Signals generated by T cell receptor (TCR) cross-linking (or phorbol 12-myristate-13-acetate + Ca2+ ionophore), glucocorticoids or ionizing radiation all stimulate apoptotic cell death in thymocytes by signals that are initially distinct from each other. However, when these stimuli were administered to thymocyte cultures that were maintained under an atmosphere containing less than 20 ppm oxygen as opposed to one that contained 18.5% molecular oxygen, cell death was inhibited or abrogated, suggesting that the induction of death by all three different stimuli depend on the presence of molecular oxygen. Studies of the effects of the cysteine analog N-acetyl cysteine (NAC) with normal thymocytes demonstrated that this antioxidant inhibited the induction of death by each of the different stimuli in a manner the paralleled anaerobiosis. Furthermore, studies with thymocytes demonstrated that the induction of nur77, a gene shown to be involved in thymocyte apoptosis signaled through the TCR or its surrogates, is not inhibited by NAC or dependent upon molecular oxygen. The possible implications for negative selection of NAC-mediated inhibition of TCR-signaled thymocyte cell death was examined in thymic organ culture. Treatment of these cultures with NAC provided significant protection against staphylococcal enterotoxin B-mediated deletion of V beta 8-expressing thymocytes.  相似文献

Arthrobacter spp. are very widely distributed in the environment (e.g., soil) but have not been described as causing disease in humans. Over a 6-year period, two reference laboratories isolated or received 11 strains which were eventually identified as belonging to the genus Arthrobacter. These strains had been initially identified as Centers for Disease Control and Prevention coryneform group B-1 and B-3 bacteria (whitishgrayish colonies of 2 mm or greater in diameter after 24 h of incubation, respiratory metabolism, absent or weak acid production from sugars, and hydrolysis of gelatin). However, chemotaxonomic investigations revealed lysine as the diamino acid of the cell wall and the presence of branched cellular fatty acids (with anteiso-pentadecanoic acid predominating) which was compatible with an assignment of the 11 isolates to the genus Arthrobacter only. Peptidoglycan and 16S rRNA gene sequence analyses demonstrated that three of the strains studied were representatives of a new Arthrobacter species for which the name Arthrobacter cumminsii sp. nov. is proposed and that one other strain represented a second new Arthrobacter species for which the name Arthrobacter woluwensis sp. nov. is proposed. To assess the role of genetic changes in sensitivity to leptin hormone in contributing to responses to long-term selection for fatness, leptin was administered to a long-term fat selected (F) and a control line (C) of mice. These lines differ almost three fold in their percentage of fat (fat%) at about 15 weeks of age. Treated (T) animals received twice-daily intraperitoneal injections of 5mg/kg leptin from 91 to 105 days of age; untreated (U) animals received equivolume injections of phosphate-buffered saline. Treated compared with untreated animals in both lines had significantly (P < 0.05) lower mean body weight, food intake and fatness at the end of test (fat%: CT 3%, CU 7.4%, FT 14.9%, FU 21.1%). The differences in response between the lines [(CT-CU)-(FT-FU)] were all non-significant (P > 0.05), however. There was a very wide range of fatness (estimated from dry matter content) among FT animals (3-29%), much higher than in FU (15-31%), CT (0.7-6.4%) and CU (2-15%) animals. While sensitivity to leptin remains in the fat line, response appears to vary among animals at the dose level used.  相似文献
