctadecadienoate. For various lipids at low levels of oxidation, the peroxide values determined by
this method were in good agreement with those obtained by conventional iodometry. The detection limit of TPO in HPLC using
absorption at 220 nm was less than 10 pmol. Consequently, total hydroperoxides in lipids at levels corresponding to less than
a peroxide value of 1 can be estimated by the TP method on a 10-mg sample. 相似文献

For the rationalization of lightning protection design of distribution lines, it is important to clarify the behavior of distribution lines when direct or nearby lightning occurs. Because of the lower insulation level than for transmission lines, in studies on lightning protection design of distribution lines, not only direct lightning strokes but also induced voltages caused by nearby strokes must be taken into account. Thus, it is necessary to grasp the frequency of occurrence on lightning phenomena around distribution lines. For this aim, lightning phenomena on TEPCO's distribution lines in use had been continuously observed for 6 years (1996 to 2001). The observation was carried out in a composite way, using still cameras and sensor for acquisition of lightning surge waveform data. Through the observation, new phenomena about lightning performance on distribution lines in the field became apparent. In some cases, in spite of a direct strike to the line, flashover did not occur. This fact means that the distribution line has a certain level of lightning resistance. Moreover, it was confirmed that AC following current generated between both ends of insulator disappeared naturally. Activation of the natriuretic peptide system lowers blood pressure and causes the excretion of salt. Atrial natriuretic peptide and B-type natriuretic peptide are the humoral mediators of this effect; they act primarily by binding to membrane-bound natriuretic peptide receptor A (NPRA) and stimulating its intrinsic guanylate cyclase activity. To study whether genetically determined differences in NPRA expression affect blood pressure we have generated mice with one, two, three, or four copies of the gene encoding NPRA (Npr1 in the mouse). Atrial natriuretic peptide-dependent guanylate cyclase activity ranged progressively from approximately one-half normal in one-copy animals to twice normal in four-copy animals (P < 0.001). On different diets (0.05%, 2%, and 8% NaCl), the blood pressures of F1 male mice having only one copy of Npr1 averaged 9.1 mmHg (1 mmHg = 133 Pa) above those of wild-type two-copy males (P < 0.001), whereas males with three copies of the gene had blood pressures averaging 5.2 mmHg below normal (P < 0.01). The blood pressures of the one-copy F1 animals were significantly higher (by 6.2 mmHg; P < 0.01) on the high-salt than on the low-salt diet. The blood pressures of four-copy F3 males were significantly lower (by 7 mmHg; P < 0.05) on the high-salt than on the low-salt diet. These results demonstrate that below normal Npr1 expression leads to a salt-sensitive increase in blood pressure, whereas above normal Npr1 expression lowers blood pressures and protects against high dietary salt. 相似文献

We previously reported that the region corresponding to amino acids 197 to 216 of the gp46 surface glycoprotein (gp46-197) served as a binding domain for the interaction between gp46 and trypsin-sensitive membrane components of the target cell, leading to syncytium formation induced by human T-cell lymphotropic virus type 1 (HTLV-1)-bearing cells. Our new evidence shows that the 71-kDa heat shock cognate protein (HSC70) acts as a cellular receptor for syncytium formation. Using affinity chromatography with the peptide gp46-197, followed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, we isolated three components (bands A, B, and C) from MOLT-4 cell lysate which exhibited specific interactions with gp46 and inhibitory activities for syncytium formation induced by HTLV-1-bearing cells. Band A and B components were identified as HSC70 and beta-actin, respectively, through amino acid sequencing by tandem mass spectrometry and immunostaining with specific monoclonal antibodies. Band C is likely to be a nonprotein component, because full activity for syncytium formation was seen after extensive trypsin digestion. Anti-HSC70 monoclonal antibody clearly blocked syncytium formation in a coculture of HTLV-1-bearing cells and indicator cells, whereas no inhibition was seen with anti-beta-actin monoclonal antibody. Furthermore, flow cytometric analysis indicated that anti-HSC70 antibody reacted with MOLT-4 cells. Thus, we propose that HSC70 expressed on the target cell surface acts as a cellular acceptor to gp46 exposed on the HTLV-1-infected cell for syncytium formation, thereby leading to cell-to-cell transmission of HTLV-1. 相似文献

Megakaryocytes are unique haemopoietic cells which undergo DNA replication, giving rise to polyploid cells. However, little is known about the mechanism of megakaryocytic polyploidization. To address this issue, we used the human megakaryocytic cell line Meg-J. In the presence of K-252a (an indolocarbasole derivative), Meg-J cells stopped proliferation and exhibited additional megakaryocytic features, including morphological changes, polyploidization, and increases in the levels of surface expression of platelet glycoprotein (GP) IIb/IIa and GPIb. Thrombopoietin (TPO) promoted the K-2 52a-induced polyploidization and megakaryocytic differentiation. In the process of K-252a-induced polyploidization, levels of expression of both cdc2 and cyclin B1 were elevated transiently and subsequently decreased. This suggested that the polyploidization process in Meg-J cells was at least in part associated with a transient elevation and subsequent decrease in the expression of cdc2/cyclin B1 complex, a critical kinase involved in G2/M cell cycle transition. 相似文献

Nineteen children with neuroblastoma (aged 2 w.-7 y.o.) were studied to evaluate the optimal scan conditions for Iodine-123-Metaiodobenzylguanidine (MIBG) scintigraphy for accurate staging at the time of diagnosis. Six and 24 hours after an injection of 123I-MIBG, whole body image and truncal spot and SPECT images were obtained. Compared with other studies (CT or MRI and bone scintigraphy), each 123I-MIBG image was evaluated visually to investigate which image can demonstrate the extent of neuroblastoma most exactly. MIBG images demonstrated primary tumors in all patients, and metastatic lymphadenopathy in 8 of 9 patients. Twenty-four hour SPECT images gave us the most detailed information about the extent of abnormal accumulation. As to bone and bone marrow lesions, 6 hour images were superior to 24 hour images in detectability. Moreover, MIBG showed many more lesions and more extended accumulation than the bone scan. 123I-MIBG scintigraphy was very useful in detecting neuroblastomas. Using a traveling-wave model that can treat the spatial evolution of light fields, quantum noise characteristics of frequency-doubling ring lasers are analyzed under single-frequency operation. Systems under consideration are the discrete system, in which a laser crystal and a nonlinear crystal are separately installed, and the self-doubling system, which is equipped with a nonlinear crystal with laser gain. It is predicted that the unsaturated gain profile does not affect noise characteristics of the discrete system: nonlinear loss-induced noise enhancement (NINE), i.e., an increase of the spontaneous emission noise associated with enlarged gain that compensates the nonlinear loss, manifests itself at a specific operating point regardless of the gain profile. In the self-doubling system, on the other hand, the manifestation of NINE will arise for higher harmonic output compared with that in the discrete system. For the self-doubling system, moreover, the profile of the unsaturated gain causes a significant difference in NINE; because NINE is associated with a large field variation, it is effectively suppressed by configuring the gain profile to be flat or increasing along the direction of laser field propagation 相似文献
