Lingual antimicrobial peptide (LAP) belongs to the β-defensin family in cattle and is found in bovine milk. However, it is unclear whether LAP is involved in the early immune response to mammary infection. The aim of the study was to investigate the changes of LAP concentration in milk after intramammary challenge with lipopolysaccharide (LPS), the gram-negative bacteria cell membrane component, in dairy cows. Milk was collected before and after LPS or phosphate-buffered saline (control) challenge every hour for 12 h on d 0 and twice daily from d 1 to 7. Somatic cell count (SCC), LAP concentration, and lactoperoxidase (LPO) activity in the milk were measured. Somatic cell count started to increase at 2 h postchallenge and remained high until d 5 (694 ± 187 × 103 to >1,000 ± 0 × 103 cells/mL at d 0; >1,000 ± 0 × 103 cells/mL at d 1 to 3; 684 ± 194 × 103 to 829 ± 108 × 103 cells/mL at d 4; 527 ± 197 × 103 to 656 ± 145 × 103 cells/mL at d 5). Somatic cell count increased in the control cows, although the levels were lower compared with those in the LPS challenge group. The LAP concentration in milk increased significantly at 2 h post-LPS-challenge and was maintained at high levels until d 2 (8.6 ± 0.6 to 17.5 ± 2.3 nM). In the control cow infused with phosphate-buffered saline, there was no increase of LAP concentration in milk (5.1 ± 0.6 to 7.2 ± 0.8 nM). Increase of LPO activity in the milk was observed at 6 h after LPS challenge and continued until d 3 (4.7 ± 0.3 to 9.4 ± 1.1 U). No increase of LPO activity was observed in the milk of control cows. The increase and subsequent decrease in LAP concentration after LPS challenge occurred earlier than those of LPO activity. In multiparous cows with LPS infusion, there was a significantly negative relationship between the days leading to the basal levels in LAP concentration and LPO activity (r = −0.75). These results suggest that LPS induces secretion of LAP into milk within hours and that LPO may have a synergistic antimicrobial function with LAP in mammary glands of dairy cows.  相似文献

A single chromophore detection using video-microscopy is one of the latest methodologies to reveal unique characteristics, which could not be obtained from ensemble measurements. Among many kinds of subjects, dynamic optical properties observed in colloidal semiconductor nanoparticles are attractive and important not only for the basis of photo-physics but also for application studies, e.g. biological labeling, electronic devices. In this study, fluorescence video-microscopy was performed on cadmium selenide (CdSe) quantum dots (QDs) spin-coated on a glass substrate. From single CdSe QDs detection, emissions at wavelengths separated over 60 nm were observed for the first time. This spectral feature was attributed to the existence of double-emissive relaxation processes in CdSe QDs. Photoluminescence intermittency was also observed both from relaxation processes. Fluorescence video-microscopy, which was advanced in biology, can be applicable for the real-time monitoring of dynamic properties in semiconductor photo-physics.  相似文献

We studied the adsorption behavior of radioactive cesium (Cs) by the non-mica minerals kaolinite, halloysite, chlorite, montmorillonite, mordenite, MnO2, TiO2, Al2O3, and FeOOH to elucidate the environmental behavior of radioactive Cs fallout from the Fukushima Daiichi Nuclear Power Plant in the Tohoku region of Japan. The adsorption and desorption experiments of Cs on the minerals were carried out at the Cs concentrations 1 × 10?4, 1 × 10?5, and 2 × 10?9 mole L?1 at pH 5.5. The desorption of Cs from the minerals was examined using 0.1 mole L?1 LiCl, NaCl, KCl, RbCl, and CsCl solutions. The sequential desorption was examined using a 0.1 mole L?1 LiCl solution, a 1 mole L?1 KCl solution, and a 1 mole L?1 HCl solution. The distribution coefficient (K d) for the minerals at the Cs concentration 10?9 mole L?1 was in the order of mordenite > illite > montmorillonite, sericite, MnO2, kaolinite, and halloysite > chlorite, TiO2, Al2O3, and FeOOH, differing from the order observed at higher Cs concentrations. After the sequential desorption by the three reagent solutions, the residual fraction of Cs was higher at the Cs concentration 10?9 mole L?1 than at higher concentrations. Approximately 40%, 40%, 50%, and 25% of the adsorbed Cs were residual in montmorillonite, mordenite, MnO2, and kaolinite, respectively, after the sequential desorption. These results strongly suggest that (1) radioactive Cs at 10?9 mole L?1 is more strongly associated with the non-mica minerals than at higher concentrations of 1 × 10?4 and 1 × 10?5 mole L?1, and (2) the non-mica minerals montmorillonite, mordenite, kaolinite, and MnO2 contributed to the fixation of the radioactive Cs fallout on Fukushima soil.  相似文献

