This study was conducted to determine the effects of cultivar, tissue susceptibility and storage temperature on the keeping
quality of minimally processed Baby and Romaine lettuces. Midrib and photosynthetic tissues were lightly processed and stored
for up to 7 days at 5 °C and 13 °C. Changes in L
* and a
* values and absorbance at 430 nm were compared. The relationship between polyphenols, phenylalanine ammonia lyase (PAL) and
polyphenol oxidase (PPO) activities and browning was examined. Measurements of L
* and a
* values on midribs showed that browning discoloration was the same for Baby as for Romaine lettuces and there was no relationship
for temperature. In addition, L
* and a
* values in photosynthetic tissue indicated a decrease in green pigmentation during storage, especially at 13 °C. For both
kinds of tissue at the end of cold storage an increase in soluble brown polymers was detected. In midribs, total phenolics
increased significantly throughout the storage period because of the tissue-wounding response. The photosynthetic tissue had
a higher phenolic content than the midrib one. For both kinds of tissue an increase in PPO activity occurred throughout cold
storage. On the other hand, PAL activity in midrib tissue only increased initially, followed by a slow decline to reach normal
levels. Browning potential estimated by L
* values correlated significantly (P>95%) with PPO activity and with absorbance at 430 nm for Romaine lettuce. Based on colour and browning potential no differences
between Romaine and Baby lettuce cultivars were observed. However, photosynthetic tissue was the most suitable tissue for
the preparation of minimally processed salad mixes because of its high phenolic content.

The aim of the work was to study the effects of processing on the unsaponifiable matter, sterols and fatty and triterpenic alcohol in ripe olive fat (Manzanilla and Hojiblanca cultivars) and to disclose the most influential factors using GLM, PCA and DA. There were significant effects of cultivars or ps on unsaponifiable matter, β-sitosterol, Δ5-avenasterol, total sterols, 1-docosanol, 1-tetracosanol (ps), erythrodiol and percentage erythrodiol + uvaol. The values of most of these parameters were within the limits established by the EU Directives for olive and pomace oils but classification of the respective oils was not conclusive. Predictive discriminant analysis using these variables permitted 100% success in the classification according to cultivars and ps (68% in the case of cross validation). Results revealed that some influential steps should be re-designed, particularly the storage phase, in order to minimise changes in the studied fat components during ripe olive processing. 相似文献

Identification of PIR/CIS3 gene was carried out by amino-terminal sequencing of a protein band released by beta-mercaptoethanol (beta-ME) from S. cerevisiae mnn9 cell walls. The protein was released also by digestion with beta-1,3-glucanases (laminarinase or zymolyase) or by mild alkaline solutions. Deletion of the two carboxyterminal Cys residues (Cys(214)-12aa-Cys(227)-COOH), reduced but did not eliminate incorporation of Pir4 (protein with internal repeats) by disulphide bridges. Similarly, site-directed mutation of two other cysteine amino acids (Cys(130)Ser or Cys(197)Ser) failed to block incorporation of Pir4; the second mutation produced the appearance of Kex2-unprocessed Pir4. Therefore, it seems that deletion or mutation of individual cysteine molecules does not seem enough to inhibit incorporation of Pir4 by disulphide bridges. In fks1Delta and gsc2/fks2Delta cells, defective in beta-1,3-glucan synthesis, modification of the protein pattern found in the supernatant of the growth medium, as well as the material released by beta-ME or laminarinase, was evident. However, incorporation of Pir4 by both disulphide bridges and to the beta-1,3-glucan of the cell wall continued. Deletion of the repetitive sequence (QIGDGQVQA) resulted in the secretion and incorporation by disulphide bridges of Pir4 in reduced amounts together with substantial quantities of the Kex2-unprocessed Pir4 form. Pir4 failed to be incorporated in alkali-sensitive linkages involving beta-1,3-glucan when the first repetitive sequence was deleted. Therefore, this suggests that this sequence is needed in binding Pir4 to the beta-1,3-glucan. 相似文献

An experimental investigation was carried out for the first time on the triterpenic acids in table olives. Maslinic acid was found in a higher concentration than oleanolic acid in the flesh of 17, unprocessed olive varieties, with the Picual and the Manzanilla varieties showing the highest and almost the lowest contents, respectively. The level of triterpenic acids in several types of commercial black and green olives ranged from 460 to 1470 mg/kg fruit, which represents a much higher value than reported for virgin olive oils. In fact, the NaOH treatment employed to debitter black and green olives reduced the concentrations of these substances in the flesh because of their solubilisation into alkaline solutions. Thus, natural black olives, which are not treated with NaOH, showed a higher concentration than 2000 mg/kg in the olive flesh. These results will contribute to the reevaluation of table olives from a nutritional and functional point of view because of the promising bioactivity properties attributed to olive triterpenic acids. 相似文献

Red wine has been reported to exert beneficial effects in preventing cardiovascular diseases probably due to their polyphenols
constituents. The vasorelaxant capacity and phenolic composition of four monovarietal young red wines Merlot, Tempranillo,
Garnacha and Cabernet-Sauvignon obtained from grapes from 2004 vintage, cultivated under the same climatic and agricultural
conditions, wine-making technology and storage conditions, were determined using vascular reactivity assay in rat aortic and
high-performance liquid chromatography, respectively. Primary cultures of vascular smooth muscle cells from Wistar rats were
also used to evaluate their survival prevention activity as well. Every studied wine present vasorelaxation effect, but the
higher value corresponds to the analyzed Merlot wine, especially rich in phenolic compounds, mainly catechins and oligomeric
proanthocyanidins and anthocyanin glycosides. This study further demonstrated previous investigations about the phenolic composition

