The presence of multiple microcracks in a structural component causes material degradation such as reduction in the stiffness or reduction in the fracture toughness of the component. In this paper, the homogenization method is used to evaluate mechanical properties of the damaged material. The adaptation of the superposition method to the homogenization method is also presented. The proposed method makes use of the finite element solution of uncracked solid and the analytical solution. The effective elastic moduli of damaged materials containing lattice-distribution microcracks are estimated by the proposed method. Furthermore, the stress fields and the stress intensity factors of the elliptical microcracks in the damaged material at a micro-mechanics scale are evaluated to illustrate microscopic behavior such as crack interaction. 相似文献

First, the near-tip stress and electric displacement fields are analytically solved for a dynamically propagating interfacial crack in a piezoelectric bimaterial. Second, from the rate formulation of the energy balance in a piezoelectric material, the path independent dynamic J integral is derived, which has the physical significance of the energy release rate. Using the present near-tip analytical solutions, the relationships between the dynamic J integral and the stress and electric displacement intensity factors are also obtained. It is shown that the path independent dynamic J integral contains the static J integral and the dynamic J integral for elastic materials, and static J integral for piezoelectric materials as special cases. Third, for an interfacial crack in a piezoelectric bimaterial, the path independent separated dynamic J integrals are derived, which have the physical significance of energy flow rates into the propagating interfacial crack tip from the individual material sides or, equivalently, the separated dynamic energy release rates. Fourth, to accurately evaluate mixed-mode stress and electric displacement intensity factors, the component separation method of the dynamic J integral is developed. Finally, the finite element analyses of a static stationary interfacial crack in a piezoelectric bimaterial subject to mechanical, electrical and combined loading are carried out to demonstrate the applicability of the generalized (dynamic) J integral and the separated J integral, and the component separation method. 相似文献

Intensity fluctuations of the longitudinal modes of a 0.8 μm AlGaAs laser were precisely measured during the occurrence of hopping between two modes. It was found from this result that mode hopping follows the stochastics of a Poisson process. The frequency of mode hopping was measured asf_{c} = [exp [-95(I/I_{th} - 1)]] times 10^{7} (Hz). whereI/I_{th} is the injection current normalized to its threshold value. Results of analog computer simulations showed that spontaneous emission worked as a triggering force for mode hopping. Results of the analysis based on the Fokker-Planck equation were compared to the experimental results, from which the root-mean-square value of the fluctuating electric field of spontaneous emission was estimated as2.3 times 10^{2) (V/m)leqlanglesim{E}_{N} leq 3.2 times 10^{2} (V/m). Plugs, i.e. droplets formed in a microchannel, may revolutionize microfluidic cell-based assays. This study describes a microdevice that handles nanolitre-scale liquid plugs for the preparation of various culture setups and subsequent cellular assays. An important feature of this mode of liquid operation is that the recirculation flow generated inside the plug promotes the rapid mixing of different solutions after plugs are merged, and it keeps cell suspensions homogeneous. Thus, serial dilutions of reagents and cell suspensions with different cell densities and cell types were rapidly performed using nanolitres of solution. Cells seeded through the plug processing grew well in the microdevice, and subsequent plug processing was used to detect the glucose consumption of cells and cellular responses to anticancer agents. The plug-based microdevice may provide a useful platform for cell-based assay systems in various fields, including fundamental cell biology and drug screening applications. 相似文献

OBJECTIVE: To estimate the prevalence of smoking and the attitudes towards the restriction of smoking at work among female nurses in the national hospitals in Japan. DESIGN: Questionnaires mailed to 14 randomly selected national hospitals and sanitariums in Japan in 1993. SUBJECTS: 2207 female nurses. MAIN OUTCOME MEASURES: Smoking status and history, and attitudes towards the restriction of smoking at work. RESULTS: The prevalence of smoking among female nurses was 18.6%, which was higher than the age-adjusted prevalence of the general female population using this study's subjects as a standardised population. Banning smoking in the hospital in which they worked was supported by 15.0%, whereas 81.6% supported the restriction of smoking. CONCLUSIONS: The results showed that smoking is more common among female nurses than among the general female adult population. The survey suggested that nurses favour restriction, but not banning, of smoking. 相似文献

