The acute resistance exercise (RE)-induced phosphorylation of mTOR-related signaling proteins in skeletal muscle can be blunted after repeated RE. The time frame in which the phosphorylation (p) of mTORS2448, p70S6kT421/S424, and rpS6S235/236 will be reduced during an RE training period in humans and whether progressive (PR) loading can counteract such a decline has not been described. (1) To enclose the time frame in which pmTORS2448, prpS6S235/236, and pp70S6kT421/S424 are acutely reduced after RE occurs during repeated RE. (2) To test whether PR will prevent that reduction compared to constant loading (CO) and (3) whether 10 days without RE may re-increase blunted signaling. Fourteen healthy males (24 ± 2.8 yrs.; 1.83 ± 0.1 cm; 79.3 ± 8.5 kg) were subjected to RE with either PR (n = 8) or CO (n = 6) loading. Subjects performed RE thrice per week, conducting three sets with 10–12 repetitions on a leg press and leg extension machine. Muscle biopsies were collected at rest (T0), 45 min after the first (T1), seventh (T7), 13th (T13), and 14th (X-T14) RE session. No differences were found between PR and CO for any parameter. Thus, the groups were combined, and the results show the merged values. prpS6S235/236 and pp70s6kT421/S424 were increased at T1, but were already reduced at T7 and up to T13 compared to T1. Ten days without RE re-increased prpS6S235/236 and pp70S6kT421/S424 at X-T14 to a level comparable to that of T1. pmTORS2448 was increased from T1 to X-T14 and did not decline over the training period. Single-fiber immunohistochemistry revealed a reduction in prpS6S235/236 in type I fibers from T1 to T13 and a re-increase at X-T14, which was more augmented in type II fibers at T13 (p < 0.05). The entity of myofibers revealed a high heterogeneity in the level of prpS6S235/236, possibly reflecting individual contraction-induced stress during RE. The type I and II myofiber diameter increased from T0 and T1 to T13 and X-T14 (p < 0.05) prpS6S235/236 and pp70s6kT421/S424 reflect RE-induced states of desensitization and re-sensitization in dependency on frequent loading by RE, but also by its cessation.  相似文献

In the present study verification is carried out of mathematical criteria that define the boundary between two hydrodynamic regimes: the gas continuous flow (GCF) regime and the pulsing flow (PF) regime. Attention is focused on the criterion derived using the model of K. Grosser, R.G. Carbonell and S. Sundaresan, AIChE J., 34 (1988) 1850–1860, and the parametric sensitivity of this criterion is analysed. Next, based on the measured values of the gas pressure drop in a reactor the verification of some selected parameters is carried out by fitting the theoretical and experimental boundary lines separating the GCF and PF regimes. Experimental results are also presented concerning the determination of parameters characterising the pulsing flow through the bed, namely the frequency of pulsations and the pulse structure. These parameters were determined for the system nitrogen–water, and also for liquids differing from water in their physicochemical properties.  相似文献

Implantation in all mammals involves shedding of the zona pellucida, followed by orientation, apposition, attachment and adhesion of the blastocyst to the endometrium. Endometrial invasion does not occur in domestic ruminants; thus, definitive implantation is achieved by adhesion of the mononuclear trophoblast cells to the endometrial lumenal epithelium (LE) and formation of syncytia by the fusion of trophoblast binucleate cells with the LE. This review highlights new information on mechanisms regulating the implantation cascade in sheep. The embryo enters the uterus on day 4 at the morula stage of development and then develops into a blastocyst by day 6. The blastocyst sheds the zona pellucida (day 8), elongates to a filamentous form (days 11-16), and adheres to the endometrial LE (day 16). Between days 14 and 16, the binucleate cells begin to differentiate in the trophoblast and subsequently migrate and fuse with the endometrial LE to form syncytia. Continuous exposure of the endometrium to progesterone in early pregnancy downregulates the progesterone receptors in the epithelia, a process which is associated with loss of the cell-surface mucin MUC1 and induction of several secreted adhesion proteins. Recurrent early pregnancy loss in the uterine gland knockout ewe model indicates that secretions of the endometrial epithelia have a physiologic role in blastocyst elongation and implantation. A number of endometrial proteins have been identified as potential regulators of blastocyst development and implantation in sheep, including glycosylated cell adhesion molecule 1 (GlyCAM-1), galectin-15, integrins and osteopontin. The epithelial derived secreted adhesion proteins (GlyCAM-1, galectin-15 and osteopontin) are expressed in a dynamic temporal and spatial manner and regulated by progesterone and/or interferon tau, which is the pregnancy recognition signal produced by the trophoblast during blastocyst elongation. The noninvasive and protracted nature of implantation in domestic animals provides valuable opportunities to investigate fundamental processes of implantation that are shared among all mammals. Understanding of the cellular and molecular signals that regulate uterine receptivity and implantation can be used to diagnose and identify causes of recurrent pregnancy loss and to improve pregnancy outcome in domestic animals and humans.  相似文献

