The goal of this study was to determine the concentrations of Mg, Ca and K ions required for the formation of metabolically active population of phosphate (P)-accumulating bacterium Acinetobacter junii. The availability of Mg, Ca and K originating from natural minerals in the conditions of severe shortage of these cations was tested. In the case of shortage of Mg, Ca and K ions in wastewater the P removal was absent due to the decay of A. junii. In the cases of Mg or K shortage in wastewater the P removal was negligible due to the decay of A. junii, while Ca was not essential for the examined bacterium. The minimal required concentrations of Mg and K in synthetic wastewater were 0.64 mg Mg/mg P and 0.50 mg K/mg P. The natural zeolitized tuffs and bentonite, either in Mg, Ca or K form, successfully replaced the lack of Mg, Ca, K and trace metals in wastewater. In this research study, the applicability of polycaprolactone‐silicon dioxide and polycaprolactone‐zinc oxide coatings used as primers on papers for offset printing, was analysed. Polycaprolactone nanocomposites with different concentrations of nanoparticles were prepared and coated onto three types of papers: uncoated, low‐coated and gloss. The main aim was to analyse the influence of the nanoparticle concentration in primer on the optical and colorimetric properties of cyan and magenta offset printed samples. To further evaluate the applicability of the primers, the tensile strength of primed samples and their surface properties were analysed. Results show that prepared primers are primarily applicable on uncoated and low coated papers. Both nanoparticles in specific concentrations increased gloss values for printed samples on low‐coated paper, and the addition of both types of nanoparticles increased the rub resistance of printed samples. Furthermore, for each type of nanoparticle, the optimal concentration at which the colour deviation on print was minimised could be defined. Adding nanoparticles to primers did not significantly improve mottling on cyan‐printed samples. However, by increasing the concentration of nanoparticles in the primer by up to 1%, the mottling on magenta‐printed samples decreased. 相似文献

Chivosazoles A and F, isolated from Sorangium cellulosum, showed high antiproliferative activity with different mammalian cell lines including human cancer cells. The chivosazoles caused a delay in G2/M phase of the cell cycle, and treated cells often contained two nuclei. By labeling F‐actin it was shown that the actin cytoskeleton of the cells starts to break down after a few minutes of treatment. In vitro polymerization assays with purified G‐actin revealed that the chivosazoles inhibit actin polymerization and also cause a depolymerization of pyrene‐labeled F‐actin microfilaments prepared in vitro. Chivosazoles are new tools for the investigation of issues concerning the actin cytoskeleton and they have a different mode of action from the known microfilament‐disrupting compounds like rhizopodin and cytochalasin D. 相似文献

The aim of this study was to explore diversity and dynamic of indigenous LAB strains associated with a long ripened hard cheese produced from raw milk and undefined natural starter such as PDO Grana Padano cheese. Samples of milk, curd, natural whey culture and cheeses (2nd, 6th, 9th and 13th months of ripening) were collected from 6 cheese factories in northern Italy. DNA was extracted from each sample and from 194 LAB isolates. tRNAAla-23S rDNA-RFLP was applied to identify isolates. Strain diversity was assessed by (GTG)5 rep-PCR and RAPD(P1)-PCR. Finally, culture-independent LH-PCR (V1–V2 16S-rDNA), was considered to explore structure and dynamic of the microbiota. Grana Padano LAB were represented mainly by Lactobacillus rhamnosus, Lactobacillus casei, Lactobacillus paracasei, Lactobacillus delbrueckii, Lactobacillus helveticus and Pediococcus acidilactici, while the structure and dynamic of microbiota at different localities was specific. The strength of this work is to have focused the study on isolates coming from more than one cheese factories rather than a high number of isolates from one unique production. We provided a valuable insight into inter and intraspecies diversity of typical LAB strains during ripening of traditional PDO Grana Padano, contributing to the understanding of specific microbial ecosystem of this cheese. 相似文献

In patients suffering from hepatic failure, the brain is subject to defined morphological and functional changes known as hepatic encephalopathia (HE). The morphological changes are dominated by glial cells (Alzheimer-type II astrocytes). It has recently been possible to demonstrate, that the retinal glia (Müller) cells undergo similar morphological changes. The present study was carried out in order to reveal if these Müller cell changes cause any characteristic functional deficits. We examined 11 patients with different stages of HE due to liver cirrhosis. Six patients were at stage 0 or 1 (group I) and five at stage 2 or 3 (group II). They underwent ophthalmological routine examination, colour vision testing and standard ERG recording. None of the patients reported impaired vision, in daylight or at night. There were no fundus abnormalities except very mild changes of the pigment epithelium and abnormal reflexes of the inner limiting membrane, especially in the higher HE stages. The number of confusions in the colour arrangement test increased with the higher stages of HE, preferably in the tritan axis. The scotopic a- and b-waves of the electroretinogram (ERG) were almost unchanged in group I and significantly decreased and delayed in group II. The photopic ERG b-wave amplitudes were changed in a similar fashion. Oscillatory potentials proved to be most sensitive to hepatotoxic changes. Their latencies were significantly delayed even in group I. Amplitudes were decreased significantly only in group II. Patients suffering from hepatic failure and accompanying HE display functional abnormalities of the retina. These are best demonstrated by the ERG, and correlate well with the degree of HE. A hypothesis is presented that relates the observed functional changes to altered neurotransmitter levels and impaired retinal glial-neuronal interaction, due to Müller cell damage caused by elevated ammonia levels. 相似文献

