There is a paucity of studies on the effect of intrauterine conditions on subsequent progeny performance in dairy cows. Using a large national data set on Irish Holstein-Friesian dairy cows, the objective of this study was to determine if intrauterine conditions, quantified by a maternal genetic variance component, significantly affected milk production, age at first calving, calving interval, somatic cell score (natural logarithm of somatic cell count) and survival in first-, second-, and third-parity female offspring. Maternal genetic variance for each trait in each parity was estimated in a linear mixed model which included, other than fixed effects, direct additive genetic, maternal genetic, cytoplasmic and permanent environmental effect of the dam, and residual component. A covariance was also estimated between the direct additive and maternal genetic components where possible. Because calves in Irish dairy herds are separated from dams at birth, a significant maternal genetic variance (with all other random effects in the model) indicates a prepartum maternal effect. A significant maternal genetic variance was estimated for 305-d milk yield in first and third lactation, somatic cell score in first lactation, and survival to second lactation from 188,144 lactations on 80,881 animals. Where estimated, a negative correlation existed between the direct additive and maternal genetic components. Regression of maternal mixed model solutions on dam milk production at different stages relative to conception revealed that greater milk yield preconception and during gestation was associated with reduced survival and milk yield and greater somatic cell count in the progeny. This study suggests that offspring survival and performance are affected by prepartum conditions that offspring experience as an oocyte, embryo, or fetus, one of which is mediated through milk production (or factors related to milk production) of the dam.  相似文献

Temperature regulation of liquid bovine semen can be difficult in field situations. Two experiments were carried out to assess the effect of storage temperature on in vitro sperm characteristics and 60-d nonreturn rate (NRR) following artificial insemination (AI) of liquid bovine semen. In experiment 1, the effect of storage of liquid bovine semen in INRA96 diluent (IMV Technologies, L'Aigle, France) at 1 of 5 storage temperatures (5, 15, or 28°C, and fluctuating between 5 and 15°C or 5 and 28°C) on total and progressive motility and kinematic parameters was assessed objectively via computer-assisted sperm analyzer on d 0, 1, 2, 3, and 4 after collection. Fluctuating temperatures were designed to mimic day- to nighttime variation. In experiment 2, we assessed the field fertility of liquid semen stored at a constant 5 or 15°C or in an unregulated manner and compared with that of frozen-thawed semen (total of n = 106,738 inseminations). In experiment 1, we detected a linear decrease in motility with increased duration of storage. Semen stored at a constant 15°C or fluctuating between 5 and 15°C had greater total motility than semen held at 5 or 28°C or fluctuating between 5 and 28°C; however, semen stored at 15°C and fluctuating between 5 and 15°C did not differ from each other. Semen held at a constant 5 or 15°C or fluctuating between 5 and 15°C, although not differing from each other, had higher progressive motility scores than that held at 28°C or fluctuating between 5 and 28°C. Semen stored at a constant 28°C exhibited poor motility and velocity values but had high progressive motion values compared with that all other storage temperatures; however, the other storage temperatures did not differ from each other in relation to motility kinematics. In experiment 2, semen stored at a constant 5°C resulted in a lower 60-d NRR (62.5%) than storage at constant 15°C or unregulated temperature or frozen-thawed semen (73.6, 74.6, and 74.4%, respectively. In conclusion, sperm stored in IRNA96 are quite tolerant in terms of storage temperature, retaining acceptable motility between 5 and 15°C. Storing semen at a constant 15°C resulted in greater in vitro sperm motility and higher NRR rates than storage at 5°C and did not differ in NRR from frozen-thawed semen or semen stored at an unregulated temperature; however, lower storage temperatures were shown to be more detrimental to sperm in vivo than unregulated storage conditions.  相似文献

Iron oxide nanoparticles (NPs) are commonly utilized for biomedical, industrial, and commercial applications due to their unique properties and potential biocompatibility. However, little is known about how exposure to iron oxide NPs may affect susceptible populations such as pregnant women and developing fetuses. To examine the influence of NP surface-charge and dose on the developmental toxicity of iron oxide NPs, Crl:CD1(ICR) (CD-1) mice were exposed to a single, low (10 mg/kg) or high (100 mg/kg) dose of positively-charged polyethyleneimine-Fe2O3-NPs (PEI-NPs), or negatively-charged poly(acrylic acid)-Fe2O3-NPs (PAA-NPs) during critical windows of organogenesis (gestation day (GD) 8, 9, or 10). A low dose of NPs, regardless of charge, did not induce toxicity. However, a high exposure led to charge-dependent fetal loss as well as morphological alterations of the uteri (both charges) and testes (positive only) of surviving offspring. Positively-charged PEI-NPs given later in organogenesis resulted in a combination of short-term fetal loss (42%) and long-term alterations in reproduction, including increased fetal loss for second generation matings (mice exposed in utero). Alternatively, negatively-charged PAA-NPs induced fetal loss (22%) earlier in organogenesis to a lesser degree than PEI-NPs with only mild alterations in offspring uterine histology observed in the long-term.  相似文献

