Ozonation of a real red‐meat‐processing wastewater was conducted in a semi‐batch reactor to explore the possibility of the water reuse. The experimental results revealed that ozone was very effective in disinfection of the red‐meat‐processing wastewater. After 8 min of ozonation with an applied ozone dose of 23.09 mg min?1 liter?1 of wastewater, 99% of aerobic bacteria, total coliforms and Escherichia coli were inactivated. Empirical models were developed to predict the microbial inactivation efficacy of ozone from the CT values for the real red‐meat‐processing wastewater. A correlation was also derived to estimate the CT values from the applied ozone dose and the ozone contact time. The results also revealed that under the ozonation condition for 99% inactivation of aerobic bacteria, total coliforms and E coli, the decrease in the chemical oxygen demand and the 5‐day biological oxygen demand of the wastewater were 10.7% and 23.6%, respectively. However, ozonation under this condition neither improved the light transmission nor reduced the total suspended solids (TSS) despite of the decolorization of the wastewater after ozonation. Polymeric capsules with a thick shell made of biodegradable and biocompatible polymer and a liquid core of perfluorooctyl bromide (PFOB) were evaluated for stability as well as for ultrasound and magnetic resonance imaging (MRI) contrast enhancement. The method of preparation allows the mean capsule diameter to be regulated between 70 nm and 25 µm and the capsule thickness‐to‐radius ratio from 0.25 to 0.54. Capsule diameter remains stable at 37 °C in phosphate buffer for at least 4 and 6 h for nanocapsules and microcapsules, respectively. The in vitro ultrasound signal‐to‐noise ratio (SNR) was measured from 40 to 60 MHz for 6 µm and 150 nm capsules: the SNR increases with capsule concentration up to 20–25 mg mL−1, and then reaches a plateau that depends on capsule diameter (13.5 ± 1.5 dB for 6 µm and 6 ± 2 dB for the 150 nm capsules). The ultrasound SNR is stable for up to 20 min for microcapsules and for several hours for nanocapsules. For nanocapsules, the thinner the shell, the larger the SNR and the more compressible the capsules. Nanocapsule suspensions imaged in vitro with a commercial ultrasound imaging system (normal and tissue harmonic imaging modes, 7–14 MHz probe) were detected down to concentrations of 12.5 mg mL−1. Injections of nanocapsules (200 µg ml−1) in mice in vivo reveal that the initial bolus passage presents significant ultrasound enhancement of the blood pool during hepatic imaging (7–14 MHz probe, tissue harmonic imaging mode). 19F‐MRI images were obtained in vitro at 9.4T using spin‐echo and gradient echo sequences and allow detecting nanocapsules in suspension (50 mg mL−1). In conclusion, these results show initial feasibility for development of these capsules toward a dual‐modality contrast agent.  相似文献

Virus detection and enumeration has become increasingly important in fields ranging from medicine and biotechnology to environmental science. Although there are a wide variety of techniques that can be used to count viruses, there is demand for a rapid and more accurate means for virus enumeration. In this work, the performance of a flow cytometer that was designed and custom-built specifically for rapid detection of single viruses was evaluated. The instrument, designated a single nanometric particle enumerator (SNaPE), was characterized and calibrated using fluorescent polystyrene nanospheres. The reliability of the instrument with respect to virus enumeration was demonstrated for three medically relevant viruses, adenovirus-5, respiratory syncytial virus, and influenza A, treated with a fluorescent nucleotide stain. In each case, the SNaPE yielded a virus particle concentration consistent with, but slightly lower than, transmission electron microscopy (TEM) results, as expected. In addition, on the basis of calibration of signal intensity, the average peak height for a given virus was correlated with genome size, as expected. In contrast to time-consuming analyses such as TEM and plaque titers, SNaPE analysis of pure virus samples (including sample handling, data collection, and data processing) can be completed within 1 h.  相似文献

