Well-run screening programs for cervical cancer in the population at risk have been shown to result in a sharp decrease in the incidence and mortality of cervical cancer in a number of large populations. Expression patterns of a recently identified biomarker family, microRNA, appear to be characteristic of tumor type and developmental origin. Several tumors have been reported to actively release exosomes carrying microRNAs. The present study has determined the association of microRNAs with cervical cancer-derived exosomes. The cervical cancer-derived exosomes were enriched in the cervicovaginal lavages specimens and the abundance of exosomes and exosomal microRNAs was detected by electron microscopy, western blot analysis, RT-qPCR and microRNA target reporter vector. The microRNA-21 and microRNA-146a, which were up-regulated in cervical cancer patients, were associated with the high levels of cervical cancer-derived exosomes. In conclusion, we demonstrated the abundance of exosomes in the cervicovaginal lavage specimens of women with cervical cancer. Furthermore, our results indicated that abnormally high levels of microRNA-21 and microRNA-146a existed in the cervical cancer-derived exosomes and the two microRNAs were functional in 293T cells. 相似文献

The templated electrodeposition of 200 nm diameter nanowires of the argentic oxynitrate Ag(Ag3O4)2NO3 phase is reported. Their high surface-to-volume ratio and the high average oxidation state of Ag make these wires promising candidates for nanoscale redox processes in which both a high volumetric charge density and a high discharge rate are required. A strategy based on Independent Component Analysis (ICA) and Uncorrelated linear discriminant analysis (ULDA) was proposed for proteomic profile analysis and potential biomarker discovery from proteomic mass spectra of cancer and control samples. The method mainly includes 3 steps: (1) ICA decomposition for the mass spectra; (2) selection of discriminatory independent components (ICs) using nonparametric Mann-Whitney U-test; and (3) selection of special peaks (m/z locations) as potential biomarkers by executing of ULDA on a mass spectra data set which was reconstructed with the m/z locations that collected from the selected discriminatory ICs. A colorectal cancer data set and an ovarian cancer data set were analyzed with the proposed method. As results, 9 and 10 m/z locations were selected as potential biomarkers for the colorectal and ovarian cancer data set respectively. The classification results of ULDA using the selected potential biomarkers yielded better results than fisher discriminant analysis (FDA) and principal component analysis (PCA), and could distinguish the disease samples from healthy controls on the independent test sets with 100% of sensitivities and specificities for the colorectal cancer dataset and 100% of sensitivity and 96.77% of specificity for the ovarian cancer dataset. 相似文献

目的:结合离子色谱法的优势,对国标方法蒸馏前处理进行改进,建立一种快速测定干红葡萄酒中亚硫酸盐的微提取—离子色谱准确定量的检测方法。方法:样品通过微提取装置释放出二氧化硫,被氢氧化钠溶液吸收生成亚硫酸根,再将其氧化为硫酸根,经0.45μm滤膜过滤除杂后,用离子色谱仪进行测定。结果:亚硫酸根离子质量浓度为1.0~50 mg/L亚硫酸根浓度与其峰面积呈良好的线性关系,相关系数为0.999 9,检出限为0.24 mg/L。添加不同水平硫酸盐加标回收率平均值为95.2%~99.1%,相对标准偏差(RSD)为6.84%~8.65%。结论:该方法的前处理操作简单、快速,杂质干扰明显减少,适合批量样品的检测。 相似文献

BACKGROUND: Since September 2008, an increased incidence of kidney stones and renal failure in infants, associated with the ingestion of infant formula contaminated with melamine has been reported in China. Furthermore, melamine was not only found in many protein‐based food commodities, but also in the feeds for cattle and poultry. So it is necessary to develop a suitable method to determine melamine. RESULTS: A capillary zone electrophoresis (CZE) method for analysis of melamine was developed by use of running electrolyte containing 35 mmol L?1 sodium dihydrogen phosphate at pH 3.5, with UV detection at 210 nm. Regression equation revealed linear relationships (r = 0.9999) between the peak‐area and the content of melamine from 0.8 to 80 µg mL?1. The detection limit was 0.08 µg mL?1. The method was successfully applied to the determination of melamine in milk powder, milk and fish feed, with the recoveries from 94.5% to 103.7%. CONCLUSION: The performance of the CZE method evaluated in terms of precision, limits of detection, accuracy and quantification were comparable and in good agreement with those obtained by the HPLC method, with the advantage of shorter analysis time and lower cost.