The effect of the drying rate of the solvent on microstructure evolution in thin films of immiscible polystyrene/polycarbonate blends was investigated by applying a stepwise change in air velocity. The polymer blend was dissolved in tetrahydrofuran and cast on a glass substrate to form a thin liquid film. The drying rate of the solvent was measured by a gravimetric technique, and the phase-separated structure in the film was observed by optical microscopy. The results indicated that cellular structures of droplet arrays were formed in the phases evolved primarily during the early stage of drying, and the droplets then coalesced in the intermediate stage to form bigger droplets. The size of the first evolved phase decreased with increasing time at a high drying rate, whereas larger droplets were obtained with longer high drying rate regimes. These results suggested that the droplets were induced by secondary phase separation which was strongly affected by the drying rate of the solvent. 相似文献

Effects of the antiosteoblastogenesis factor Semaphorin 4D (Sema4D), expressed by thrombin-activated platelets (TPs), on osteoblastogenesis, as well as osteoclastogenesis, were investigated in vitro. Intact platelets released both Sema4D and IGF-1. However, in response to stimulation with thrombin, platelets upregulated the release of Sema4D, but not IGF-1. Anti-Sema4D-neutralizing monoclonal antibody (mAb) upregulated TP-mediated osteoblastogenesis in MC3T3-E1 osteoblast precursors. MC3T3-E1 cells exposed to TPs induced phosphorylation of Akt and ERK further upregulated by the addition of anti-sema4D-mAb, suggesting the suppressive effects of TP-expressing Sema4D on osteoblastogenesis. On the other hand, TPs promoted RANKL-mediated osteoclastogenesis in the primary culture of bone-marrow-derived mononuclear cells (BMMCs). Among the known three receptors of Sema4D, including Plexin B1, Plexin B2 and CD72, little Plexin B2 was detected, and no Plexin B1 was detected, but a high level of CD72 mRNA was detected in RANKL-stimulated BMMCs by qPCR. Both anti-Sema4D-mAb and anti-CD72-mAb suppressed RANKL-induced osteoclast formation and bone resorptive activity, suggesting that Sema4D released by TPs promotes osteoclastogenesis via ligation to a CD72 receptor. This study demonstrated that Sema4D released by TPs suppresses osteogenic activity and promotes osteoclastogenesis, suggesting the novel property of platelets in bone-remodeling processes. 相似文献

The effects of pear vinegar (PV), which was specially brewed for enhanced galacturonic acid content, on the DSS-induced ulcerative colitis (UC) mouse model were evaluated. PV improved clinical symptoms, colon inflammation, and histological tissue injury in the DSS-induced acute UC mouse model. Moreover, PV suppressed inflammation due to acute UC by suppressing the myeloperoxidase (MPO)-mediated activation of inflammatory cells such as leukocytes and decreasing the serum concentration of IL-6. Our results demonstrated the protective action of PV in the DSS-induced acute UC mouse model. On the other hand, commercial apple vinegar did not show a protective effect in the DSS-induced acute UC mouse model. The structures of the E277A isomaltase mutant from Saccharomyces cerevisiae in complex with isomaltose or maltose were determined at resolutions of 1.80 and 1.40 Å, respectively. The root mean square deviations between the corresponding main-chain atoms of free isomaltase and the E277Α-isomaltose complex structures and those of free isomaltase and the E277A-maltose complex structures were found to be 0.131 Å and 0.083 Å, respectively. Thus, the amino acid substitution and ligand binding do not affect the overall structure of isomaltase. In the E277A-isomaltose structure, the bound isomaltose was readily identified by electron densities in the active site pocket; however, the reducing end of maltose was not observed in the E277A-maltose structure. The superposition of maltose onto the E277A-maltose structure revealed that the reducing end of maltose cannot bind to the subsite + 1 due to the steric hindrance from Val216 and Gln279. The amino acid sequence comparisons with α-glucosidases showed that a bulky hydrophobic amino acid residue is conserved at the position of Val216 in α-1,6-glucosidic linkage hydrolyzing enzymes. Similarly, a bulky amino acid residue is conserved at the position of Gln279 in α-1,6-glucosidic linkage-only hydrolyzing α-glucosidases. Ala, Gly, or Asn residues were located at the position of α-1,4-glucosidic linkage hydrolyzing α-glucosidases. Two isomaltase mutant enzymes – V216T and Q279A – hydrolyzed maltose. Thus, the amino acid residues at these positions may be largely responsible for determining the substrate specificity of α-glucosidases. 相似文献