A technique was developed and tested to measure the local thickness of a droplet or a liquid film on a surface of an opaque or thick single transparent plate by an interference fringe pattern that was easily formed by reflecting laser lights. Monochromatic epi-illumination through an objective lens of a conventional microscope was provided by a 5 mW or 300 mW laser and a filter to remove the noise caused by laser speckle. The incremental height difference of the liquid layer between neighboring maxima or minima of fringes was evaluated from the wavelength of the laser light and the refractive index of the liquid. Estimation error of a local inclination angle was discussed using ray tracing under parallel illumination approximation. Droplet profiles evaluated from the interferogram that were obtained by the present fringe method agreed well with those by Laser Focus Displacement Meter. Measurement was made to ensure the usefulness of the present technique. It was made clear that the contact angle of a liquid droplet could be obtained precisely and swiftly, even in small size or small contact angle, and that instantaneous three-dimensional profile of a liquid film on a bubble moving in a microchannel could be measured. The fringe method had sufficient potential to obtain more detailed information about three-dimensional characteristics of liquid film in flows such as the generation, breakdown, and growth of waves and the liquid film on a bubble at the beginning of movement.  相似文献

Blast furnace water-cooled slag (BFWS) has been solidified hydrothermally with tobermorite formation. The experimental results showed that the addition of fly ash and quartz was favorable to the formation of tobermorite, and the strength development of solidified body depended on both of the tobermorite formation and filling degree of formed tobermorite in the spaces between BFWS particles. The fly ash added appeared to have a higher reactivity than the quartz used during the initial hydrothermal processing due to the higher solubility of glassy phase in fly ash. The tobermorite formation seemed to be very sensitive to the fly ash content, e.g., the addition of fly ash 10-20 mass% was favorable to tobermorite formation, while the excessive addition of fly ash (> 20 mass%) appeared to impede the tobermorite formation. The excessive addition of quartz was also shown to exert a negative effect on the tobermorite formation, which causes strength deduction.  相似文献

In this system, we introduced a trigger logic which can handle double-pulse events due to high count rate in future X-ray missions and several application on the ground experiments. We report on performance of 8-input superconducting quantum interference devices (SQUIDs) for multiplexing transition-edge sensor signals by using frequency-domain multiplexing. We found the typical critical current and the flux noise to be 17–19 μA and 0.7–1.1 μ , respectively. We also measured the crosstalk current between the input coils of the SQUIDs, and found that the mutual inductance was consistent with the design value, 800 pH. We confirmed that the cross talk current due to the mutual inductance was reduced by the flux-locked-loop (FLL) feedback, and its reduction rate was consistent with 1/(1+ℒ), where ℒ is the FLL feedback gain. We also show the result of 2-channel DC-driven TES signals readout using the 8-input SQUIDs.   相似文献

Tetrachloroethylene (PCE) is one of the major pollutants and is degraded by dissimilation by dehalorespiring bacteria. The dehalorespiring bacteria are anaerobic, and most cannot be cultured by conventional agar plating methods. Therefore, to identify the dehalorespiring bacteria that dissimilatively degrade PCE, a cultivation-independent method is required. To achieve accurate and detailed analysis of the bacteria, we developed a novel stable isotope probing (SIP) method. This technique involves 2 steps, namely, a labeling step, in which a labeled carbon source is incorporated into the sample's DNA, and an analysis step, in which the DNA is isolated, fractionated, and analyzed by polymerase chain reaction denaturing gradient gel electrophoresis (PCR-DGGE). Subsequently, 16S rRNA sequencing and phylogenetic analysis were performed to identify the bacteria. Initially, we examined the effectiveness of this method by using Dehalococcoides ethenogenes 195 consortium as a defined model system. The result indicated the method was able to correctly identify the dehalorespiring bacteria D. ethenogenes 195 from the consortium. Moreover, in an artificially contaminated microcosm experiment, we confirmed that the method was able to identify the indigenous dehalorespiring bacteria Dehalobacter sp. Thus, we concluded that this novel method was a feasible tool to identify dehalorespiring bacteria in natural environments.  相似文献