Four adaptation options for ‘Nightingale’-type hospital ward buildings devised with practising clinicians are presented and evaluated. The adaptations recover functionality in an archaic ward configuration by delivering care to current UK National Health Service (NHS) models whilst preserving resilience to summer overheating. The investigation builds on recent work that demonstrates the significant resilience to heatwaves enjoyed by such traditionally constructed communal dormitories, the dominant UK hospital type between the late 1850s and 1939. Nightingale wards are potentially well-ventilated naturally, with good dilution of airborne pathogens. Although condemned as outdated by health ministers in recent years, many remain in use. As financial retrenchment suggests economical, creative refurbishment of hospitals will be required rather than new-build and replacement, the authors argue for health estates’ strategies that place value on resilience in a changing climate. Proposed adaptation options are investigated to assess resulting internal airflows and patient exposure to airborne pathogens. Options are costed and payback periods calculated to the standard public sector methodology. The proposed adaptations save time and cost over new-build equivalents. Selection of the most appropriate option is dependent on the characteristics of the patient cohort and care required.  相似文献

The cross and down channel flows are analyzed in the center of an idealized leakproof intermeshing twin screw extruder chamber. The respective velocity components are assumed to vary only with channel depth. Because the screw flights block the cross channel flow, fluid circulates between two complementary channel depths in the cross channel direction just as predicted for single screw extruders. In addition, fluid circulates between an independent set of channel depths in the down channel direction due to the seal provided by the second screw lands. When the two fluid motions are considered simultaneously, a fluid particle is predicted to follow a complex path over a number of channel depths during its residence time in the extruder. This unique flow also causes particles which are initially near one another to eventually move to significantly distant locations. Furthermore, a wide range of velocities and shear rates is experienced by a fluid particle as it moves to the various channel depths. The strain predicted by two approaches is nearly uniform for the twin screw extruder product in striking contrast to the distribution of absolute strains found in single screw devices. The strain uniformity, wide shear history, and fluid separation predicted by this analysis of a limiting case may help explain the good mixing capabilities of these devices.  相似文献

The p21WAF1/CIP1 gene is regulated by p53 and encodes a cyclin-dependent kinase (Cdk)-inhibitor involved in senescence and cell quiescence. The role of p21 as a negative regulator of cell proliferation suggests that it may function as a tumor suppressor gene. However, only a few mutations of the p21WAF1/CIP1 gene have been reported to date. In order to assess potential p21WAF1/CIP1 gene alterations in human bladder cancer, we have examined this gene and its encoded product in a well-characterized cohort of 27 primary bladder tumors. Mobility shifts by single-strand conformation polymorphism in the p21WAF1/CIP1 gene were identified in 2 cases. Sequencing analyses revealed that one of these cases had point mutations in the 3' untranslated region, while the other case had a frame shift mutation at positions 322 (C to A) and a deletion of 8 nucleotides (323-->331; CCG-->ACG, codon 81 Arg-->Thr) that produced a stop signal at codon 83 (Gly--Stop). This tumor had a p21-negative phenotype by immunohistochemistry, but did not lose any allele. We further characterized these cases by the study of TP53 mutations using single-strand conformation polymorphism (PCR-SSCP) and sequencing, as well as immunohistochemical assays. Seven mobility shifts were identified and seven cases showed p53 nuclear accumulation. The two cases displaying mutated p21WAF1/CIP1 had wild-type TP53. It is concluded that p21WAF1/CIP1 gene aberrations are infrequent in bladder carcinoma but may be occasionally identified in primary bladder tumors.  相似文献

In this letter, we present three new criteria for early stopping and error detection in turbo decoding. The approaches are based on monitoring the mean of the absolute values of the log-likelihood ratio of the decoded bits, which we show to be directly related to the variance of the metachannel. We demonstrate that this mean value increases as the number of errors in a frame decreases, and as a result, propose the simple mean-estimate criterion. We show that the systematic component of a terminated recursive systematic convolutional encoder used in turbo codes provides a built-in cyclic redundancy check (CRC). To further improve the performance, we also propose the mean-sign-change (MSC) criterion and the MSC-CRCeb criterion, in which a short external CRC code and the built-in CRC are concatenated with the MSC criterion.  相似文献

The Passover-related gene family plays significant roles in cellular connectivity. Mutations in three family members from Drosophila and from Caenorhabditis elegans alter a few specific electrical synapses. The passage of cobalt between Drosophila neurons was used to assay the presence of gap junctional connections. The giant fiber in the wild type has specific gap junctional connections in the brain and in the thorax. In flies mutant for Passover, cobalt cannot pass into or out of the giant fiber in either the anterograde or the retrograde directions. A large number of other gap junctional connections remain unaffected. This demonstrates that the Passover gene is necessary for gap-junctional communication between the neurons of the Drosophila giant fiber system.  相似文献
