ALG-2 is a penta-EF-hand Ca(2+)-binding protein and interacts with a variety of intracellular proteins. Two types of ALG-2-binding motifs have been determined: type 1, PXYPXnYP (X, variable; n = 4), in ALIX and PLSCR3; type 2, PXPGF, in Sec31A and PLSCR3. The previously solved X-ray crystal structure of the complex between ALG-2 and an ALIX peptide containing type 1 motif showed that the peptide binds to Pocket 1 and Pocket 2. Co-crystallization of ALG-2 and type 2 motif-containing peptides has not been successful. To gain insights into the molecular basis of type 2 motif recognition, we searched for a new hydrophobic cavity by computational algorithms using MetaPocket 2.0 based on 3D structures of ALG-2. The predicted hydrophobic pocket designated Pocket 3 fits with N-acetyl-ProAlaProGlyPhe-amide, a virtual penta-peptide derived from one of the two types of ALG-2-binding sites in PLSCR3 (type 2 motif), using the molecular docking software AutoDock Vina. We investigated effects of amino acid substitutions of the predicted binding sites on binding abilities by pulldown assays using glutathione-S-transferase -fused ALG-2 of wild-type and mutant proteins and lysates of cells expressing green fluorescent protein -fused PLSCR3 of wild-type and mutants. Substitution of either L52 with Ala or F148 with Ser of ALG-2 caused loss of binding abilities to PLSCR3 lacking type 1 motif but retained those to PLSCR3 lacking type 2 motif, strongly supporting the hypothesis that Pocket 3 is the binding site for type 2 motif.  相似文献

PES/Pebax and PEI/Pebax composite membranes were prepared by coating the porous PES and PEI substrate membranes with Pebax-1657. The morphology and performance of the prepared membranes were investigated by SEM and CO2 and CH4 permeation tests. The CO2 permeances of 28 and 52 GPU were achieved for PES/Pebax and PEI/Pebax composite membranes, respectively, with CO2/CH4 selectivities almost equal to that of Pebax (26). The experimental data were further subjected to a theoretical analysis using the resistance model. It was found that the porosity and the thickness of the dense section of PES substrate were an order of magnitude higher than those of PEI substitute. The porosity/thickness ratio of PEI substrate was, however, higher than PES, explaining the higher permeance of PEI/Pebax composite membrane. Substrates with porosities much higher than the Henis-Tripodi gas separation membrane were used in this work, aiming to achieve the selectivity of Pebax, rather than those of the substrate membrane materials.  相似文献

A chemical decomposition of polyurea (PUA) by hydrolysis under high pressure of carbon dioxide (CO2) was proposed. The hydrolysis of PUA was carried out at 190 °C for 2 h under 7.0 MPa of CO2 in the presence of water. The hydrolysis reaction gave white residual solid and water soluble compound. The white residual solid was characterized to be degraded PUA by FT-IR spectrum and elemental analysis. 1H-NMR spectra of the water soluble compound revealed that the hydrolysis of PUA produced diamine only from repeating unit of PUA, which was a component of PUA. This hydrolysis gave the corresponding diamine for quantitative yields.  相似文献

CO2 absorbents were prepared from polyethylene glycol and the polyamidines having N,N′-disubstituted amidine structure in the main chain synthesized through acid-catalyzed melt polycondensation of orthoesters and α,ω-diamines. The homogeneous binary mixtures with the polyamidines captured CO2 much more efficiently under CO2 flow than the one with polyethyleneimine. Furthermore, we investigated the CO2 capture and release by the binary mixtures in terms of effects of the volatility and the structure of polyamidines, temperature, and polyethylene glycol. Taking into consideration the results thus obtained, we conducted CO2 capture/release cycles with the CO2 capture step at 40 °C and with the CO2 releasing step at 80 °C in an alternating manner, thereby demonstrating the repeatability of CO2 capture and release by the binary system of the polyamidine and polyethylene glycol.  相似文献

Radiophotoluminescence phenomena have been widely investigated on various types of materials for dosimetry applications. We report that an aluminoborosilicate glass containing 0.005 mol% copper exhibits intense photoluminescence in the visible region induced by X-ray and γ-ray irradiation. The luminescence is assigned to the 3d94s1 → 3d10 transition of Cu+. The proportionality of the intensity of the induced photoluminescence to the irradiation dose was confirmed up to 0.5 kGy using 60Co γ-ray irradiation. Based on the spectroscopic results, a potential mechanism was proposed for the enhancement of the photoluminescence. The exposure to the ionizing radiation generates electron-hole pairs in the glass, and the electrons are subsequently captured by the Cu2+ ions, which are converted to Cu+ and emit the luminescence. For the glass containing 0.01 mol% copper, the pronounced enhancement of the photoluminescence was not observed because the reverse reaction, ie, the capture of the holes by the Cu+ ions, becomes prominent. The photoluminescence induced by the irradiation was stably observed for the glasses kept at room temperature and even for the glasses heat-treated at 150°C. However, the induced photoluminescence could be eliminated by the heat treatment at a temperature at 500°C, and the glass returned to the initial pre-irradiation state. The Cu-doped aluminoborosilicate glass is a potential candidate for use in dosimetry applications.  相似文献

Molecular dynamics simulations and dynamic mechanical analysis experiments were employed to investigate the mechanical behavior of metallic glasses subjected to iteration deformation in a nominally elastic region. It was found that cyclic deformation leads to the formation of irreversible shear transformation zones (STZs) and a permanent uniform strain. The initiation of STZs is directly correlated with the atomic heterogeneity of the metallic glass and the accumulated permanent strain has a linear relation with the number of STZs. This study reveals a new deformation mode and offers insights into the atomic mechanisms of STZ formation and low temperature uniform plastic deformation of metallic glasses.  相似文献

Phase-pure perovskite Pb(Zn x Mg1– x )1/3Nb2/3O3 solid solution (PZ x M1– x N) is obtained for x ≦ 0.7 by heating a milled stoichiometric mixture of PbO, Mg(OH)2, Nb2O5, and 2ZnCO3·3Zn(OH)2·H2O at 1100°C for 1 h. Percent perovskite ( f P) with respect to total crystalline phase decreases with increasing temperature of subsequent heating then increases to 900°C for the mixtures where x ≦ 0.8 and milled for 3 h. For mixtures with x = 0.9 and x = 1, f P decreases monotonically. Curie temperature increases almost linearly with increasing x up to x = 0.7. The maximum dielectric constant at 1 kHz is 2×104 and 1.7×104 for the mixture with x = 0.4 and x = 0.7, respectively. The stabilization mechanism of strained perovskite is discussed.  相似文献

Novel membranes based on sulfonated poly (phenylene oxide) (SPPO) was developed. SPPO membranes in the hydrogen form were converted to metal ion forms. The effect of exchange with metal ions including monovalent (Li+, Na+, K+), divalent (Mg2+, Ba2+, Ca2+) and trivalent (Al3+) ions was investigated in terms of permeation rate and permeation rate ratios for CO2 and CH4 gases. Both dense homogeneous membranes and thin‐film composite (TFC) membranes were studied for their gas separation characteristics. The effect of membrane preparation conditions and operating parameters on the membrane performance were also investigated. The selectivity of the TFC membrane increased as the cationic charge density increased as a result of electrostatic cross‐linking. TFC membrane of very high selectivity was achieved by coating a thin layer of SPPO‐Mg on a PES substrate. © 2000 John Wiley & Sons, Inc. J Appl Polym Sci 76: 735–742, 2000  相似文献