Two ready-to-eat crawfish processing plants were monitored for 2 years to study the impact of Listeria control strategies, including employee training and targeted sanitation procedures, on Listeria contamination. Environmental, raw material, and finished product samples were collected weekly during the main processing months (April to June) and tested for Listeria spp. and Listeria monocytogenes. Before implementation of control strategies (year 1), the two processing plants showed Listeria spp. prevalences of 29.5% (n = 78) in raw, whole crawfish, 5.2% (n = 155) in the processing plant environment, and 0% (n = 78) in finished products. In year 2, after plant-specific Listeria control strategies were implemented, Listeria spp. prevalence increased in raw crawfish (57.5%, n = 101), in the processing plant environment (10.8%, n = 204), and in the finished product (1.0%, n = 102). Statistical analysis showed a significant increase in Listeria spp. prevalence (P < 0.0001) and a borderline nonsignificant increase in L. monocytogenes prevalence (P = 0.097) on raw material in year 2. Borderline nonsignificant increases were also observed for Listeria spp. prevalence in environmental samples (P = 0.082). Our data showed that Listeria spp. prevalence in raw crawfish can vary significantly among seasons. However, the increased contamination prevalence for raw materials only resulted in a limited Listeria prevalence increase for the processing plant environment with extremely low levels of finished product contamination. Heat treatment of raw materials combined with Listeria control strategies to prevent cross-contamination thus appears to be effective in achieving low levels of finished product contamination, even with Listeria spp. prevalences for raw crawfish of more than 50%. 相似文献

ABSTRACTOccupancy patterns are necessary to estimate energy demand and evaluate thermal comfort in households. Because of this, many European countries are developing representative domestic schedules to replace outdated criteria. This paper evaluates the state of knowledge of UK domestic occupancy patterns and develops new domestic occupancy profiles for England. The presented research (1) characterizes methods for collecting occupancy data and inferring patterns; (2) identifies and assesses the quality of categories of occupancy patterns used in building simulation; and (3) develops updated occupancy profiles. A systematic scoping review identified social and monitoring surveys as the most deployed data-collection methods. A systematic literature review also established that the occupancy categories most frequently used in UK building simulation are (a) a family with dependent children where the parents work full time; and (b) a retired elderly couple who spend most of their time indoors. The interview sample from the English Housing Survey 2014–15 was used to map household typologies. Results show that categories (a) and (b) combined amount to only 19% of England’s households, which suggest models are over-reliant on these groups. Considering this result, the paper develops occupancy patterns for England derived from 2015 UK Time Use Survey diaries for each household typology previously identified. 相似文献

The nucleotide sequence of a fragment from the left arm of Saccharomyces cerevisiae chromosome VII has been determined. Analysis of the 14,607 bp DNA segment reveals nine open reading frames (ORFs) longer than 300 bp. G2827 is the SEC 7 gene, an essential coatomer complex subunit. G2834 encodes SSM1b, a ribosomal protein. The G2838 product shows homology to hypothetical yeast proteins, YIF0 and YE09, of unknown function. The G2830 product shows homology with the cell division protein FtsJ from Escherichia coli, with two hypothetical proteins from yeast, YCF4 and YBR1, and with R74.7, a hypothetical protein from Caenorhabditis elegans. Two of the ORFs are completely internal to longer ones and a third is partially embedded in G2850. The remaining ORFs give no significant homology with proteins in the databases. We present the results of a non-destructive measuring method allowing us to characterize the evolution of solder joints during thermal cycling ageing tests. The method uses a high resolution optical probe to detect selectively pure Joule and Peltier thermal responses of the solder joint subject to a given current pulse. The results show the Peltier and Joule responses to be good indicators for the evaluation of the age and the degradation of solder joints. 相似文献

Methylmercury (MeHg) is one of the most dangerous toxic pollutants spread throughout the earth. Chronic MeHg intoxication by contaminated food ingestion is the most common threat to human health, including impairment to the developing fetus. The present study aims at investigating the effects of maternal exposure to MeHg during gestation and lactation on the spinal cord of offspring. Pregnant rats received oral doses of MeHg (40 μg/kg/day) over a period of 42 days (21 gestation and 21 lactation). Control animals received the vehicle only. Total mercury concentration was measured in blood samples from offspring collected at the 41st postnatal day. Counting of motor neurons and immunoreactivity for myelin basic protein (MBP) were assessed in the spinal cords in both control and MeHg-intoxicated animals. Our results showed that MeHg promoted an increase in blood Hg levels. In addition, it caused a reduction in the number of spinal cord motor neurons as well as decreased MBP immunoreactivity in the cervical, thoracic and lumbar segments. Our present findings suggest that MeHg intoxication during rat pregnancy and lactation is associated with a pattern of motor neuron degeneration and downregulation of myelin basic protein in different segments of a developing spinal cord. Further studies are needed to establish the effect of MeHg intoxication in both young and adult rats. 相似文献
