Antioxidant (AO) activity of cold pressed pumpkin (Cucurbita pepo L.) seed oil, produced from three naked and one hulled variety, was assessed using a DC polarographic assay based on a hydrogen peroxide scavenge (HPS). Results are expressed as the decrease of the anodic oxidation current of hydrogen peroxide obtained upon addition of methanolic extract of the investigated oils. Strict correlations of HPS and (1) radical scavenging capacity against the stable free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH) (0.99), (2) the induction period estimated by a Rancimat test (0.99) and (3) total phenolic content estimated by Folin-Ciocalteu (FC) assay (0.99) were obtained. In addition, a significant correlation of HPS and the content of δ-tocopherol (0.87), squalene (0.67) and color CIE a* (−0.89) was found. Based on the results reported, the polarographic assay was found to be suitable for determination of AO activity as an indicator of the quality and oxidative stability of oil.  相似文献

Nitroxoline, a well-known antimicrobial agent, has been identified in several independent studies, and on different molecular targets, as a promising candidate to be repurposed for cancer treatment. One specific target of interest concerns cathepsin B, a lysosomal peptidase involved in the degradation of the extracellular matrix (ECM), leading to tumor invasion, metastasis and angiogenesis. However, dedicated optimization of the nitroxoline core is needed to actually deliver a nitroxoline-based antitumor drug candidate. Within that context, 34 novel nitroxoline analogs were synthesized and evaluated for their relative cathepsin B inhibitory activity, their antiproliferative properties and their antimicrobial activity. More than twenty analogs were shown to exert a similar or even slightly higher cathepsin B inhibitory activity compared to nitroxoline. The implemented modifications of the nitroxoline scaffold and the resulting SAR information can form an eligible basis for further optimization toward more potent cathepsin B inhibitors in the quest for a clinical nitroxoline-based antitumor agent.  相似文献

From the perspective of its inherent promises, difficulties, elusiveness, and relative discontinuity, the pursuit of integration in planning could be compared to the Quest for the Holy Grail. The aim of this paper is to identify the main commitments and impediments to pursued integration in the former Yugoslavia’s planning from the late 1950s until the late 1980s, drawing on the theoretical discourse, legislative, and institutional framework and the case study of the spatial plan preparation in the Socialist Republic of Serbia. During the chosen period, different aspects of integrated approach to spatial and urban planning were pursued in a context of creating and failing to deliver a unique Yugoslav version of the welfare state – the self-management socialism. In this light, the intended integration is contrasted to poor outcomes, which led to the loss of legitimacy and the final dissolution of the state, its ideology, governance, and socio-economic planning. The conclusion reflects on the main merits and pitfalls of the former Yugoslavia’s experiment to illuminate some general risks associated with the integrated approach in planning.  相似文献

Summary: Inverse gas chromatography under finite surface coverage conditions (IGC‐FC) was used for the investigation of sorption of hexane, benzene, chloroform and tetrahydrofuran on macroporous crosslinked poly[(glycidyl methacrylate)‐co‐(ethylene glycol dimethacrylate)], poly(GMA‐co‐EGDMA), and copolymer modified with ethylene diamine, EDA, poly(GMA‐co‐EGDMA)‐en, in the temperature range 333–363 K. Two poly(GMA‐co‐EGDMA) samples with different porosity parameters were synthesized by suspension copolymerization and modified using a ring‐opening reaction of the pendant epoxy groups with EDA. The specific surface areas of the initial and modified copolymer samples were determined by the BET method from low temperature nitrogen adsorption isotherms (77 K). The adsorption isotherms determined from chromatographic peaks of adsorbates were analyzed using the BET theoretical model and used for estimation of the surface area, isosteric heat of adsorption and the adsorption energy distribution on the surface of the initial and modified copolymer samples. It was observed that the adsorbate nature and the properties of the solid surface of the initial and modified copolymer governed the uptake of adsorbate by the copolymer. The deviations observed for the Sa values obtained by the BET method from the low temperature nitrogen adsorption isotherms and hexane was attributed not only to the difference in molecule size, but also to the specific polymer‐adsorbate interactions. The isosteric heat of adsorption approached a constant value with increasing adsorbate loading. A large variation in the shapes of the qstα curves resulted from strong acid‐base interactions, as well as dissolution phenomena. Of the four studied adsorbates, chloroform was shown to have the greatest affinity towards the surface of the investigated polymer samples.

The skin, being the largest organ in the human body, is exposed to the environment and suffers from both intrinsic and extrinsic aging factors. The skin aging process is characterized by several clinical features such as wrinkling, loss of elasticity, and rough-textured appearance. This complex process is accompanied with phenotypic and functional changes in cutaneous and immune cells, as well as structural and functional disturbances in extracellular matrix components such as collagens and elastin. Because skin health is considered one of the principal factors representing overall “well-being” and the perception of “health” in humans, several anti-aging strategies have recently been developed. Thus, while the fundamental mechanisms regarding skin aging are known, new substances should be considered for introduction into dermatological treatments. Herein, we describe melatonin and its metabolites as potential “aging neutralizers”. Melatonin, an evolutionarily ancient derivative of serotonin with hormonal properties, is the main neuroendocrine secretory product of the pineal gland. It regulates circadian rhythmicity and also exerts anti-oxidative, anti-inflammatory, immunomodulatory, and anti-tumor capacities. The intention of this review is to summarize changes within skin aging, research advances on the molecular mechanisms leading to these changes, and the impact of the melatoninergic anti-oxidative system controlled by melatonin and its metabolites, targeting the prevention or reversal of skin aging.  相似文献

The major role of 24-hydroxylase (CYP24A1) is to maintain 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) homeostasis. Recently, it has been discovered that CYP24A1 also catalyses the hydroxylation of 20(OH)D3, producing dihydroxy-derivatives that show very effective antitumorigenic activities. Previously we showed a negative correlation of vitamin D receptor (VDR) and CYP27B1 expression with progression, aggressiveness and overall or disease-free survivals of skin melanomas. Therefore, we analyzed CYP24A1 expression in relation to clinicopathomorphological features of nevi, skin melanomas and metastases. In melanocytic tumors, the level of CYP24A1 was higher than in the normal epidermis. The statistically highest mean CYP24A1 level was found in nevi and early stage melanomas. With melanoma progression, CYP24A1 levels decreased and in advanced stages were comparable to the normal epidermis and metastases. Furthermore, the CYP24A1 expression positively correlated with VDR and CYP27B1, and negatively correlated with mitotic activity. Lower CYP24A1 levels correlated with the presence of ulceration, necrosis, nodular type and amelanotic phenotypes. Moreover, a lack of detectable CYP24A1 expression was related to shorter overall and disease-free survival. In conclusion, the local vitamin D endocrine system affects melanoma behavior and an elevated level of CYP24A1 appears to have an important impact on the formation of melanocytic nevi and melanomagenesis, or progression, at early stages of tumor development.  相似文献