In order to provide the necessary phase equilibria data for understanding the development of the Widmanstatten pattern in iron meteorites, we have redetermined the Fe-Ni-P phase diagram from 0 to 100 pct Ni, 0 to 16.5 wt pct P, in the temperature range 1100° to 550°C. Long term heat treatments and 130 selected alloys were used. The electron microprobe was employed to measure the composition of the coexisting phases directly. We found that the fourphase reaction isotherm, where α+ liq ⇌ γ+ Ph, occurs at 1000° ± 5°C. Above this temperature the ternary fields α+ Ph + liq and α+ γ+ liq are stable and below 1000°C, the ternary fields ⇌+ γ + Ph and γ + Ph + liq are stable. Below 875°C a eutectic reaction, liq → γ + Ph, occurs at the Ni-P edge of the diagram. Altogether nineteen isotherms were determined in this study. The phase boundary compositions of the two-and three-phase fields are listed and are compared with the three binary diagrams. The α + γ + Ph field expands in area in each isotherm as the temperature decreases from 1000°C. Below 800°C the nickel content in all three phases increases with decreasing temperature. The phosphorus solubility in α and γ decreases from 2.7 and 1.4 wt pct at 1000°C to 0.25 and 0.08 wt pct at 550°C. The addition of phosphorus to binary Fe-Ni greatly affects the α/α + γ and γ/α + γ boundaries below 900°C. It stabilizes the α phase by increasing the solubility of nickel (α/α +γ boundary) and above 700°C, it decreases the stability field of the γ phase by decreasing the solubility of nickel(@#@ γ/α + γ boundary). However below 700°C, phosphorus reverses its role in γ and acts as a γ stabilizer, increasing the nickel solubility range. The addition of phosphorus to Fe-Ni caused significant changes in the nucleation and growth processes. Phosphorus contents of 0.1 wt pct or more allow the direct precipitation ofa from the parent γ phase by the reaction γ ⇌ α + γ. Examined the effect on sexual behavior of 57 male Sprague-Dawley rats of reduced olfactory system input to the preoptic area and medial forebrain bundle (MFB). Bilateral electrolytic lesions in the MFB just caudal to the preoptic area virtually abolished mating behavior. Electrolytic lesions or sham lesions placed bilaterally in the olfactory tubercle produced decrements in the mating behavior. There was no evidence of gonadotropic dysfunction in any of the groups, nor were there significant differences in their mean body weights at sacrifice. (22 ref.) The authors develop an efficient, recursive algorithm for determining the economic power dispatch of thermal generators within the unit commitment environment. The algorithm uses the equal incremental fuel cost criterion as its basis. In the algorithm, the fuel cost functions of the thermal generators are modeled by quadratic polynomials and the transmission losses are discounted. A method for incorporating the operation limits of the online generators and limits due to ramping generators is developed. The algorithm is amenable to computer implementation using the artificial intelligence programming language Prolog. The performance, of the algorithm was demonstrated through its application to the evaluation of the costs of dispatching 13 thermal generators within a generator schedule in a 24-h schedule horizon  相似文献

Because of its importance to cell function, the free-energy metabolism of the living cell is subtly and homeostatically controlled. Metabolic control analysis enables a quantitative determination of what controls the relevant fluxes. However, the original metabolic control analysis was developed for idealized metabolic systems, which were assumed to lack enzyme-enzyme association and direct metabolite transfer between enzymes (channelling). We here review the recently developed molecular control analysis, which makes it possible to study non-ideal (channelled, organized) systems quantitatively in terms of what controls the fluxes, concentrations, and transit times. We show that in real, non-ideal pathways, the central control laws, such as the summation theorem for flux control, are richer than in ideal systems: the sum of the control of the enzymes participating in a non-ideal pathway may well exceed one (the number expected in the ideal pathways), but may also drop to values below one. Precise expressions indicate how total control is determined by non-ideal phenomena such as ternary complex formation (two enzymes, one metabolite), and enzyme sequestration. The bacterial phosphotransferase system (PTS), which catalyses the uptake and concomitant phosphorylation of glucose (and also regulates catabolite repression) is analyzed as an experimental example of a non-ideal pathway. Here, the phosphoryl group is channelled between enzymes, which could increase the sum of the enzyme control coefficients to two, whereas the formation of ternary complexes could decrease the sum of the enzyme control coefficients to below one. Experimental studies have recently confirmed this identification, as well as theoretically predicted values for the total control. Macromolecular crowding was shown to be a major candidate for the factor that modulates the non-ideal behaviour of the PTS pathway and the sum of the enzyme control coefficients.  相似文献

Molecular dosimetry for polycyclic aromatic hydrocarbon-DNA adducts, genetic predisposition to cancer, and their interrelationships are under study in numerous laboratories. This report describes a modified 32P-postlabeling assay for the detection of polycyclic aromatic hydrocarbon-DNA adducts that uses immunoaffinity chromatography to enhance chemical specificity and quantitative reliability. The assay incorporates internal standards to determine direct molar ratios of adducts to unmodified nucleotides and to assess T4 polynucleotide kinase labeling efficiency. High performance liquid chromatography is used to assure adequacy of DNA enzymatic digestion. The assay was validated using radiolabeled benzo(a)pyrene-diol-epoxide modified DNA (r = 0.76, P < 0.05) thereby assessing all variables from enzymatic digestion to detection. Thirty-eight human lung samples were examined and adducts were detected in seven. A subset of samples also was examined for benzo(a)pyrene-diol-epoxide-DNA adducts by immunoaffinity chromatography, high performance liquid chromatography, and synchronous fluorescence spectroscopy. A high correlation between the two assays was found (P = 0.006). The lung samples were then analyzed by the polymerase chain reaction for the presence of mutations in the cytochrome P-450 (CYP) 1A1 and glutathione S-transferase mu (GST mu) genes. A positive association was identified for adduct levels and GST mu null genotypes (P = 0.038). No correlation was found between polycyclic aromatic hydrocarbon-adduct levels and CYP1A1 exon 7 mutations. Age, race, and serum cotinine were not related to adduct levels. Multivariate analysis indicated that only the GST mu genotype was associated with polycyclic aromatic hydrocarbon-DNA adduct levels. This work demonstrates that the 32P-postlabeling assay can be modified for chemically specific adduct detection and that it can be used in the assessment of potentially important genetic factors for cancer risk. The absence of a functional GST mu gene in humans is likely one such factor.  相似文献
