Diphenyl-Methane Based Thyromimetic Inhibitors for Transthyretin Amyloidosis

Thyromimetics, whose physicochemical characteristics are analog to thyroid hormones (THs) and their derivatives, are promising candidates as novel therapeutics for neurodegenerative and metabolic pathologies. In particular, sobetirome (GC-1), one of the initial halogen-free thyromimetics, and newly synthesized IS25 and TG68, with optimized ADME-Tox profile, have recently attracted attention owing to their superior therapeutic benefits, selectivity, and enhanced permeability. Here, we further explored the functional capabilities of these thyromimetics to inhibit transthyretin (TTR) amyloidosis. TTR is a homotetrameric transporter protein for THs, yet it is also responsible for severe amyloid fibril formation, which is facilitated by tetramer dissociation into non-native monomers. By combining nuclear magnetic resonance (NMR) spectroscopy, computational simulation, and biochemical assays, we found that GC-1 and newly designed diphenyl-methane-based thyromimetics, namely IS25 and TG68, are TTR stabilizers and efficient suppressors of TTR aggregation. Based on these observations, we propose the novel potential of thyromimetics as a multi-functional therapeutic molecule for TTR-related pathologies, including neurodegenerative diseases.     

基于词性特征的明喻识别及要素抽取方法

比喻是一种利用事物之间的相似点建立关系的修辞方式。明喻是比喻中最常见的形式,具有明显的喻词,例如"像",用于关联本体和喻体。近年来高考语文散文类鉴赏题中多有考查明喻句的试题,为了解答此类鉴赏题,需要识别比喻句中的本体和喻体要素。该文提出了基于词性特征的明喻识别及要素抽取方法。首先将句子中词向量化表示与词性特征向量化表示进行融合,将融合后的向量输入到BiLSTM中进行训练,然后利用CRF解码出全局最优标注序列;最后得到明喻识别和要素抽取的结果。公开数据集上的实验结果表明,该方法优于已有的单任务方法;同时也将该文方法应用于北京高考语文鉴赏题中比喻句的识别与要素抽取,验证了方法的可行性。     

弹性元件参数优化对扑翼机构转速波动影响研究

以特拉华大学机械系统实验室所研制的样机模型进行建模,该模型传动机构采用曲柄摇杆机构.传动机构在运转的过程中,翅翼会在气动力和惯性力作用下产生周期性波动,导致电机转速波动增大,影响机构的平稳运行,产生振动和噪声.在仿真试验中引入弹簧元件,可有效降低电机转速波动,利用正交试验设计方法对影响电机转速波动的影响因素进行极差分析,得出弹簧的连接点位置是主要因素,弹簧的刚度系数是次要因素,弹簧原长影响最小.搭建了物理实验平台,验证了仿真结果和正交试验设计方法的正确性.     

基于情境感知工具辅助碎片化学习行为的研究

碎片化学习难以集中记录,存在无法迅速定位到自己需要的学习内容等用户体验问题.传统方法不能采集真实情境下的用户数据,无法满足研究人员的研究需求.基于此,开发一种基于用户行为的捕捉工具——CAUX(Context-Aware User Experience,CAUX),设计了具有情境感知能力的数据采集模块,自动捕捉指定App内的用户行为数据.CAUX对用户干扰性低,可以辅助研究人员捕捉典型的碎片化学习行为.对利用CAUX采集到的数据进行处理,并结合人工方法进行分析,可以发现不同情境下的用户行为和用户体验问题.     

Phase constitution,microstructure and mechanical properties of a Ni-based superalloy specially designed for additive manufacturing

In this study, a kind of Ni-based superalloy specially designed for additive manufacturing (AM) was investigated. Thermo-Calc simulation and differential scanning calorimetry (DSC) analysis were used to determine phases and their transformation temperature. Experimental specimens were prepared by laser metal deposition (LMD) and traditional casting method. Microstructure, phase constitution and mechanical properties of the alloy were characterized by scanning electron microscopy (SEM), transmission scanning electron microscopy (TEM), X-ray diffraction (XRD) and tensile tests. The results show that this alloy contains two basic phases, γ/γ', in addition to these phases, at least two secondary phases may be present, such as MC carbides and Laves phases. Furthermore, the as-deposited alloy has finer dendrite, its mean primary dendrite arm space (PDAS) is about 30-45 μm, and the average size of γ' particles is 100-150 nm. However, the dendrite size of the as-cast alloy is much larger and its PDAS is 300-500 μm with secondary and even third dendrite arms. Correspondingly, the alloy displays different tensile behavior with different processing methods, and the as-deposited specimen shows better ultimate tensile stress (1,085.7±51.7 MPa), yield stress (697±19.5 MPa) and elongation (25.8%±2.2%) than that of the as-cast specimen. The differences in mechanical properties of the alloy are due to the different morphology and size of dendrites, γ', and Laves phase, and the segregation of elements, etc. Such important information would be helpful for alloy application as well as new alloy development.     

催化剂级配技术在加氢改质装置上的工业应用

基于柴油产品质量升级的需求,中国石化大连石油化工研究院(FRIPP)开发了FHIDW加氢改质技术及配套FF-46加氢精制催化剂、FC-14B加氢改质催化剂和FDW-3临氢降凝催化剂.该技术及催化剂级配体系在格尔木炼油厂0.8 Mt·a-1加氢改质装置上的工业应用情况表明,级配催化剂体系对原料适应性强,装置运转平稳,操作灵活性高,催化剂失活速率慢,产品分布合理且质量优异,在降低柴油产品凝点的同时可以明显改善其密度和十六烷值,解决了改造前装置生产重柴油密度偏低的问题,为炼油厂生产符合国V质量标准的柴油提供了技术保障.
