The clearing of tropical rain forest in the Amazon basin has created large areas of cattle pasture that are now declining in productivity. Practices adopted by ranchers to restore productivity to degraded pastures have the potential to alter soil N availability and gaseous N losses from soils. We examined how soil inorganic N pools, net N mineralization and net nitrification rates, nitrification potential and NO and N2O emissions from soils of a degraded pasture responded to the following restoration treatments: (1) soil tillage followed by replanting of grass and fertilization, (2) no-till application of non-selective herbicide, planting of rice, harvest followed by no-till replanting of grass and fertilization, and (3) the same no-till sequence with soybeans instead of rice. Tillage increased soil NH4+ and NO3? pools but NH4+ and NO3? pools remained relatively constant in the control and no-till treatments. Cumulative rates of net N mineralization and net nitrification during the first 6 months after treatment varied widely but were hightest in the tilled treatment. Emissions of NO and N2O fluxes increased with tillage and with N fertilization. There were no clear relationships among rates of N fertilizer application, net N mineralization, net nitrification, NO, N2O and total N oxide emissions. Our results indicate that pasture restoration sequences involving tilling and fertilizing will increase emissions of N oxides, but the magnitude of the increase is likely to differ based on timing of fertilizer application relative to the presence of plants and the magnitude of plant N demand. Emissions of N oxides appear to be decreased by the use of restoration sequences that minimize reductions in pasture grass cover.  相似文献

Mercury is a severe environmental pollutant with neurotoxic effects, especially when exposed for long periods. Although there are several evidences regarding mercury toxicity, little is known about inorganic mercury (IHg) species and cerebellum, one of the main targets of mercury associated with the neurological symptomatology of mercurial poisoning. Besides that, the global proteomic profile assessment is a valuable tool to screen possible biomarkers and elucidate molecular targets of mercury neurotoxicity; however, the literature is still scarce. Thus, this study aimed to investigate the effects of long-term exposure to IHg in adult rats’ cerebellum and explore the modulation of the cerebellar proteome associated with biochemical and functional outcomes, providing evidence, in a translational perspective, of new mercury toxicity targets and possible biomarkers. Fifty-four adult rats were exposed to 0.375 mg/kg of HgCl2 or distilled water for 45 days using intragastric gavage. Then, the motor functions were evaluated by rotarod and inclined plane. The cerebellum was collected to quantify mercury levels, to assess the antioxidant activity against peroxyl radicals (ACAPs), the lipid peroxidation (LPO), the proteomic profile, the cell death nature by cytotoxicity and apoptosis, and the Purkinje cells density. The IHg exposure increased mercury levels in the cerebellum, reducing ACAP and increasing LPO. The proteomic approach revealed a total 419 proteins with different statuses of regulation, associated with different biological processes, such as synaptic signaling, energy metabolism and nervous system development, e.g., all these molecular changes are associated with increased cytotoxicity and apoptosis, with a neurodegenerative pattern on Purkinje cells layer and poor motor coordination and balance. In conclusion, all these findings feature a neurodegenerative process triggered by IHg in the cerebellum that culminated into motor functions deficits, which are associated with several molecular features and may be related to the clinical outcomes of people exposed to the toxicant.  相似文献

Monoacylglycerol (MAG) and diacylglycerol (DAG) are two natural components found in most edible oils and fats. Conventional synthesis of MAG and DAG is usually conducted by glycerolysis of triacylglycerol (TAG) at high temperatures (above 200°C) in the presence of an alkaline catalyst. In this work, the synthesis of MAG and DAG using enzymatic glycerolysis of olive oil was investigated using Tween 80 as surfactant, n-butanol as co-surfactant and the novel lipase in free/liquid formulation Lipozyme TL 100L as catalyst. Experimental design was used to evaluate the effect of enzyme load and reaction temperature on the feedstock conversion. Enzyme load and system temperature were significant variables in the statistical design and the best condition was found at 35°C, 7.5 vol% of Lipozyme TL 100L and glycerol to oil volumetric ratio of 2:1 with conversion of TAG at approximately 98% after 2 h of process. A mathematical model based on the Ping-Pong Bi-Bi mechanism was used to describe the reaction kinetics. The model adequately described the behavior of the system and can be a useful tool for the design of reactors in larger scales.  相似文献

New data on phase equilibria for Sacha inchi seed oil in carbon dioxide have been measured using a variable volume cell phase equilibria system at temperatures of 303, 313 and 323 K and at pressures ranging from 4.3 to 27.7 MPa. The CO2 mole fraction varied from 0.7488 to 0.9997. At the studied concentrations, phase transitions of vapor-liquid, liquid-liquid-vapor and liquid-liquid were observed. Sacha inchi oil contains 47% of omega-3 fatty acids, with a ratio of 0.76:1 for omega-6:omega-3, which is good for human health. The Peng-Robinson equation of state was used to describe the experimental data. A qualitative agreement was obtained between experimental and calculated data for the binary system CO2 and Sacha inchi seed oil.  相似文献

