In an effort to improve the properties of cyclodextrin glucanotransferase (CGTase) as an antistaling enzyme, error-prone PCR was used to introduce random mutations into a CGTase cloned from alkalophilic Bacillus sp. I-5 (CGTase I-5). A mutant CGTase[3-18] with the three mutations M234T, F259I and V591A was selected by agar plate assay. Sequence alignment of various CGTases indicated that M234 and F259 are located in the vicinity of the catalytic sites of the enzyme and V591 in the starch binding domain E. The cyclization activity of CGTase[3-18] was dramatically decreased by 10-fold, while the hydrolyzing activity was increased by up to 15-fold. These mutations near subsite +1 (M234T) and at subsite +2 (F259I) are likely to alter the enzyme activity in a concerted manner, promoting hydrolysis of substrate while retarding cyclization. The addition of CGTase[3-18] reduced the retrogradation rate of bread by as much as did the commercial antistaling enzyme Novamyl during 7-day storage at 4 degrees C. No cyclodextrin (CD) was detected in bread treated with CGTase[3-18], whereas 21 mg of CD per 10 g of bread was produced in bread treated with wild-type CGTase. 相似文献

Temperature programmed reduction (TPR) and oxidation (TPO) are used extensively in catalyst characterisation. In this paper, we examine the use of TPR/TPO cycles for the characterisation of a range of molybdates and single oxides. In particular we observe that the first cycle differs from that of subsequent analyses, even when the maximum temperature is limited to that used in the catalytic reaction. The effect is independent of heating rates and cooling atmospheres and has been demonstrated using different bed configurations. This observation has significance when these oxides are used in periodic flow reactors that involve many cyclical reduction/oxidation. 相似文献

This study presents an investigation of the effect of the different crystalline phases of each blend component on miscibility when blending poly(vinylidene fluoride) (PVDF) and its copolymer poly[(vinylidene fluoride)‐ran‐trifluorethylene] [P(VDF–TrFE)] containing 72 mol % of VDF. It was found that, when both components crystallized in their ferroelectric phase, the PVDF showed a strong effect on the crystallinity and phase‐transition temperature of the copolymer, indicating partial miscibility in the crystalline state. On the other hand, immiscibility was observed when both components, after melting, were crystallized in their paraelectric phase. In this case, however, a decrease in crystallization temperatures suggested a strong interaction between monomers in the liquid state. Blend morphologies indicated that, in spite of the lack of miscibility in the crystalline state, there is at least miscibility between PVDF and P(VDF–TrFE) in the liquid state, and that a very intimate mixture of the two phases on the lamellar level can be maintained upon crystallization. © 2002 Wiley Periodicals, Inc. Experimental investigations have been carried out for spherical and non-spherical particles using beds comprised of single-sized particles and mixtures in the size and particle density ranges of 439 to 1524 μm and 1303 to 4948 kg/m3, respectively. Five conical fluidizers with varying apex angles of 8.86, 14.77, 19.60, 32.0 and 43.2 degrees were used. Experimental values of minimum velocity and bed pressure drop with air as the fluidizing medium have been compared with their respective values obtained from different models available in the literature. The agronomic effectiveness of P fertilizers, as sources of phosphorus for crops, was evaluated using the quantities, Pf, of phosphorus taken up byLolium perenne grown on 14 soils during greenhouse experiments in pot cultures. The Pf quantities were determined using32P-labelled fertilizers. Data were analysed using a new concept: the Isotopic Relative Agronomic Effectiveness (IRAE). The IRAE value was defined as the ratio of the Pf quantity, taken up by a crop, of a tested fertilizer over the Pf quantity, taken up by a crop, of a fertilizer used as standard. In our experiments diammonium phosphate (DAP) was used as standard P fertilizer and two rock phosphates, the North Carolina rock phosphate (NCPR) and a calcium-iron-aluminium phosphate (Phospal), were tested. As a linear relationship between Pf(NCPR) quantities and Pf(DAP) quantities was obtained, with r2 = 0.95, when the application rates increased from 15 mgP (kg soil)–1 to 200 mgP (kg soil)–1, it is conciuded that IRAE values for a given fertilizer, other than the standard fertilizer, could be determined with a single rate of application. As regards soil pH in the range 4.7 to 8.2 the IRAENCPR is related to soil pH by a curvilinear relationship: log IRAENCPR = –(0.44) pH + 4.05 with r2 = 0.89. The average of IRAEphospal values was 0.15 with a standard error = 7% irrespective of soil pH. Then a logarithmic relationship was obtained between IRAE values of the two tested fertilizers and their water P-solubility determined at the soil pH where they were applied. 相似文献

The effect of fertilization with urea and ammonium nitrate on nitrogen mineralization was studied in a series of laboratory incubation experiments. The samples (humus layer) were collected from field experiments with different applications of fertilizers during a period of 11–14 years. The nitrogen fertilization influenced some chemical properties of the humus layer such as pH, N-Kjeldahl content and the content of inorganic nitrogen, especially in the samples from North Sweden. The sensory (odor and flavor) and physicochemical characteristics of tertiary butylhydroquinone (TBHQ) treated and butylated
hydroxyanisole/toluene (BHA/BHT) treated liquid canola shortenings, subjected to accelerated storage (Schaal oven test at
65°C) and deep fat heating (at 185°C), were determined. Data for the Schaal oven test indicate that TBHQ was effective in
retarding oxidative rancidity in liquid canola shortenings. However, addition of the commonly used mixture of BHA/BHT to canola
shortenings resulted in only a slight decrease in oxidation during schaal oven storage. The results obtained from deep fat
