Cerebellar ataxias (CA) comprise a heterogeneous group of neurodegenerative diseases characterized by a lack of motor coordination. They are caused by disturbances in the cerebellum and its associated circuitries, so the major therapeutic goal is to correct cerebellar dysfunction. Neurotrophic factors enhance the survival and differentiation of selected types of neurons. Liver growth factor (LGF) is a hepatic mitogen that shows biological activity in neuroregenerative therapies. We investigate the potential therapeutic activity of LGF in the 3-acetylpiridine (3-AP) rat model of CA. This model of CA consists in the lesion of the inferior olive-induced by 3-AP (40 mg/kg). Ataxic rats were treated with 5 µg/rat LGF or vehicle during 3 weeks, analyzing: (a) motor coordination by using the rota-rod test; and (b) the immunohistochemical and biochemical evolution of several parameters related with the olivo-cerebellar function. Motor coordination improved in 3-AP-lesioned rats that received LGF treatment. LGF up-regulated NeuN and Bcl-2 protein levels in the brainstem, and increased calbindin expression and the number of neurons receiving calbindin-positive projections in the cerebellum. LGF also reduced extracellular glutamate and GABA concentrations and microglia activation in the cerebellum. In view of these results, we propose LGF as a potential therapeutic agent in cerebellar ataxias. 相似文献

Drug eluting stents are associated with late stent thrombosis (LST), delayed healing and prolonged exposure of stent struts to blood flow. Using macroscale disturbed and undisturbed fluid flow waveforms, we numerically and experimentally determined the effects of microscale model strut geometries upon the generation of prothrombotic conditions that are mediated by flow perturbations. Rectangular cross-sectional stent strut geometries of varying heights and corresponding streamlined versions were studied in the presence of disturbed and undisturbed bulk fluid flow. Numerical simulations and particle flow visualization experiments demonstrated that the interaction of bulk fluid flow and stent struts regulated the generation, size and dynamics of the peristrut flow recirculation zones. In the absence of endothelial cells, deposition of thrombin-generated fibrin occurred primarily in the recirculation zones. When endothelium was present, peristrut expression of anticoagulant thrombomodulin (TM) was dependent on strut height and geometry. Thinner and streamlined strut geometries reduced peristrut flow recirculation zones decreasing prothrombotic fibrin deposition and increasing endothelial anticoagulant TM expression. The studies define physical and functional consequences of macro- and microscale variables that relate to thrombogenicity associated with the most current stent designs, and particularly to LST. 相似文献

Automotive fuel cell technology has made considerable progress, and hydrogen fuel cell vehicles are regarded as a possible long-term solution to reduce carbon dioxide emissions, reduce fossil fuel dependency and increase energy efficiency. Even though great strides have been made, durability is still an issue. One key challenge is controlling MEA contamination. Metal ion contamination within the membrane and the effects on fuel cell performance were investigated. Given the possible benefits of using stainless steel or aluminum for balance-of-plant components or bipolar plates, cations of Al, Fe, Ni and Cr were studied. Membranes were immersed in metal sulfide solutions of varying concentration and then assembled into fuel cell MEAs tested in situ. The ranking of the four transition metals tested in terms of the greatest reduction in fuel cell performance was: Al3+ ? Fe2+ > Ni2+, Cr3+. For iron-contaminated membranes, no change in cell performance was detected until the membrane conductivity loss was greater than approximately 15%. 相似文献

The nuclear debate in Australia is shaped by the nation's substantial base of energy resources, including its large reserves of uranium and recent rises in the market price of uranium. But the debate also echoes Australia's past in the development of programs for nuclear weapons. The proposals include uranium enrichment, the storage of nuclear waste in Australia, and the introduction of nuclear reactors. A national inquiry has been instituted by the government. These developments, stemming from a range of factors, have revived the opposition to uranium mining and nuclear power which has for two decades held a dominant role in Australian politics. The issue of what should be Australia's energy path in a time of climate change is beginning to be examined. The importance of the decisions, whether to be a player in the nuclear club with its attendant multiple hazards, or to play a quite different leadership role in the development and adoption of sustainable energy strategies and technologies, cannot be exaggerated. 相似文献

Highway projects in metropolitan areas often require that adjacent utilities be relocated to accommodate new or expanded highway facilities. Traditionally, utility owners have been responsible for their own relocations prior to highway construction. However, utility relocation delay was reported as one of the root causes of overall project delays in highway construction. In the USA, one strategic approach sporadically taken over the past 15 years by some state departments of transportation (DOTs) has been to combine utility relocation work with the highway contractor's scope of work. While the combined strategy offers many benefits, it does have its own set of challenges and disadvantages. Many of these difficulties involve disagreements between DOTs and utility owners over the appropriateness of the combined strategy for particular types of relocations. Hence, a negotiation‐based decision support model (DSM) for determining whether to use the combined strategy was developed to provide guidance to both DOTs and utility decision makers during the planning and design phases of highways. A literature review and interviews with experts from both DOTs and the utility industry were conducted to identify significant factors as well as their impact levels on the decision. A DSM aiming at synthesizing the factors and orchestrating the negotiation process for the decision was proposed and verified. As suggested by the model evaluators, the proposed DSM can improve the quality of this complex decision‐making process, especially for less experienced project managers or designers. 相似文献