Highly branched alkoxysilane (HB) units were prepared in situ via a Michael‐type reaction between pentaerythrithol triacrylate and aminopropyltriethoxysilane. These units were used as an inorganic component for the modification of cellulose acetate (CA) films using the sol–gel process. The thermal and dynamic‐mechanical behaviors, the morphology, and the dimensional stability of the modified CA films were analyzed. The siloxane‐modified CA films showed thermal stability similar to pure CA, but the residue content at 900°C increased with the addition of HB units. The morphology of these films was characterized by siloxane nanodomains dispersed in the CA matrix, with good interfacial adhesion between the phases. Moreover, the CA/siloxane nanocomposite films showed improved dimensional stability in comparison with CA, i.e., in the presence of HB, the dimensional change was reduced to around 50% of the value observed for pure CA. Finally, a complex dynamic‐mechanical behavior was obtained for the nanocomposite films, as a consequence of the heterogeneous morphology. © 2011 Wiley Periodicals, Inc. The structural selectivity of the DNA-binding antitumor drug ditercalinium was investigated by competition dialysis with a series of nineteen different DNA substrates. The 7H-pyridocarbazole dimer was found to bind to double-stranded DNA with a preference for GC-rich species but can in addition form stable complexes with triplex and quadruplex structures. The preferential interaction of the drug with four-stranded DNA structures was independently confirmed by electrospray mass spectrometry and a detailed analysis of the binding reaction was performed by surface plasmon resonance (SPR) spectroscopy. The BIAcore SPR study showed that the kinetic parameters for the interaction of ditercalinium with the human telomeric quadruplex sequence are comparable to those measured with a duplex sequence. Slow association and dissociation were observed with both the quadruplex and duplex structures. The newly discovered preferential binding of ditercalinium to the antiparallel quadruplex sequence d(AG(3)[T(2)AG(3)](3)) provides new perspectives for the design of drugs that can bind to human telomeres.  相似文献

Summary The dielectric behaviour through DETA system has been studied for six PVC samples of distinct molecular microstructure in terms of content and associated surroundings and conformation of the local configurations mmr termini of isotactic sequences. Both the dielectric studies and the microstructure of the samples, as obtained by stereospecific substitution reaction of an additive-free commercial PVC are to be published in detail separately. For purposes of clarity some of the results are mentioned in Experimental section. The phenomenological correlation between the β parameter as defined by Ngai's theory and the microstructure of the samples, quantities both issued from the above quoted work, is considered in the present paper with the objective of providing some novel ideas on the physical meaning, at molecular level, of the Ngai's coupling parameter, in the light of mobility constraints and local free volume associated with mmr structure at its likely conformations. Thionation using the Lawesson’s reagent (LR) led to quantitative and selective replacement of carbonyl groups (C=O) with thionocarbonyl groups (C=S). Mixtures of thionotriglycerides were obtained by reaction of 2,4-bis (p-methoxyphenyl)-1,3-dithia-2,4-diphosphetane-2,4-disulfide in suspension with triglycerides (TG). With a molar ratio (LR)/(TG)=3∶2, there was complete exchange of C=O with C=S in the carboxylic groups of the TG. The yield is over 90%. The two P=S ligands in the LR confer a double reactivity, producing a high power of thionation. Fourier transform infrared spectroscopy was employed for quantitative and qualitative characterization of the thionated TG and for monitoring the progress of partial or total thionation as conditioned by the molar ratios of the starting compounds. Thus, with molar ration LR/TG of 0.108, 0.36, and 0.72, we obtained around 6, 20, and 40% of C=O conversion, respectively.  相似文献

Poly(ethylene-co-vinylalcohol)-grafted-poly(3-hydroxybutyrate-co-hydroxyvalerate) (EVOH-g-PHBV) was prepared by a catalyzed grafting of PHBV onto immiscible EVOH. These reactions were performed at high temperature in the molten state. The choices of Tin(II) bis(2-ethylhexanoate) (Sn(Oct)2) and 1,5,7-Triazabicyclo[4.4.0]dec-5-ene (TBD), as catalyst, were justified by a first study in an internal mixer. The catalyst quantity, reaction temperature and reaction time were optimized to apply them in the conditions of a reactive extrusion process with a co-rotating twin screw extruder. High reaction temperature, from 180 °C to 220 °C, associated to intensive mixing led to efficient grafting of PHBV onto EVOH, compatible with short residence times required.Rheological, thermal and morphological analyses were conducted to characterize the obtained copolymers. Molecular weights were determined by SEC. SEM imaging were made on cryo-fractured surfaces of blends extruded with and without catalyst. In most of reaction conditions, a compatible blend was obtained with a fine micro phase separation. Grafted copolymers were only obtained when well defined and controlled conditions are applied.  相似文献

The aim of this work was to obtain a new stable food product from cashew apple juice encapsulated by spray-drying technique using chitosan–whey protein isolate systems. The materials were evaluated according to their physicochemical stability during storage at different conditions and characterized with respect to their particle size distribution and structure. Encapsulated commercial and natural juices showed homogenous and unimodal particle size distribution with diameters ranging from 0.2 to 5.0 μm and from 0.2 to 40.0 μm, respectively. Both juices demonstrated higher physicochemical stability for vitamin C and color measurements than their respective non-encapsulated juices. X-ray diffractograms evidenced that the powder juices after the 140th day of storage were still in amorphous state. These results indicate that the new product was effective in protecting sensitive compounds present in the food matrix and it was also able to remain stable throughout the study period.  相似文献