The relationship describing the shock wave velocity was formulated for the trickle-bed reactor operating at periodically changed feeding the bed with liquid phase. The values of shock wave velocity calculated from derived equations were compared with experimental values obtained for both fast and slow mode of base–pulse periodic liquid feeding and using liquids differing in physicochemical properties. A good agreement between these two sets of values of shock wave velocity was obtained. It has to be emphasized that the relationship (Eq. (26)) derived in this study enables to estimate the values of the shock wave velocity when only mean values of variables of a process are known.  相似文献

The vitamin E (α-tocopherol) and free and bound malondialdehyde (MDA) in ventricular heart muscle and myocardial membrane from Wistar-Kyoto (W/K) normotensive and spontaneously hypertensive (SH) rats have been measured directly by high performance liquid chromatography (HPLC). Thiobarbituric acid-reactive substance (TBA-RS) in the myocardium and heart-muscle membrane of the two strains was also quantified by a colorimetric TBA test. It was found that SH-rat myocardium and myocardial membrane contained more than 3-fold less α-tocopherol than did heart muscle and cardiac membrane of the normotensive rat. Coincident with this relative vitamin E deficiency were several-fold greater amounts of MDA and TBA-RS in SH-rat myocardium and myocardial membrane. Most (87%) of the MDA in SH-rat heart muscle, but only 40% in W/K-rat heart muscle, was free (i.e., unbound). These results offer direct evidence that SH-rat myocardium is vitamin E-deficient and highly peroxidative, relative to cardiac muscle of the normotensive W/K parent strain. The lower vitamin E content of SH-rat myocardium is particularly striking, because SH-rat myocardial membrane was found to contain ∼35% more phospholipid than myocardial membrane in the W/K rat. Although the amounts of myocardial TBA-RS are greater in the SH strain, they do not reflect the actual MDA profiles of the heart muscles or the heart membranes and cannot be used as a quantitative index of cardiac oxidative-injury status due to non-MDA TBA-RS in both strains.  相似文献

In laboratory experiments we observe that ice particles (≤100 μm) entrained in a low pressure atmosphere (~1 mbar) get trapped by temperature gradients between three reservoirs at different temperature. Confining elements are a peltier element at 250 K (bottom), a liquid nitrogen reservoir at 77 K (top), and the surrounding vacuum chamber at 293 K. Particle levitation and trapping is modeled by an interplay of thermophoresis, photophoresis, and gravity. A number of ice particles are trapped simultaneously in close spatial distance to each other at least up to minutes and are accessible for further experiments.  相似文献

The process of injection‐molding net‐shape parts from thermotropic liquid‐crystalline polymers results in a skin‐core macrostructure. The underlying orientation in the core and the skin may differ both in magnitude and direction. A combination of near‐edge X‐ray absorption fine structure (NEXAFS) spectroscopy and two‐dimensional wide‐angle X‐ray scattering (2D WAXS) in transmission was used to characterize the orientation in injection‐molded plaques fabricated from thermotropic liquid‐crystalline copolyesters based on either 4,4′‐dihydroxy‐α‐methylstilbene or 6‐hydroxy‐2‐naphthoic acid/6‐hydroxybenzoic acid. NEXAFS is presented as a noninvasive in situ means of determining surface layer orientation that samples to a depth of as little as 2 nm and does not require slicing or ultramicrotoming of the samples. The effects of various processing conditions on the surface orientation in the region of the centerline of square injection‐molded plaques are presented and discussed. Comparisons are made between orientation parameters obtained by 2D WAXS in transmission, which is dominated by the microstructure in the core, and the NEXAFS technique. © 2005 Wiley Periodicals, Inc.
