Hydroxyproline is one of the most prevalent amino acids in animal proteins. It is not a genetically encoded amino acid, but, rather, it is produced by the post-translational modification of proline in collagen, and a few other proteins, by prolyl hydroxylase enzymes. Although this post-translational modification occurs in a limited number of proteins, its biological significance cannot be overestimated. Considering that hydroxyproline cannot be re-incorporated into pro-collagen during translation, it should be catabolized following protein degradation. A cascade of reactions leads to production of two deleterious intermediates: glyoxylate and hydrogen peroxide, which need to be immediately converted. As a result, the enzymes involved in hydroxyproline catabolism are located in specific compartments: mitochondria and peroxisomes. The particular distribution of catabolic enzymes in these compartments, in different species, depends on their dietary habits. Disturbances in hydroxyproline catabolism, due to genetic aberrations, may lead to a severe disease (primary hyperoxaluria), which often impairs kidney function. The basis of this condition is accumulation of glyoxylate and its conversion to oxalate. Since calcium oxalate is insoluble, children with this rare inherited disorder suffer from progressive kidney damage. This condition has been nearly incurable until recently, as significant advances in substrate reduction therapy using small interference RNA led to a breakthrough in primary hyperoxaluria type 1 treatment.  相似文献

In the present study, we investigated critical factors in letter-sound acquisition (i.e., letter-name knowledge and phonological awareness) with data from 653 English-speaking kindergartners in the beginning of the year. We examined (a) the contribution of phonological awareness to facilitating letter-sound acquisition from letter names and (b) the probabilities of letter-sound acquisition as a function of letter characteristics (i.e., consonant–vowel letters, vowel–consonant letters, letters with no sound cues, and vowel letters). The results show that letter-name knowledge had a large impact on letter-sound acquisition. Phonological awareness had a larger effect on letter-sound knowledge when letter names were known than when letter names were unknown. Furthermore, students were more likely to know the sounds of consonant–vowel letters (e.g., b and d) than vowel–consonant letters (e.g., l and m) and letters with no sound cues (e.g., h and y) when the letter name was known and phonological awareness was accounted for. Sounds were least likely to be known for letters with no sound cues, but reliable differences from other groups of letters depended on students' levels of phonological awareness and letter-name knowledge. Researchers have long been interested in understanding the conditions under which evaluations will be more or less consistent or context-dependent. The current research explores this issue by asking when stability or flexibility in evaluative responding would be most useful. Integrating construal level theory with research suggesting that variability in the mental representation of an attitude object can produce fluctuations in evaluative responding, we propose a functional relationship between distance and evaluative flexibility. Because individuals construe psychologically proximal objects more concretely, evaluations of proximal objects will tend to incorporate unique information from the current social context, promoting context-specific responses. Conversely, because more distal objects are construed more abstractly, evaluations of distal objects will be less context-dependent. Consistent with this reasoning, the results of 4 studies suggest that when individuals mentally construe an attitude object concretely, either because it is psychologically close or because they have been led to adopt a concrete mindset, their evaluations flexibly incorporate the views of an incidental stranger. However, when individuals think about the same issue more abstractly, their evaluations are less susceptible to incidental social influence and instead reflect their previously reported ideological values. These findings suggest that there are ways of thinking that will tend to produce more or less variability in mental representation across contexts, which in turn shapes evaluative consistency. Two experiments investigated situational information is used to identify behavior (assimilative identification) and to adjust dispositional inferences from the identified behavior (inferential adjustment). Participants heard an ambiguous or unambiguous evaluation of a job candidate by an evaluator who was under situational pressure to present either a positive or negative evaluation. In Experiment 1 participants were under low or high cognitive load. In Experiment 2 the situational information was either validated or invalidated. Results showed that cognitive load and invalidation eliminated the use of situational information for inferential adjustment. Behavior ambiguity determined the use of situational information for assimilative identification. The results suggest that the use of situational information for assimilative identification is resource independent but inflexible, whereas inferential adjustment is flexible but resource dependent. Three studies investigated the influence of mood states on the processing of positive and negative information regarding caffeine consumption and on the impact of this information on one's mood, attitudes, and intentions. The results were consistent with the predictions of the mood-as-a-resource hypothesis: First, the induction of positive mood in high (compared with low) caffeine consumers enhanced recall of negative information about caffeine consumption. Second, processing information about caffeine consumption undermined the positive mood of high (but not low) caffeine consumers. Third, the induction of positive mood enhanced the impact of negative information about caffeine on high (compared with low) caffeine consumers' attitudes and intentions toward caffeine consumption. Four studies tested the hypothesis that temporal distance increases the weight of global dispositions in predicting and explaining future behavior. Study 1 found that the correspondence bias was manifested more strongly in predictions of distant future behavior than of near future behavior. Study 2 found that participants predicted higher cross-situational consistency in distant future behavior than in near future behavior. Study 3 found that participants sought information about others' more global dispositions for predicting distant future than near future behavior. Finally, Study 4 found that participants made more global causal attributions for distant future outcomes than for near future outcomes. The results were interpreted as supporting the assumption of construal level theory that perceivers use more abstract representations (higher level construals) to predict and explain more distant future behaviors. On the basis of the assumption that positive experiences enhance perceived ability to cope with the discomfort associated with negative performance feedback (NF), it was hypothesized that (1) positive experiences increase willingness to accept negative but useful feedback and that (2) individuals seek positive experiences before accepting NF. Exp 1 found that past success increased Ss' interest in unrelated NF. Exp 2 found that positive mood increased Ss' interest in NF. Exp 3 investigated the amount of time Ss spent reading about their past success while waiting for new feedback. When the new feedback was mandatory, the time Ss spent reading about their past success increased with the anticipated negativity of the new feedback. However, when the new feedback was optional, the time Ss spent reading about their past success was an inverted-U function of the anticipated negativity of the new feedback. The goal of this study was to develop a technology for prolonged storage of pepper fruit at low temperature, based on physical treatments and packaging materials. Physical and compositional changes of red bell pepper fruit (Capsicum annuum L. cv.’Selika’) were monitored during 21 days of cold storage (at 2 °C) plus 3 days at 20 °C (market simulation). Fruits were treated with tap and hot water and stored without packaging or sealing each fruit in individual shrink packaging (Cryovac®). This research revealed that individual shrink packaging following prestorage with hot water rinse (55 °C for 15 s) over brushes, significantly reduced weight loss, softening, decay incidence and chilling injury, while maintaining a quality. The wrapped fruit ripened normally (carotenoid content increased) during shelf‐life period, when shifted to 20 °C after unwrapping. This study showed that antioxidant levels of pepper fruit may be preserved during storage.  相似文献

Analyses of data from a randomized field experiment with 1,801 participants (A. D. Vinokur et al, see record 1996-09746-001) examined the long-term effects of a job-search workshop (JOBS) and the independent effects of demographic and psychological factors on reemployment and mental health outcomes. Two years after the JOBS workshop, the experimental group had significantly higher levels of reemployment and monthly income, lower levels of depressive symptoms, lower likelihood of experiencing a major depressive episode in the last year, and better role and emotional functioning compared with the control group. Baseline job-search motivation and sense of mastery had both direct and interactive effects (with experimental condition) on reemployment and mental health outcomes, respectively.