The insertion of unconditioned latissimus dorsi muscle at the humerus was reattached in anesthetized goats to the first stage piston of a two stage mechanical-to-hydraulic energy convertor for a skeletal muscle powered ventricular assist device. To study a range of forces, pistons of different cross sectional areas were evaluated during thoracodorsal nerve stimulation. Each energy convertor piston was coupled hydraulically to an actuator on a Thoratec VAD (Thoratec Laboratories, Berkeley, CA) in a mock circulatory loop. Maximum force (70.1 +/- 10.8 N) was greatest for the largest piston, and stroke length (4.0 +/- 0.7 cm) was greatest for the smallest piston. However, maximum stroke work (1.2 +/- 0.5 J) and muscle powered VAD ejected stroke volume (45 +/- 17 ml) were greatest for the middle size piston. These results are consistent with a biomechanical model of whole muscle contraction that predicts that there is an optimum force that produces maximum cycle work. Thus, with a two stage energy convertor, by changing the ratio of the cross sectional areas of the energy convertor and muscle powered VAD actuator pistons, the effective mechanical advantage for the muscle can be optimized to produce more work output and muscle powered VAD flow. Skeletal muscle powered devices using such an energy convertor could provide completely implantable circulatory support free from batteries and other power conditioning hardware required with electromechanical systems. 相似文献

The distribution of mitochondria within retinal glial (Müller) cells and neurons was studied by electron microscopy, by confocal microscopy of a mitochondrial dye and by immunocytochemical demonstration of the mitochondrial enzyme GABA transaminase (GABA-T). We studied sections and enzymatically dissociated cells from adult vascularized (human, pig and rat) and avascular or pseudangiotic (guinea-pig and rabbit) mammalian retinae. The following main observations were made. (1) Müller cells in adult euangiotic (totally vascularized) retinae contain mitochondria throughout their length. (2) Müller cells from the periphery of avascular retinae display mitochondria only within the sclerad-most end of Müller cell processes. (3) Müller cells from the vascularized retinal rim around the optic nerve head in guinea-pigs contain mitochondria throughout their length. (4) Müller cells from the peripapillar myelinated region ('medullary rays') of the pseudangiotic rabbit retina contain mitochondria up to their soma. In living dissociated Müller cells from guinea-pig retina, there was no indication of low intracellular pH where the mitochondria were clustered. These data support the hypothesis that Müller cells display mitochondria only at locations of their cytoplasm where the local O2 pressure (pO2) exceeds a certain threshold. In contrast, retinal ganglion cells of guinea-pig and rabbit retinae display many mitochondria although the local pO2 in the inner (vitread) retinal layers has been reported to be extremely low. It is probable that the alignment of mitochondria and the expression of mitochondrial enzymes are regulated by different mechanisms in various types of retinal neurons and glial cells. 相似文献

BACKGROUND: The intranasal allergen Provocation test (INT) is the standard procedure to verify the effect of an allergen when a nasal allergy is suspected. It may be especially useful if results from skin tests or in vitro tests conflict with those from the history. In the clinical and research setting, INT may be used to study the efficacy of different treatment modalities such as pharmacotherapy or immunotherapy. The technique of INT has not been well standardized in the past. Variations exist in planning the optimum time for testing, especially in seasonal allergies. METHODS: We examined influences of different test dates with regard to the pollen season on the result of INT. A total of 19 patients (eight females, 11 males, aged 26.1 +/- 4.8 years) with a history of at least two years of allergic rhinitis to birch pollen, positive skin prick test, and RAST (> or = CAP class II) to birch pollen. INT was performed during the winter-period after approximately six months without natural pollen exposition (V1), during the birch pollen season (V2) and six weeks after the birch pollen season (V3). Allergens were applied using a pump spray (0.1 ml) in concentrations of 10, 50, 100, 500, 1000, 5000, 10,000, 50,000, and 100,000 SQ units per ml (SQ/ml). RESULTS: The threshold concentrations were significantly lower at V2 (100 SQ/ml) and V3 (500 SQ/ml) if compared to V1 (1000 SQ/ ml).
